dc.description.abstractObjectives: Cell therapy has been extensively studied as a gene complementation approach in such genetic diseases as Duchenne muscular dystrophy (DMD). Adipose tissue has recently been identified as an alternative, uniquely abundant and accessible source of pluripotent cells. In the present work we investigated myogenic potentials of adipose-tissue-derived cells (ATDCs) as mesenchymal stem cells (MSCs). Methods: Human ATDCs were obtained by liposuction and cultured in three different media; control, myogenic and conditioned media. The following observation was made to evaluate differentiation with using immunoflurescence study and western blotting. Results: Conversion of ATDCs to a myogenic phenotype is observed by indirect immunoflurescence study of MyoD and Myf-5 in regardless of media type. However, secondary myogenic regulatory factors (Myf-6 and myogenin) and desmin are negative in all different culture condition. Conclusion: Our findings suggest that human adipose-tissue-derived cells might have a mesencymal stem cell population and myogenic potentials. Since human adipose tissue is plentiful, easily harvested in large quantity under local anesthesia with little patient discomfort, it may be a good source of an alternative cell therapy for DMD patients.-
