현재까지 많은 종류의 정자 처리 방법이 알려져 왔으며 각각의 방법으로 정자를 처리한 후에 정자의 상태가 어떻게 변하는가도 많은 연구가 되어졌다. 정자처리 방법은 운동성 정자의 비율. 세균 제거 능력 등에 의해 우수성이 결정되며 특히 운동성 정자의 회수율이

좋을수록 희소정자중의 치료에 유리하다. 최근 Sephadex를 정자 처리에 간편하게 사용할 수 있도록 bead 형태로 변형시킨 장치가 개발되었고, 현재 임상 적응이 시도되고 있다.

이에 Sephadx bead column의 치료적 가치를 조사하기 위해 다음과 같은 연구를 시행하였다. 세브란스 병원에 내원한 불임 부부중 남성 요인이 없는 23명을 대상으로 Sephadex bead column을 이용한 정자 처리 방법을 일반적인 swim-up 방법과 동시에 시행하여 처리 후

정자수 및 운동성 정자의 농도를 측정하여 Sephadex bead column의 운동성 정자 선택 능력을 조사하였다. 아울러 남성 요인에 의한 불임 환자 20명의 정액을 Sephadex bead column으로 처리하여 운동성 정자의 회수율을 정상군과 비교하여 회소정자증 흑은 무력정자증

환자에게 Seohadx bead column을 이용한 방법이 얼마나 치료적 가치가 있는가를 조사하였다. 그 결과 정상군에서 Sephadex bead column 방법을 이용하였을 때 회수된 총 정자수 및 운동성 정자회수율이 swim-up 방법에 비해 높았으며, Sephadex bead column 방법을 이용하였을 때 운동성 정자의 회수율은 정상군과 회소정자증 혹은 무력정자증군 간에 차이가 없었다. 이상의 결과로 보아 Sephadex bead column을 이용한 정자 처리방법은 시술이 비교적 간단하고, 보편적으로 사용되어 왔던 swim-up 방법에 비해 높은 운동성 정자

회수율을 나타내었다. 따라서 Sephadex bead column 방법은 남성 요인에 의한 불임 환자의 치료를 위한 정자 처리 방법으로 많이 활용될 수 있으리라 생각한다.

To date, many kinds of sperm preparation method and their clinical efficiency has

been studied in the field of reproductive technology. The recovery rate of motile

sperm is very important factor in treatment of male factor column was developed for

sperm preparation and applied to clinical use. The semen samples of 23 normal male

were prepared by Sephadex bead column and swim-up method at same time. Then sperm

count and recovery rate of motile sperm were compared after preparation by two

methods. Twenty semen samples of oilgospermia or asthenospermia patients were

prepared using Sephadex bead column. The recovery rates of normal control group and

oligospermia or asthenospermaia group after Sephadex bead column preparation were

compared. The sperm count and recovery rate of motile sperm after Sephadex bead

column were significantly higher than those of swim-up method. The recovery rates

of motile sperm after prepartion with Sephadex bead column were significantly

higher than those of swim-up method. The sperm count and recovery rate of motile

sperm after Sephadex bead column were signiicantly higher than those of swin-up

method. The recovery rates of motile sperm after preparation with Sephadex bead

column was not significantly different in control and oligospermia or

asthenospermia group. From these date, we can postulate that Sephadex bead coumn is

successful in recovering motile sperm and can be used in clinical situatuon in male

factor infertility.
