N-hexane 및 benzene이 백서 경골 신경에 미치는 영향에 관한 조직계측학적 연구

[영문] [한글] 벤젠과 노말헥산이 말초신경에 미치는 영향을 관찰하기 위하여 아급성중독 상태를 유발 할 수 있는 기중농도인 6,000ppm의 노말헥산과 2,000ppm의 벤젠을 흰쥐에 2주, 4주, 6주 동안 흡입시킨 후 경골신경의 섬유직경, 수초두께, 축삭의 직경을 계측하였다. 경골신경을 절제하여 고정 및 탈수한 후 epon 812에 포매하여 1μm 두께로 잘라 광학현 미경으로 관찰하고 1,000배로 한시야씩 건너뛰며 사진을 찍은 다음 최종배율이 2,000배가 되도록 인화하였다. 신경섬유 직경의 계측은 원형은 1회, 타원형은 장경과 단경을 각각 계측하여 평균값을 이용하였고 수초의 두께는 얇은 쪽을 택해 1회 계측하였다. 모든 계측치는 SPSS Package program을 이용하여 컴퓨터로 처리분석하여 다음과 같은 결과를 얻었다. 1. 수초의 두께는 벤젠 흡입군에서 대조군과 차이가 없었으나 노말헥산 4주 및 6주 흡 입군에서 대조군에 비해 감소하는 경향을 보였다. 2. 신경섬유의 직경과 수초두께 및 축삭직경은 대조군과 실험군에서 상호 비례적인 상 관관계를 보였다. 3. 동일한 직경의 신경섭유를 비교할 때 축삭직경과 수초의 분포는 벤젠을 흡입한 군에 서는큰 차이를 보이지 않았으나 노말헥산 4주 및 6주 흡입군에서는 수초두께가 작은 섬유 의 분포가 많았다. 이상의 결과로 미루어 보아 노말헥산 흡입군의 신경섬유 수초가 같은 직경의 대조군 및 벤젠흡입군 보다 상대적으로 얇아진 것은 노말헥산이 신경섬유의 성장을 저하시키기 때 문인 것으로 사료된다. A Morphometrical Study of Tibial Nerve Fibers of Rats Influenced by Benzene and n-H exane Young Soo Lee Department of Medical Science The Graduate School, Yonsei University (Directed by Professor Young Hahn Moon, M.D.) In order to observe the effects of benzene and n-hexane on the peripheral nerve, tibial nerves of rat were observed after exposing them to two air concentrations of these organic solvents(6,000 ppm of n-hexane and 2,000 ppm of benzene), which were known to be the levels to cause subacute toxicity, for the three different periods: two, four and six weeks. Tibial nervy was removed, fixed, dehydrated, embeded in epon 812, and then sectioned 1 micron thick. The sections were observed under the 200× magnification, and photographed while scanning the different fields. The prints were finally enlarged to 2,000× magnigication. The average diameter for the circularly sectioned fibers, major and minor for the ovoidally sectioned fibers were measured. The thickness of the thinner part of the myelin sheath was also measured. The SPSS trackage computer program was used for the statistical analysis of these data. The results were as follows. 1. Myelin sheath thickness of tibial nerve fiber for tIne two different n-hexane exposed groups (four and six weeks) were reduced compared with the control group. 2. There were positive relationships among nerve fiber diameter, myelin sheath thickness and axon diameter for bosh exposed and control groups. 3. There was no significant difference in mrelin sheath thickness for the equal diameter nerve fiber between the exposed group and the control group, but the greater number of thin myelin sheath were observed for the n-hexane exposed group compared with the control group. Thus, it is concluded that n-hexane tends to reduce the growth rate of nerve fiber more so than benzene. While these results shed light on understanding the effects of benzene and n-hexane, the duration of exposure was not long enough to apply these results to real world situation. To further understand the mechanism of nerve fiber degeneration caused by the organic solvents, both epidemiological and biochemical studies should accompanied this kind of study.
