Abstract
p62/SQSTM1은 선택적 자가포식의 수용체 및 scaffold 단백질 역할을 하는 다기능적인 단백질이다. 선택적 자가포식에서, 이 단백질은 유비퀴틴화 된 단백질과 오토파고좀 사이의 가교역할을 한다. 뿐만 아니라, mTORC1, NF-κB, Keap1-Nrf2 경로를 포함한 다양한 세포 내 신호전달의 signaling hub로 작용한다. 이렇게 p62가 다양한 기능을 가지는 것은 도메인들이 다양한 파트너 단백질과 상호결합 할 수 있기 때문이다. 이때, p62는 인산화나 유비퀴틴화 같은 번역 후 변형에 의해 어떤 결합 파트너와 상호작용하여 어떤 세포 내 기능을 할 것인지가 결정된다고 알려져 있다. 그러나 유비퀴틴화와 반대 기전인 탈유비퀴틴화는 상대적으로 연구된 바가 적다. 본 연구에서는 USP family 중 하나인 USP13이 p62에 직접 결합하여 PB1 도메인의 유비퀴틴을 제거하는 탈유비퀴틴화 효소임을 확인하였다. 또한 USP13에 의한 p62의 탈유비퀴틴화에 의해 p62의 유비퀴틴-프로테아좀 시스템을 통한 분해가 저해됨으로써, 안정성이 증가되고 집합체 형성이 촉진되는 것을 관찰 할 수 있었다. 뿐만 아니라, p62 집합체의 증가는 오토파지 활성화 및 Keap1과의 결합 증가로 이어짐을 확인하였다. 이는 Keap1이 Nrf2의 분해를 유도하여 항산화 반응의 음성조절자로 작용하는 것을 저해하고, 결과적으로 Nrf2에 의한 항산화 반응이 활성화되는 것을 확인하였다. 결론적으로, 본 연구는 USP13에 의한 p62의 탈유비퀴틴화가 p62의 분해를 저해하고 집합체 형성을 촉진한다는 결과를 통해, 비정상적인 p62에 관련된 질병에 USP13이 영향을 줄 가능성을 시사하고 있다.
p62/SQSTM1 is a multifunctional protein that serves as a receptor protein for selective autophagy and a scaffold protein. In selective autophagy, p62 functions as a bridge between poly-ubiquitinated proteins and the autophagosome. In addition, p62 acts as a signaling hub for many cellular pathways, including mTORC1, NF-κB, and Keap1-Nrf2. It was shown that the post-translational modifications of p62, such as ubiquitination and phosphorylation, determine its binding partners and regulate their intracellular functions. However, the opposite process of the p62 ubiquitination, the deubiquitination mechanism is still unclear. It was hypothesized that the unknown pathway of p62 deubiquitination might regulate its stability and functions. In this study, it was found that ubiquitin-specific protease 13 (USP13), one of the USP family, directly bound to p62 and removed ubiquitination at its PB1 domain. Also, USP13 mediated p62 deubiquitination, which enhanced the stability of p62 and its subsequent aggregate formation. Furthermore, increased p62 aggregation activated autophagy and inhibited Keap1 from functioning as a negative regulator of the antioxidant response, which promoted the activation of Nrf2. Therefore, it is suggested that the abnormal regulation of p62 and USP13 might affect autophagy-related diseases.
