백서의 7, 12-dimethylbenz(a)anthracene 유도 악하선 종양발암과정에서의 효소 특이활성도에 대한 연구

악성종양의 조기 진단과 진행과정 및 그 발생기전에 대한 많은 연구가 활발히 진행되고

있으며, 이중 암세포에 대한 표현형 표식자(phenotypic markers)로써 사용되는 조직 항

원과 효소돌은 진단 수단으로써 그 존재가 점차 중요해지고 있다. 저자는 백서 악하선에

지용성 발암제인 7, 12-dimethyl-benz(a)anthracene(DMBA) 압착결정소구(pellet)를 매식

하여 유도한 타액선 종양 발생과정에서 효소활성도 변화를 관찰하고자 백서의 우측 악하

선을 실험군으로 하여 DMBA 압착결정소구출 매식하고 좌측을 대조군으로 하여 매 3주 간

격으로 5마리씩 희생시켜 좌, 우측 악하선을 적출한 후 생화학적방법으로 γ-glutamyl tr

anspeptidase (GGT), 5'-Nudeotidase, Omithine decarboxylase (ODC) 및 AcetyL-Co A car

boxylase에 대한 효소활성도를 측정하여 다음과 같은 결론을 얻었다.

1. 대조군인 좌측 악하선의 무게 변동은 거의 없었으나, 실험군의 경우 현저히 증가하

여 실험 15주째에는 약 7배로 증가하였다.

2. 대조군의 경우 실험 전 기간에 걸쳐 각 효소들의 활성도 변화가 관찰되지 않았다.

3. 실험군에서 GGT 활성도는 실험 9주째에 초기의 약 8배로 1.766±0.082units/mg of D

NA의 최고치를 보였다.

4. 5'-Nucleotidase 활성도는 실험 6주째에 급격히 증가하였고 9주째에 최고치인 362.1

±53.2μmoles/mg of DNA 였다.

5. ODC 활성도도 역시 실험 9주째에 최고치인 26.2±4.8nmoles/mg of DNA를 보었고, 그

후 급격히 감소하여 실험말기에는 대조군과 유사하였다.

6. Acetyl-A carboxylase 활성도는 다른 효소보다 다소 빠른 실험 6주째에 최고치인 0.

178±0.013units/mg of DNA를 보였고, 9주째부터 급격히 감소하여 15주째에는 대조군과

유사하였다.

이상의 결과로 보아 이들 효소들은 종양발생과정에서 전암세포에 대한 표식자로써 유용

할 것으로 여겨지며, 인체 종양의 조기진단과 진행에 따른 예후판정에 활용하기 위해서는

향후 혈청 및 종양조직에서의 보다 연관적인 연구가 필요할 것으로 사료된다.





A STUDY ON SPECIFIC ACTIVITIES 0F ENZYMES IN 7, 12-DIMETHYLBENZ(A)ANTHRACENE(DMBA)

-INDUCED RAT SUBMAXILLARY GLAND CARCINOGENESIS

In recent years tissue antigens and enzymes that will serve as phenotypic markers

for malignant cells are becoming increasingly important as diaglostic aids. This

study was undertaken to investigate the specific activities of these enzymes in

DMBA-induced rat submaxillary gland carcinogenesis. One hundred and twenty

Sprague-Dawley rats about 100gms of body weight were used. In experimental group,

DMBA pellet (5mg) was implanted into right submaxillary gland and sham operation

was performed into left gland to serve as control.

The animals were sacrificed every three weeks up to 15 weeks. Submaxillary glands

were excised on both sides and enzyme assays for γ-glutamyl transpeptidase (GGT),

5'-Nucleotidase, Omithine decarboxylase (ODC) and Acetrl-Co A carboxylase were

carried out biochemically.

The obtained results were as fellows :

1. In control group, there was no significant weight change of submaxillary

gland, while in experimental group, weight was increased remarkably about 7-fold at

15th week since DMBA implantation.

2. In control group there was no change in specific activities of enzymes during

the experimental period.

3. GGT activity was rapidly increased reaching a peak of 1.766±0.082units/mg of

DNA, 8-fold greater than that of onset.

4. 5'-Nucleotidase activity was increase reaching a peak of 362.1±53.2μmoles/mg

of DNA at 9th weeks.

5. ODC activity was rapidly increased, reaching a peak of 26.2±4.8nmoles/mg of

DNA at 9th week and quickly returned to that of control at 15th week.

6. Acetyl-Co A carboxylase activity was rapidly increased earlier than other

enzymes, reaching a peak of 0.178±0.013units/mg of DNA at 6th week and quickly

declined to the control level at 15th week.
