토끼 방광근 운동에 대한 Vasoactive Intestinal Polypeptide의 영향

골반신경자극에 의한 방광배뇨근의 흥분성 수축반응은 atropine에 의해 완전히 봉쇄되

지 않는데 이는 비콜린성 비아드레날린성 흥분성 신경이 존재하기 때문이며 이의 신경전

달물질은 ATP나 펩타이드성 물질이라고 알려져 있다. 또한 방광에는 억제성인 비콜린성

비아드레날린성 신경이 발견 되었으며 이의 신경 전달물질로는 5-hydroxytryptamine (5-H

T)과 vasoactive intestinal polypeptide (VIP)가 주장되고 있다.

VIP는 돼지의 소장에서 처음 분리되어 위장호르몬으로 인정되었으나 그후 중추신경계

뿐 아니라 말초신경계에도 널리 분포되어 있음이 밝혀져 있다.

평활근에 대한 VIP의 작용기전은 아직까지 확실히 밝혀지지 않고 있으나 칼슘의 이동을

촉진함으로써 활동전압의 발생을 억제하고 평활근막에 대한 칼륨의 투과도를 증가시키는

것과 관계가 있는 것으로 추측되며 β 아드레날린성 신경이나 prostaglandin과는 무관하

다고 보고있다.

이번 실험에서는 토끼 방광의 배뇨근과 삼각근을 이용하여 콜린성 및 아드레날린성 효

과에 대한 VIP의 영향을 살펴보고 그 기전을 칼슘 및 칼륨 이동과 연관지어 검색하였다.

실험동물로는 체중 2kg안팎의 잡종토끼를 사용하여 희생시킨 다음 방광의 배뇨근과 삼

각근 절편을 만들어 37℃의 Tyrode용액 1ml가 들어있는 이중벽 organ bath에 현수하였으

며 95%O^^2 -5%CO^^2 혼합 가스를 공급하였다. 방광근절편이 안정된 긴장도를 나타낼 때

약물을 투여하여 절편의 수축곡선변동을 Polygraph (Grass, Model 7, USA)에 묘사하였으

며 다음과 같은 결과를 얻었다.

1. Vasoactive intestinal polypeptide (VIP)는 방광의 배뇨근과 삼각근의 자발운동 및

긴장도를 억제시켰다.

2. 방광의 배뇨근절편에서 VIP는 콜린성 약물에 의한 흥분반응을 억제하였다.

3. 방광의 삼각근절편에서 VIP는 아드레날린성 약물에 의한 흥분반응을 억제하였다.

4. 칼슘배제 고칼륨 탈분극 영양액 내에서 칼슘에 의해 유발되는 방광의 배뇨근과 삼각

근절편의 긴장도는 VIP에 의해 의의있게 억제되었다.

5. VIP는 칼륨에 의한 방광의 배뇨근과 삼각근절편의 긴장도 증가를 현저하게 억제하였

다.

이상의 결과로_보아 VIP는 토끼 방광의 배뇨근과 삼각근절편의 자발운동을 비롯하여 콜

린성 약물 및 아드레날린성 약물에 의한 수축작용을 억제하는 바 이러한 VIP의 방광수축

억제작용에는 칼슘의 세포내 유입억제와 칼륨통로 개방이 관여할 것으로 추측된다.





Effects of Vasoactive Intestinal Polypeptide on Motility of Rabbit Urinary Bladder

It has been known that the excitatory response of the detrusor to pelvic nerve

stimulation is largely atropine resistant and mediated by non-cholinergic,

non-adrenergic, excitatory nerves.

Recently, 5-hydroxytryptamine (5-HT) and vasoactive intestinal polypeptide (VIP)

have been suggested as a possible non-cholinergic, non-adrenergic inhibitory

transmitter in the urinary bladder of the pig and of man.

VIP, a 28-amino acid peptide isolated from porcine duodenum, has been detected by

immunocytochemistry and radioimmunoassay in many parts of the peripheral as well as

central nervous systems.

The effect of VIP on urinary tract smooth muscle has been well documented, but

the results are still conflicting. It may produce relaxation or contraction, or be

inactive.

In this study, the effect of VIP was investigated, using the isolated rabbit

urinary bladder strip with special reference to mobilization of Ca**2+ and K**+

ions.

Bladder strips of 5-10 mm long were carefully prepared from dome and trigone of

the rabbit and were suspended in an organ bath containing 1 ml of Tyrode solution

maintained at 37℃. Tyrode solution was aerated with 95% oxygen and 5% carbon

dioxide. One end of the bladder strip was attached at the bottom of the bath and

the other end to the force displacement transducer (FT.03, Grass, USA).

When a stable tension level of the strip had been attained, drugs were added to

the organ bath and the change of motility of the strip was recorded on a Polygraph

(Grass Model 7, USA).

The results obtained were as fellows;

1. VIP inhibited the spontaneous contraction of detrusor and trigone strips of

rabbit bladder.

2. VIP exerted relaxation of detrusor strip contraction induced by cholinergic

drugs.

3. The contraction of trigone strip induced by adrenergic drugs was relaxed by

VIP.

4. In calcium-free high-potassium depolarizing solution, VIP exerted a relaxing

effect on detrusor and trigone strip contraction induced by CaCl^^2.

5. VIP exerted a relaxing effect on detrusor and trigone strip contraction

induced by KCI.

From the above results, it was concluded that VIP inhibits the spontaneous

motility as well as cholinergic- or adrenergic- induced contraction, and that these

effects were associated with the inhibition of Ca**+2 influx and K**+ -channel

opening.
