소사체 치아에서의 유전자지문 분석을 위한 실험적 연구

법의학 분야의 개인식별에 있어서 소사체의 경우, 그 정도에 따라 시체의 심한 형태학적 변형이 감정을 불가능하게 하기도 하며, 또한 열에 의한 혈액과 근육 및 뼈와 같은 조직의 탄화는 유전자분석과 같은 법의분자생물학적 감정도 불가능하게 한다. 그러나 치아는 인체에서 가장 견고하고 열과 같은 외부적 환경에 쉽게 파괴되지 않는 구조와 성분으로 구성되어 있으면서도 유전자 분석에 필요한 세포들이 존재한다.

이에 본 연구에서는 이러한 치아의 특성을 이용하여, 소사체의 경우에 다른 장기나 조직에서 검출되지 않는 온도에서 개인식별에 관련된 유전자가 치아에서 검출되는지 여부를 확인하고자 실험을 시행하였다.

실험은 남, 여에서 발치 한 제3대구치 64개를 열처리 시행하지 않은 치아와 100℃,150℃,200℃에 각각 45분, 90분, 120분 및 250℃에 45분 열처리한 치아로 구분하여, 각 치아에서 DNA를 추출한 후 종합효소연쇄반응을 이용한 증폭절편다형(AMP-FLPs)을 실시하고, 현재 개인식별을 위한 유전자분석에서 중요하게 다루는 HumTH01과 HumCD4 유전좌위의 STR 대림유전자의 검출여부를 관찰하여 다음과 같은 결과를 얻었다.

1. 발거후 특기할 열처리를 시행하지 않은 치아에서는 모든 치아에서 HumTH01과 HvmCD4 유전좌위의 대립유전자를 검색할 수 있었다.

2. 100℃로 45분, 90분, 120분 열처리한 치아에서도 모든 치아에서 HumTH01과 HumCD4 유전좌위의 대립유전자를 검색할 수 있었다.

3. 150℃, 200℃로 열처리한 치아에서 HumTH01과 HumCD4 유전좌위의 대립유전자는 45분, 90분, 120분 처리한 경우에 검색이 가능했다.

4. 250℃로 열처리한 치아에서는 HumTH01과 HumCD4 유전좌위의 대립유전자를 검색할 수 없었다.

이상의 결과를 종합해 볼 때, 어느 정도와 열에 의해 발생한 소사체의 경우에 타장기에서 개인식별에 필요한 DNA가 검출되지 않더라도 치아에서는 DNA검출이 가능하므로, 치아가 법의치과학적 분자생물학 개인식별을 위한 자료로서의 가치가 매우 높은 것으로 생각되어 앞으로 실제감정에 적용할 수 있으리라 사료된다.

In the field of individual identification in forensic science, if the body is charred, it is sometimes impossible to identify the morphologic changes and charred tissue such as blood, muscle and bone can not be identified by forensic microbiologic method such as DNA typing.

So the author used the characteristics of teeth which is relatively firm compare to other organs and stable to external environment such as heat and also possess cells needed far the DNA typing. The author conducted the experiment on teeth to detect DNA related to individual identification regarding to temperature in which other charred organs can not be detected.

The experiment was done on 64 extracted third molars consisted of unheated ones, and heated teeth to 100℃, 150℃,200℃ for 45 min,90 min, and 120 min respectively and to 250c for 45 min. DNA was extracted from each tooth and amplified fragment length polymorphism procedure(AMP-FLPs) using polymerase chain reaction(PCR) was applied and observed fur the matching DNA in HumTH01 and HumCD4 locus and the followings are the results:

1. It was able to detect matching DNA in HumTH01 and HumCD4 locus in every teeth which no heating has been done.

2. It was able to detect matching DNA in HumTH01 and HumCD4 locus in every teeth healed to 100℃ for 45, 90 and 120 min.

3. It was able to detect matching DNA in HumTH01 and HumCD4 locus in teeth heated to 150℃, 200℃ for 45,90, 120 min.

4. It was impossible to detect matching DNA in HumTH01 and HumCD4 locus in teeth heated to 250℃.

So, it is possible to extract DNA from teeth that otherwise can not be extracted from other organs in the charred body and it can be concluded that teeth are highly reliable and applicatable as forensic odontology for individual identification.
