백서 두개골 결손부에서 홍화씨 추출물의 골조직 재생 유도 효과

Abstract
[한글]치주치료의 궁극적 목표는 치주질환에 의해 소실된 치주조직의 재생이다. 이를 위하여 다양한 치료법과 재료의 개발에 많은 노력을 기울여 왔다. 근래에는 생약제재의 치주조직 재생능력이 관심의 대상이 되고 있으며 이 중 홍화씨는 전통적으로 골절의 치료나 지혈작용을 위해 사용되어 왔다.

이에 본 연구에서는 Sprague - Dawley rat의 두개골에서 임계 크기 결손을 형성하여 홍화씨를 n-hexane과 chloroform으로 추출한 잔사를 MeOH로 추출한 수용성 분획을 적용하였을 때 결손부 치유양상을 알아보기 위해 국소 적용을 한 군을 실험군으로, 아무런 처치도 하지 않은 군을 대조군으로 설정하여 실험하고 술 후 각각 2주, 4주, 8주의 치유결과를 조직학적, 방사선학적으로 비교 분석하여 다음과 같은 결론을 얻었다.



1. 신생골 길이는 대조군, 실험군에서 각각 술후 2주에 102.91 ± 22.05, 178.29 ± 24.40을 보였고 4주에 130.95 ± 39.24, 242.62 ± 50.33을 보였으며 8주에 181.53 ± 76.35, 240.36 ± 22.00을 보였다(단위, μm). 2주, 4주에서 대조군과 비교해 유의성 있는 차이를 보였다(P<0.05).



2. 신생골 면적은 대조군, 실험군에서 각각 술후 2주에 2962.06 ± 1284.48, 10648.35 ± 1631.84를 보였고 4주에 5103.25 ± 1375.88, 9706.78 ± 1481.81을 보였으며 8주에 8046.02 ± 818.99, 12057.06 ± 3740.47을 보였다(단위, μm2). 2주, 4주, 8주에서 대조군과 비교해 유의성 있는 차이를 보였다(P<0.05).

3. 신생골 밀도는 대조군, 실험군에서 각각 술후 2주에 14.26 ± 6.33, 25.47 ±4.33를 보였고 4주에 20.06 ± 9.07, 26.61 ± 2.78을 보였으며 8주에 22.99 ± 3.76, 27.29 ± 1.54를 보였다(단위, %). 2주에서 유의성 있는 차이를 보였다(P<0.05).

이상의 결과에서 볼 때 백서 두개골 결손부에 홍화씨 추출물을 사용한 경우, 초기에는 골밀도, 면적, 길이를 증가시킬 수 있으며, 그 이후에는 면적과 길이를 증가시킬 수 있다고 사료된다.



[영문]The ultimate goal of periodontal therapy is the regeneration of periodontal tissue and repair of function. For more than a decade there have been many efforts to develop materials and methods of treatment to promote periodontal wound healing. Recently many efforts are concentrated on the regeneration potential of material used in oriental medicine. In some in vitro and in vivo experiments, there have been many evidences that these materials have an effect on bone regeneration.

The purpose of this study was to evaluate histologically and radiologically in Sprague-Dawley rats the effects of safflower seed extracts on the regeneration of the calvarial defects surgically produced.

So in this study, the critical size defects were surgically produced in the calvarial bone of 30 Sprague-Dawley rats using the 8mm trephine bur. The safflower seed extract was applied into the defect of each rat in experimental group, whereas nothing was applied into the defect of each rat in control group. Rats were sacrificed at 2, 4, 8 weeks following operation and histomorphometric and radiodensitometric analysis were performed.

1. The newly formed bone length was 102.91 ± 22.05, 178.29 ± 24.40 at 2 week in the each control, experimental group, 130.95 ± 39.24, 242.62 ± 50.33 at 4 week and 181.53 ± 76.35, 240.36 ± 22.00 at 8 week(unit,μm). In the 2, 4 week, there were statistically significant difference between control and experimental group (P<0.05).



2. The newly formed bone area was 2962.06 ± 1284.48, 10648.35 ± 1631.84 at 2 week, 5103.25 ± 1375.88, 9706.78 ± 1481.81 at 4 week, 8046.02± 818.99, 12057.06 ± 3740.47 at 8 week (unit, μm2). In every week, there were statistically significant difference between control and experimental group (P<0.05).



3. The radiopacity was 14.26 ± 6.33, 25.47 ± 4.33 at 2 week, 20.06 ± 9.07, 26.61 ± 2.78 at 4 week, 22.99 ± 3.76, 27.29 ± 1.54 at 8 week (unit, %). In the 2 week, there was statistically significant difference between control and experimental group (P<0.05).



In conclusion, the results of the present study suggest that safflower seed extract initially has an effect on the newly formed bone area, length and radiopacity when it is applied to the calvarial defect of Sprague - Dawley rat. Then. the material has an effect on newly formed bone area and length.
