Peripheral nerve damage is commonly observed in leprosy patients, but the pathogenic processes of nerve damage are still not well understood. In the present study, we produced monoclonal antibodies to protein components from human central nerve system (CNS) in an attempt to investigate the immunological mechanisms of nerve damage and to detect nerve antigen from sera in leprosy patients. Seven monoclonal antibodies (six were IgG1 and one was IgG2a in iso type) were produced; five antibodies were reactive with 38 kDa AND 43 kDa molecules from human CNS; other two antibodies were cross-reactive with human myelin basic protein and human muscle protein in ELISA. No antibody was reactive with rabbit peripheral nerve system protein. Although further evaluations of these antibodies are required to know the specific binding site of CNS protein and the reactivity to other tissue protein, this study suggests that the monoclonal antibodies may be useful in studying the immunological mechanisms of nerve damage in leprosy.-
