악하선에서의 Na+, K+-activated adenosine triphosphatase의 특성에 관한 실험적 연구

dc.description.abstract[영문] [한글] 고양이의 악하선에서 microsome 분획을 얻어 deoxycholate를 처리한 후 Na-K-ATPase활 성을 측정하고 악하선에 있는 이 효소의 특성을 구명하기 위하여 이 효소 활성에 대한 in cubation시간, incubation용액의 조성, 수종약물의 영향, PH 및 온도의 영향등을 관찰하 여 다음과 같은 결론을 얻었다. 1. Microsome 분획을 얻는 과정에서 deoxycholote를 2번 처리한 결과 Na -K -ATPase활성이 0.58 0.04μmol pi/㎎ Protein/min, Mg-ATPase 활성은 0.42 0.03 μmol pi /㎎ Protein/min으로 나타났다. 2. 효소의 활성도는 incubation시간이나 효소 농도에 따라 증가하였다. 3. Incubation 용액내 ATP와 Mg 농도비와 Na과 K의 농도비가 일정할 때 최대활성도를 나타내었는데 2mM ATP, 3mM Mg, 100mM Na 그리고 10mM K일때 최대활성도를 나타내었다. 4. Na-K-ATPase의 최대활성도는 PH 7.5에서 나타났으나 Mg-ATPase의 활성도는 시험된 범위내에서 PH가 증가할수록 증가하였다. 5. Na-K-ATPase 활성도는 ouabain, ethacrynic acid에 의해서 억제되었으나 NaN^^3, ac etylcholine, 항암제인 mitomycin-C에 의해서는 영향을 받지 않았다. 6. Arrhenius plot 결과 25℃에서 회절되었으며 25℃보다 낮은 온도 범위에서는 높은 온도 범위에서보다 activation energy가 증가되었다. 이상의 결과로 보아 고양이의 악하선에는 다량의 Na-K-ATPase를 함유하고 있으며, 그 특성은 다른 조직의 Na-K-ATPase와 유사한 효소 특성을 가진 것으로 사료된다. An Experimental Study on Characteristics of Na**+, K**+ -activated Adenosine Triphosphatase in Submaxillary Gland In Kyo Jeong.D.D.S. Department of Dental Science, the Graduate School, Yonsei University (Directed by Prof. Eui Wung Lee.D.D.S.,M.S.D.,Ph.D.) The Na-K-ATPase was isolated from cat submaxillary gland by a deoxyclolate-treated procedure. In order to determine the characteristics of Na-K-ATPase of submaxillary gland, effects of incubation time, medium composition, temperature and various drugs were investigated. The results were summarized as fellows: 1. The specific activities of Na-K-ATPase and Mg-ATPase were 0.58 0.04 and 0.42 0.03μmol Pi/㎎ protein/min, respectively, in twice deoxycholate-treated microsome. 2. The initial velocity of the Na-K-ATPase reaction was directly proportional to the incubation time and to the enzyme concentration in the medium. 3. The rate of the reaction was dependent on the concentrations of ATP, Mg, Na, K and H ions in the medium, showing maximal activity at 2mM ATP, 3mM Mg, 100mM Na and 10mM K. 4. The maximal activity of Na-K-ATPase was observed in pH 7.5 but Mg-ATPase activity increased by increasing of medium pH. 5. The Na-K-ATPase activity was inhibited by ouabain and ethacrynic acid, but not affected by NaN^^3, acetylcholine and mitomycin-C. 6. The Arrhenius plot showed biphasic pattern and the higher activation energy at lower range(10-25℃) than higher temperature range(25-37℃). These results indicate that the Na-K-ATPase of submaxillary gland of cat is similar to that of the other tissue in its physiological and biochemical properties .-
