Abstract

In literature, we are able to find many reports dealing with adrenal glands and stress, especially in physiological and biochemical aspects of adrenal activities investigated by means of administration of certain drugs, various types of stress and operations. Although the morphological changes of adrenals induced by a relatively short period of exposure to physical stressors has also been well documented, there are only a few reports desoribing the adrenal changes in the rate which are exposed to physical stressors for a relatively long period of time.

In this investigation, 50 male and female healthy rats weighing 130 to 150 gms, were selected and they were devided into 4 groups, one being the control and the rest experimental.

Each of the experimental groups was daily exposed to three different types of physical stressors; sun-light, G-force and electric shook, respectively, for 35 days.

The control group consists of 6 male and 6 female (total 12) healthy rats. They were kept and fed in well controlled animal cages to avoid undesirable stress causing from their living a conditions.

The sun-light exposure group consisting of 6 male and 5 female (total 11) rats were exposed to direct sun-light for 30-120 minutes at 2 to 3 p.m. from May 29 to July 2, 1958. Two rats died within one week during the experimental period.

The G-force group consisting of 7 male and 6 female (total 13) rats were centrifuged 250 rotations per minute for 5 minutes daily for 35 days. The length of the centrifuge arm was 21.5cm. Five rats died within 2 weeks.

The electric shock group consisting of 7 male and 7 female (total 14) rats were given electric shook by applying electrio currents on the frontal head once a day for 55 days. Four rats died within one week and one at the eleventh day.

Peripheral eosinophilo counts were done every fourth day in all groups including the control group, in order to estimate the degree of stress.

Body weights of the rats in all groups were measured daily during the entire experimental period.

All the rats were killed at the end of the experiments (i.e. 35th day) and both adrenals were carefully excised immediately. Combine weight of both adrenals of each rat were recorded and they were kept in fixative for further examination.

Following results were obtained in this observation.

1. Sun-light exposure, G-force and electric shock resulted significant changes in the adrenal glands of the rats. These changes were common in all the experimental groups.

A) Adrenal weights of the rats in all experimental groups were markedly increased as compared with those of control group. The average ratios of adrenal weight to body weight and adrenal weight in each group were as follow.

Adrenal glands of female rats were more increased in weight than those of male when compared with the control group. The average ratios of the adrenal weight to body weight and adrenal weight were shown below. (rats died during the experiments were excluded)

C) Main histological changes in adrenals of the rats in all the experimental groups were as follow.

(1) The rats died in early experimental stage(the majority of death were occured within one week) showed.

cortical thickening.

disappearance of sudanophilic material from the cortical tissue, congestion and widening of the sinusoids in the cortex and the medulla, decreased chromaffin reaction in the medullary cells.

(2) The rats finished the experiments showed, cortical thickening, increased sudanophilic material in the cortical tissue, mild congestion and widening of sinusoide in the cortex and the medulla, narrow zona glomerulosa.

2. Perpheral eosinophile cell counts showed a marked and a sudden decrease during the first 5 days (first day: 37±36.6) in the sun-light exposure group and the first 9 days (first day 39±1.5) in the G-force group.

These decrease followed by a slow increase in number but they have never recovered to the control level. The number of eosinophiles in the rats of the electria shock group showed a relatively mild decrease at begining but from the 5th experimental day they revealed a slow decrease. At the 9th day the average counts was 136??28.3 and it was followed by an irregular fluctuation, continuing to the last day of the experiment.

Rats died during the experiments in each group showed a more marked decrease in the eosinophile counts.

3. The changes in body weight are shown below.

Body weights of rats have decreased at the beginning and kept at a level lower than the initial body weights during the first 6 days in the sun-light exposure group and the first 2 days in the G-force group, and the rafter the body weight curves have gradually increased almost in parallel with that of the control group.

The electrio shock group showed little decrease in the average body weight either at the begining or during the whole experimental period, but it showed a marked fluctuation and a slow incline of body weight-curve.

In all the groups, the rats died at the early phase of the experiments showed a marked decrease in the body weight.
