인체 다형핵백혈구에 의한 candida albicans 탐식작용시 혈청성분 분포에 관한 연구

Candida albicans의 침범시 면역글로불린 G와 보체성분 C3가 opsonin으로 작용하여 다

형핵백혈구에 의한 탐식작용에 기여하는 것으로 알려져 있다.

이에 저자는 정상혈청과 열처리 비동화혈청에서의 인체 다형핵백혈구의 Candida albica

ns에 대한 탐식능의 차이와 동시에 전자현미경적 관찰을 통하여 혈청성분중 면역글로불린

G와 보체성분 C3의 결합부위를 관찰하고, 정상혈청과 열처리 비동화혈청간의 식균화 효

과를 추구하여 다음과 같은 결론을 얻었다.

1. 다형핵백혈구의 Cindida albicans에 대한 탐식능은 정상혈청에서는 89.5 0.9%, 열처

리 비동화혈청에서는 57.2 0.9%, HBSS(Hank's Balanced Salt Solution)에서는 34.8 1.2%

의 순이었다.

2. 다형핵백혈구에 탐식된 Candids alblcans의 미세구조 소견은, 세포 섬유장구조층이

시간이 경과함에 따라 감소되었으며, Candida albicans 주위에 다형핵백혈구내의 azuroph

ilic 및 specific 과립과 전자alf도가 동일한 물질들이 침착되었다.

3. Peroxidase- labeled antihuman 면역글로불린 G로 처리한 Candida albicans의 전자

현미경적 관찰에서, 정상혈청에서는 세포벽과 섬유상구조층에 높은 밀도의 면역글로불린

G 침착이 있었으나, 열처리 비동화혈청에서는 면역글로불린 G의 침착밀도가 낮았으며 pho

sphate bufferal saline에서는 면역글로불린 G의 침착이 없었다.

4. Peroxidase-labeled antihuman C3로 처리한 Candida albicans의 전자현미경적 소견

은 정상혈청에서 세포표면 섬유상구조층에 높은 밀도의 C3 침착이 있었으며, 열처리 비동

화혈청에서는 C3 침착이 없었다.

Opsonins, IgG and complement components C3, are known to aid in the Phagocytosis

of C. albicans.

In this study, the author investigated ability of phagocytosis between normal

human serum and heat inactivated serum by use of the phagocytic index.

The author observed electron-microscopically, the binding site of IgG and C3 in

an attempt to find the opsonical differences between normal serum and heat

inactivated serum.

The results were summarized as follows:

1. Phagocytic activities of Polymorphonuclear leukocyte (PMN) were compared to C.

albicans, normal human serum was most active (89.5 0.9%), and heat inactivated

serum(57.2 0.9%)were least active. HBSS (34.8 1.2%) were found to be poor in

phagocytosis.

2. The ultra structural changes were observed electron-microscopically. The cell

wall of C. albicans which was engulfed by PMN became thicker, and electron dense

materials like azurophile and specific granules were deposited.

3. When C. albicans were treated by Peroxidase-labeled antihuman IgG, there was a

high density of IgG on the cell wall and fibrillic structure of C. albicans in

normal human serum. But lower density of IgG were observed in the heat inactivated

serum. Under HBSS, there were no IgG on the cell wall of C. albicans.

4. Although a hi71 density of C3 was deposited on the fibrillic structures of the

cell wall of C. albicans which were treated by peroxidase-labeled antihuman C3 in

normal human serum, there were deposited no C3 in the heat inactivated seam.
