Resveratrol protects streptozotocin-induced diabetes by preventing pancreatic β-cell apoptosis through inhibition of the cleavage of poly(ADP-ribose) polymerase

dc.description.abstract[한글] 서론: 레스베라트롤 (Resveratrol;3,5,4’-trihydroxystilbene; RSV) 은 폴리 페놀계통 파이토알렉신의 한 종류이다. 대사성 질환에 대한 RSV의 효과는 다양하며, 최근에는 레스베라트롤이 혈당을 낮추는 효과가 있음이 확인되었다. 본 연구는 RSV 전 처치를 통한 췌장 보호 효과 및 이에 대한 기전을 조사하였다. 대상 및 방법: 200에서 230 그램의 수컷 Sprague-Dawley 랫트 (Sprague-Dawley rat)에 스트렙토조토신 (Streptozotocin ; STZ)을 투여하여 당뇨를 유발하였다. 실험 동물은 총 4군으로 분류하였다.; 대조군, STZ을 이용한 당뇨 군, RSV 15mg/kg 를 1주일간 하루 2회 복강 내 주사하여 전 처치한 후 STZ를 투여한 군 (RSV15) + STZ, 그리고 니코티나마이드 (nicotinamide;NA) 로 전처치 후 STZ를 투여한 군으로 분류하였다. 복강내 포도당 주사 후 72시간 째 내당능 검사를 시행하였고 기전 연구를 위해 STZ 투여 3 시간 및 6 시간 48시간 후의 췌장 조직 변화도 관찰하였다. 결과: RSV(15) 및 NA를 전처치한 군에서는 STZ 투여 후에도 당뇨가 발생하지 않았다(P = 0.005). 전처치를 시행한 군이 시행하지 않은 군에 비교하여 유의하게 STZ 투여 후 공복 혈당이 낮았다 (control vs. STZ vs. RSV(15) + STZ vs. NA + STZ; 120.02 ± 7.12 vs. 343.03 ± 12.55 vs. 157.90 ± 72.18 vs. 124.00 ± 14.92 mg/dL; P = 0.007). 각 군들간에 활성형 poly(ADP-ribose) polymerases (PARP) 의 발현 정도에 차이가 있었으며, 전 처치를 시행하지 않은 군은 3시간 후 활성형 PARP 가 발현되나 RSV 전 처치를 시행한 군에서는 활성형 PARP 가 관찰되지 않았다. 결론: 상기 결과를 종합할 때, RSV는 췌장 베타세포 보호 효과가 있을 것으로 사료되며, 당뇨병 치료제로서의 새로운 가능성이 있으리라 사료된다. [영문] Background/Aims: Resveratrol (3,5,4’-trihydroxystilbene; RSV) is one kind of polyphenolic phytoalexin. Until now, many effects on metabolic diseases of RSV have been reported. Recently anti-hyperglycemic effect of RSV was reported. This study was aimed to evaluate the effect of RSV pretreatment on pancreas protection and the detailed mechanism involving in this effect. Method: Male Sprague-Dawley rats (SD rats), weighing 200 to 230 g were divided in 4 groups; control (non-pretreatment / no STZ), STZ (non-pretreatment + STZ), RSV(15) (RSV 15 mg/kg b.i.d. for 1 week) + STZ, nicotinamide (NA) + STZ group. Diabetic condition was obtained with streptozotocin (STZ 70 mg/kg i.p.) injection. Each group included 5 ~ 10 SD rats and intraperitoneal glucose tolerance test (IPGTT) was performed at 72 houts after STZ injection. For the evaluation of molecular mechanism, the pancreas of SD rats at 3 hours, 6 hours, and 48 hours after STZ injection was obtained at each group additionally. Result: All of the rats without pretreatment had diabetes and none of the rats in RSV(15) and NA developed diabetes after STZ injection (P = 0.005). The level of fasting blood glucose was significantly low in pretreatment group, compared with non-pretreatment and STZ injection group (control vs. STZ vs. RSV(15) + STZ vs. NA + STZ; P = 0.007). The difference in expression of total length PARP was not observed among each group. But the cleavage of PARP was inhibited in RSV pretreatment group which is prominent at 3 hour after STZ injection in non-pretreatment group. Conclusion: RSV has a significant effect on protection of pancreatic β-cell. Considering the approved effect on variable metabolic disorder, it might be used as a new potential drug for diabetes.-
