STUDIES ON FINE STRUCTURE OF TRICHOMONAS TENAX AND TRICHOMONAS HOMINIS

This study was carried out to observe the fine structure of Trichomonas tenax and

Trichomonas hominis, and to discuss their structural differences.

The flagellates were cultured in modified Boeck & Drbohlav's diphasic medium and

were subcultured every other day.

The procedures for electron microscopy are briefly as follows'

The Protozoa were fixed with 3% glutaraldehyde in 0.1 M phosphate buffer (pH 7.4)

for 2 hours (4 ℃). They were washed and left overnight in 0.1 M phosphate buffer

(4℃), and post-fixed with 1% osmium tetroxido in phosphate buffer solution for 1

hour and centrifugated (500 rpm). 1% warm agar was added into the sediment and was

divided into 1 mm cubes. They were dehydrated in a grading series of ethanol from

60%, embedded in epon 812, cut into sections 500 A° thick with glass knives using

Sowall MT2 Poter Blum Ultra-microtome, and mounted on copper grids. The samples

were stained with saturated uranly acetate and leed citrate and examined with

Hitachi HU-11E-I electron microscope. Alternate section of 1 micron thick were

stained with basic fuchsin and examined with a light microscope to confirm the

flagollates.

The findings of electron microscopic observation are summerized as follows;

A. Trichomoras tenax

1. The nucleus was ovoid and surrouned by double nuclear membrane. Dense mass

assumed to be a nucleolus was observed in nucleus.

Endeplasmic reticulum was seen around the nuclear membrane by 3-4 layers and was

scattered in other part of the cytoplasm.

2. Fine black deposits characteristic of glycogen were scattered in the

cytoplasm.

Uniformly dense, round or rod-shaped dense granules were seen in cytoplasam.

Vacuoles were about 2 micron in diameter and contained ingested materials. They

were limited by the membrane same as the cytoplasmic membrane.

3. The axostyle appeared to be a cup-shaped structre comprised a single row of 41

fibrils, each about 120 A° in diameter and constant distance apart. The opened

side of axostyle which directed toward the nucleus enclosed glycogen deposits.

5 recurrent flagella and 1 anterior flagellum had 9 pairs of peripheral and 1

pair of central fibrils. The flagella were surrounded by disticnt outer membrane.

The undulating membrane, elongated extension of the cytoplasmic membrane were

observed to curve over the recurrent flagella.

4. Blepharoplast, cytostome, parabasal body, costa, Golgi apparatus and

mitochondria were not observed.

B. Trichomonas hominis

1. The nucleus was surrounded by the typical double nuclear membrane

characteristic of protozoan nuclei.

Endoplasmic reticulum was distributed only in a part around the nucleus, and was

less developed than T. tenax.

2. The structures of glycogen and dense granules wore almost same as in. t.

tenax, however dense granule was not observed in the vacuole-distributed region.

3. The micrographs showed details of cytostome engulfing a bacteria, accesoric

rootlet fiber and costa.

4. Parabasal body, blepharoplast, Golgi apparatus and mitochondria were not

observed.
