This study was undertaken to observe the DMBA- induced buccal pouch carcinoma in hamster and the effects of single dose of antineoplastic agents : Bleomycin and cis-Platinum.
In this study, ninety two golden Syrian hamsters, weighing about 100 grams were used.
The first experiment was carried out to produce experimentally induced - buccal pouch carcinoma by the application of 0.5% DMBA in heavy mineral oil on buccal pouch mucosa of hamsters in every other day for 12 weeks.
Experimental animals were divided into two groups, sixty six experimental group treate with 0.5% DMBA in heavy mineral oil and twenty six control group treated with saline solution as placebo.
The animals of experimental group were sacrificed serially on the 6th, 7th, 8th, 9th, 10th, 11th, 12th, 13th, 14th, 15th. weeks after application of carcinogen.
The second experiment was done by the administration of antineoplastic agents, Bleomucin and cis-Platinum in the hamster of DMBA induced buccal pouch carcinoma.
36 animals of experimental group and 24 animals of control group were diveded into two groups(Ⅰ,Ⅱ).
Group Ⅰ of experimental and control group were administrated Bleomycin
Group Ⅱ experimental and control group were administrated Bleomycin and cis-Platinum.
All animals were sacrificed serially on the 1st, 3rd, 7th, 14th, 21st, 28th day after intra peritoneal injection of antineoplastic agents.
At sacrifice, tumor masses were excised and examined grossly, microscopically and electromicroscopically.
After examination and comparison of all specimens, following results were obtained;
1. Grossly, in the first experiment, papillomas were developed at 45% in the 7th week, 75% in the 9th week, 100% in the 12th week respectively, and mortality was about 30% during the experiment.
2. Microscopically, in the first experiment, epithelial hyperplasia in the 6th week, papilloma in the 8th week, microinvasion in the 13th week and infiltrative squamous cell carcinoma in the 15th week were observed respectively.
3. In the second experiment, decreasing tendency of pleomorphism was observed from the 21st day in group Ⅰ, the 14th day in group Ⅱ.
4. Microscopically, in the second experiment, marked vascular dilatation was observed in the 21st day of group Ⅰ, the 14th day of group Ⅱ and that was prominent in group Ⅱ than that of group Ⅰ.
5. In the second experiment, vacuolar degeneration was observed in the 1st day of group Ⅰ, Ⅱ of experimental group and inflammation and necrosis were observed in Ⅰ, Ⅱ experimental groups.
Edema was observed in the 21st day of Ⅰ group, in the 14th day of Ⅱ experimental group.
6. Electronmicroscopically, vacuolation was observed in Ⅰ, Ⅱ experimental groups in the second experiment and discontinuities of basement membrane was also observed in all experimental groups.
7. In the second experiment, pseudopodia was prominent in the 3rd day of Ⅰ experimental group and mitochondrial changes were observed in all experimental and control groups and that was prominent in group Ⅰ than that of group Ⅱ.
