The Intrinsic Role of Viperin in Metabolic Diseases

Abstract
Viperin is a multifunctional interferon (IFN)-inducible protein. It was identified from the IFN-gamma-treated human macrophages. Viperin normally localizes to the endoplasmic reticulum (ER) and has antiviral activity. However, this protein re-localizes from ER to mitochondria during human cytomegalovirus infection. There, viperin interacts with trifunctional protein, an essential enzyme for fatty acid β-oxidation, and modulates cellular lipid metabolism. Several studies have reported the involvement of fatty acid β-oxidation in various metabolic phenotypes, such as thermogenesis and macrophage polarization. I observed that viperin is naturally expressed in various metabolic organs including two main fat depots, white adipose tissue and brown adipose tissue. Here I hypothesize that viperin may play a role in metabolic diseases such as diabetes, obesity and atherosclerosis. Initially, I investigated viperin effect on metabolic phenotypes of wild type (WT) and viperin knockout (KO) mice fed regular chow (RC) or high-fat-diet (HFD). I found that viperin affects metabolism in adipose tissues of mice. Viperin KO mice exhibit reduction of fat mass and adipocyte size and improvement of HFD-induced glucose tolerance. The KO mice also show higher rate of oxygen consumption and energy expenditure. I observed that the expression levels of fatty acid β-oxidation- and thermogenesis-related genes are increased in adipose tissues of viperin KO mice when compared with those of WT mice. These thermogenic phenotypes are attributed to an adipocyte autonomous mechanism that regulates fatty acid -oxidation. These data indicate that viperin negatively regulates thermogenesis in adipocyte of adipose tissues, resulting from inhibition of fatty acid -oxidation. Next, I also investigated the role of viperin in the development of atherosclerosis. I generated viperin-/-/apolipoprotein E (ApoE)-/-double knockout (DKO) mice. Using these mice, I found that atherosclerotic lesions of DKO mice are more severe than those of ApoE KO mice fed RC or western-diet (WD). I showed that viperin is expressed in macrophage in atherosclerotic lesions of ApoE KO mice fed WD. In addition, viperin expressed in M1, M2 -like macrophages of mice carotid atherosclerotic plaques, which are important in atherosclerosis. These data suggested that viperin might be involved in the regulation of the formation of macrophages in atherosclerotic lesions. To investigate whether viperin affects polarization of macrophages, bone marrow was isolated from WT or viperin KO mice. I show that viperin is basally expressed in murine bone marrow cells including monocytes. Its expression is maintained in bone marrow monocyte-derived macrophages (BMDMs). In BMDMs, viperin deficiency promotes polarization of macrophages and secretion of M1 and M2 cytokines. Collectively, these findings indicate that viperin plays a role as a metabolic regulator in metabolic diseases.

Viperin은 인터페론에 의해 유도되는 다기능의 단백질이다. Viperin은 사람 대식세포에서 인터페론 감마를 처리 후 동정되었다. Viperin은 일반적으로 소포체에 위치하며, 항 바이러스 작용을 한다. 하지만 Human cytomegalovirus 감염시 이 단백질은 소포체에서부터 이동하여 미토콘드리아로 위치하게 된다. 이동된 Viperin은 지방산 베타 산화에 필수적인 효소인 trifunctional protein과의 상호작용을 통해 세포의 지방대사를 조절한다. 최근 연구를 통해 viperin이 흰색지방과 갈색지방을 포함한 대사와 관련된 조직에서 자연적으로 발현하고 있는 것을 관찰하였다. 이러한 현상을 바탕으로 viperin이 당뇨, 비만 그리고 동맥경화와 같은 대사질환에서 역할을 할 수도 있다는 가설을 설정하였다. 해당가설을 증명하기 위하여 먼저, 야생형과 viperin 유전자제거 쥐에서 일반식이와 고지방식이 후 viperin이 대사표현형에 미치는 영향을 조사하였다. 그 결과 viperin이 지방에서 대사에 영향을 미친다는 것을 발견하였다. Viperin 유전자 제거 쥐에서 지방무게의 감소, 지방세포의 크기 감소와 고지방식이로 인해 증가된 포도당 내성이 향상된 것을 관찰하였다. 유전자 제거쥐는 또한 높은 산소소비량과 에너지 소비량을 보였다. 지방산 베타 산화와 열발생과 관련된 유전자의 발현이 viperin 유전자 제거 쥐의 지방조직에서 야생형에 비해 높은 것을 확인해였다. 이를통해, 이러한 열발산과 관련된 표현형은 지방산 베타 산화가 조절됨으로써의 지방세포의 자발적인 대사의 변화로 인함 이라는 것을 알수 있었다. 이러한 결과들은 viperin이 지방산 베타 산화를 조절함으로써 지방조직에서 열발산의 저해할수 있다는 것을 밝혀냈다. 다음으로, 동맥경화의 발달과정에서 viperin이 미치는 영향을 알아보고자 하였다. 이를 위해여viperin과 아포지방단백 E(Apolipoprotein E)의 이중 유전자제거(double kncokout) 쥐를 제작하였다. 해당 쥐를 사용하여 일반식이 혹은, 서구화된 식이를 했을 때 아포지방단백 E의 단일 유전자 제거마우스에 비해 이중유전자 제거마우스에서 더 심한 동맹경화 경변을 관찰하였다. 서구화된 식이를 한 아포지방단백 E 유전자 제거쥐의 동맥경화 경변상에 존재하는 대식세포에서 vipeirn의 발현을 확인하였다. 또한 동맥경화에서 중요한 M1, M2 대식세포에서 viperin의 발현을 확인하였다. 이 결과들은 동맥경화상에서 대식세포의 형태형성에 viperin이 관여할수 있다는 것을 암시한다. Viperin이 대식세포의 M1, M2 분화에 미치는 영향을 알아보기 위하여, 골수세포를 야생형과 viperin 유전자를 제거한 쥐에서 분리하였다. Viperin은 기본적으로 쥐의 골수세포에서 발현 되고 있었다. 이러한 발현은 골수유래대식세포(BMDMs)에서도 유지되고 있었다. BMDMs 에서 vieprin이 없는 경우 대식세포의 분화가 더 잘일어나며 M1, M2와 관련된 사이토카인들의 발현이 증가되는 것을 확인하였다. 종합적으로 해당 연구결과들은 viperin이 대사질환들에서 대사를 조절하는 역할을 수행하는 새로운 가능성을 시사한다.
