본 연구에서는 5명의 사람으로부터 끽연 후 남은 담배꽁초를 수거하여 1, 3, 5, 7, 15일간 실온에서 방치한 다음 DNA를 추출, 성별 검사,D1S80 유전좌위의 AMP-77Ps의 검색, HLA-DQA1 유전자의 다형성을 검색하여 법의학적 개인식별에 응용할 수 있는지 여부를 조사한 바 다음과 같은 연구 결과를 얻었다.

1. 15일간 실온에서 방치한 담배꽁초에서도 ONA 추출이 가능하므로 중합효소반응을 이용한 법의학적 개인식별에 이용할 수 있다.

2. 중합효소반응에 의한 X-Y homologous amelogenin gene을 증폭하면, 타액반 피검물(궐연)에서 성별의 판별이 가능하다.

3. 타액반 피검물(궐연)과 같은 법의학적 시료에서 DIS80 유전좌위의 증폭은 bovine serum albumin의 첨가와 hot start PCR을 시행함으로써 AMP-FLPs 검색이 가능하다.

4. HLA-DQA1 유전자의 다형성 검사는 Amplitype**TM HLA-DQ**α Forensic Kit를 사용하여 타액반 피검물(궐연)에서 간편하고 빠르게 유전자형의 결정이 가능하다.

이상의 연구를 종합해 볼때, 담배꽁초에서의 DNA의 추출, 성별 검사, D1S80 유전좌위의 AMP-FLPs 검색, HLA-DQA1 유전자의 다형성 결정은 15일이 지난 담배꽁초 시료에서도 성공적으로 수행될 수 있으므로, 담배꽁초는 법의학적 개인식별을 위한 훌륭한 분자생물학적 시료가 될 수 있다고 사료된다.

Cigarette butts from 5 smokers were gathered and then, placed in room temperature

for 1, 3, 5, 7, 15 days. The possible use of the cigarette butts for individual

indentification was evaluated in sex determination, amplification of D1S80 locus,

polymorphisms of HLA-DQA1 gene from the extracted DNA.

1. DNA extraction was possible in cigarette butts were left in room temperature

for 15 days, so it can be applicatable to individual identification by polymerase

chain reaction (PCR),

2. Amplification of X-Y homologous amelogenin gene by PCR made it possible to

identify the sex in saliva stains (cigarette butts).

3. Amplification of D1S80 locus can be acquired from adding the bovine serum

albimine and hot start PCR procedures from forensic samples such as as saliva

stains (cigarette butts), so the AMP-FLPs examining is possible.

4. Genotype could be determined simply and rapidly using Amplitpe**TM HLA-DQ**α

forensic kit in examining the HLA-DQA1 gene.

From the investigation DNA extraction sex determination, amplification of D1S80

locus, polymorphisms of HLA-DQA1 gene were successfully done even through the

cigarette butts were left for 15days at room temperature. Therefore cigarette butts

are highly reliable and applicatable as molecular biologic samples for individual

identification.
