[한글]

저자는 indomethacin이 Hamster협낭에서 DMBA유도 상피암 발암과정 중의 Langer-hans세포와 prostaglandins에 미치는 영향을 관찰하고자 체중 120g내외의 Hamster 60마리를, 정상군 6마리, 용매인 heavy mineral oil을 주 3회 도포한 대조군 6마리, 0.5 % DMBA를 주3회 도포하며 ml당 20ug의 indomethacin을 녹인 음료수를 공급한 실험 Ⅰ군 24마리, 0.5% DMBA만을 주3회 도포한 실험Ⅱ군 24마리로 각각 나누어, 육안적 관찰과 함께 정상 및 대조군은 실험 5주, 실험 Ⅰ, Ⅱ군은 실험 5,7,9,11,13,15,17주째에 2마리씩 회생시켰고 나머지는 18주째에 전부 회생시켜 광학현미경적 관찰은 H-E stain과 PAP stain을 하여 검경하였고 prostaglandins검출은 thin layer chromatography를 시행하여 densitometer scan과 Packard tri-carb counter로 arachidonic acid 대사산물의 상대적 구성비와 대조군에

대한 비율등을 연구하여 다음과 같은 결론을 얻었다.

1. 실험군의 육안적 소견은 10주째 까지는 실험 Ⅰ군이 Ⅱ군보다 1∼2주 늦게 반응하였으나 10주 이후에는 오히려 증상이 심하였다

2. 대조군의 조직학적 소견은 정상군과 별차 없었으며 실험군에서는 5주째 과각화증, 7주째 극세포증, 9주째 상피이형성, 13주째 편평세포암의 소견을 보였고 Ⅰ군과 Ⅱ군간의 소견은 별 차 없었다.

3. 모든 실험동물의 상피층에서는 Langerhans세포가 관찰되지 않았다. 정상군과 대조군의점막하층에서는 PAP양성 반응 세포가 관찰되었으며 실험군의 점막하층에서는 5주째부터 PAP양성 반응 세포가 증가하다가 11주째부터 감소하기 시작하였고 13주째에는 암부위에서는 대조군보다 낮은 분포를, 18주째에는 암병소에서 떨어진 부위에서도 대조군과 같은 낮은 분포를 보였으며 Ⅰ군과 Ⅱ군간의 차이는 거의 없었다.

4. Arachidonic acid대사산물은 모든 실험동물에서 공히 prostaglandin D^^2, prostaglandin E^^2, prostaglandin F^^2α의 순으로 검출되었으며 6-keto-prostaglandin F^^lα, thromboxane B^^2 는 검출되지 않았다.

5. 실험 Ⅰ군에서의 기간에 따른 arachidonic acid대사산물의 상대적 양의 변화에서 prostaglandin D^^2는 감소를, prostaglandin E^^2는 증가를 보이나 차이가 적었으며 prostaglandin F^^2α는 차이가 거의 없었다. 대조군의 상대적 양에 비해서는 소폭의 변화를

나타냈다.

6. 실험 Ⅱ군에서의 변화는 9주째부터 prostaglandin D^^2의 감소와 prostaglandin E^^2의 증가가 뚜렷하였으나 prostaglandin F^^2α는 시간에 따른 차이가 별로 없었다. 대조군의 상대적 양에 비해도 같은 변화를 보였다.

7. 실험군의 prostaglandin D^^2, prostaglardin E^^2, prostaglandin F^^2α의 양을 기간에 따라 대조군의 양과 비교하면 prostaglandin D^^2와 prostaglandin F^^2α는 미약한 변화를, prostaglandin E^^2는 뚜렷한 증가상을 보였다.

8. 실험기간에 따른 실험 Ⅰ군과 Ⅱ군간의 prostaglandin D^^2는 차이가 거의 없으며 prostaglandin E^^2는 15주이후 실험Ⅰ군에서는 증가상을 보이지 않으나 실험Ⅱ군에서는 급격하며 지속적인 증가상을 보이고 prostaglandin F^^2α 는 17주 이후 소폭의 차이를 나타냈다.

[영문]

This study was undertaken to investigate the effect of indomethacin on Langerhans cells and prostaglandins in 9,17-dimethyl-1,2-benzanthracene (DMBA) induced carcinogenesis of hamster buccal pouch.

In this study, sixty hamsters-about 120 gram of body weight-were used classifying as 1) six hamsters of normal group received no treatment, 2) six hamsters of control group treated with heavy mineral oil 3 times a week, 3) twenty four hamsters of experimental group Ⅰ treated with 0.5% DMBA in heavy mineral oil 3 times a week and administered 20 μg/ml indomethacin in drinking water ad lib, 4) twenty four hamsters of experimental group Ⅱ treated with 0.5% DMBA in heavy mineral oil 3 times a week.

The animal or normal and control groups were sacrificed 5th and 18th week, while those of experimental group Ⅰ and Ⅱ were sacrificed 5th, 7th, 9th, 11th, 13th, 15th, 17th and 18th week after experiment. The buccal pouches were excised. The tissues for hematoxylin-eosin stain and peroxidase antiperoxidase stain were fried in 10% formalin and processed in a routine manner, and the tissues for thin layer chromatography were kept frozen -70℃. Densitometer scan and Packard-tri-carb counter were utilized for the thin layer chromatography of arachidonic acid metabolites,

Following results were obtained.

1) In general, the mucosal changes of hamsters in experimental group Ⅰ appeared 1∼2 weeks earlier than those in experimental group Ⅱ during the early 10 weeks, but intra-oral changes after 10 weeks showed more distinct in experimental group Ⅰ.

2) The histologic findigs of control tissues represented similar to those of normal group. The tissues of experimental groups showed hyperkeratosis at 5th week, acanthosis at 7th week, dysplasia at 11th week, and squamous cell carcinoma at 13th

week. There were no significant differences in microscopic findings between experimental group Ⅰ and Ⅱ.

3) Langerhans cell was not observed within epidermis of all groups, however, PAP positive cells were observed within submucosal area. In experimental group, PAP positive cells of submucosal area were increased from 5th week and decreased from

11th week respectively. At 13th weeks cancerous area had fewer PAP positive cells than in central group and at 18th week, the PAP positive cells in non- cancerous area showed approximately the same number as contral group.

4) The arachidonic acid metabolites synthesized in entire tissue homogenates included prostaglandin D^^2, prostaglandin E^^2, prostaglandin F**2α in that order of relative abundances, but not included 6- keto-prostaglandin F^^lα and thromboxane B^^2.

5) The relative abundances of arachidonic acid metabolites in experimental groups were a slight decrease in prostaglandin D^^2 and a slight increase in prostaglandin E^^2, however, there was little variation in prostaglandin F^^2α during experiment.

6) The relative abundances of arachidonic acid metabolites in experimental group Ⅱ were a significant decrease in prostaglandin D^^2 and a significant increase in prostaglandin E^^2, however, there was little variation in prostaglandin F^^2α

during experiment.

7) In ratios of prostaglandins in experimental and control groups, there were a slight variation in prostaglandin D^^2 and prostaglandin F^^2α but a significant increase in prostaglandin E^^2.

8) The ratios of prostaglandin D^^2 in experimental group Ⅰ and Ⅱ compared to control group were slight differences. The ratio of prostaglandin E^^2 in experimental group Ⅰ was increased up to the 15th week, while in experimental group Ⅱ showed continuous increase. The ratio of prostaglandin F^^2α showed slight differences after 17th week.
