심근세포는 α1-아드레날린 수용체(α1-adrenergic receptor, α1-AR)의 자극에 의해 비대 반응을 나타낸다. Gh는 새롭게 밝혀진 G 단백질 의 일종으로 조직형 글루타민전이효소(transglutaminase)와 α1-AR의 신호전달 기능을 동시에 가진다. 아직 Gh를 통해 비대반응을 일으키는 세포내 신호 전달 경로는 규명되어 있지않다. RGS (Regulator of G protein signaling)는 Gαi와 Gαq를 통한 신호 전달을 완전히 차단하는 단백질로서, 백서의 심근세포에서는 RGS4의 형태로 존재한다. 본 연구에서는 RGS4를 과발현시킨 신생백서의 심근세포를 이용하여 norepinephrine 자극에 의해 비대반응을 일으키는 세포내 신호전달 경로에서 Gh가 어떻게 관여하는지 알아보고자 하였다.

비대반응의 지표로는 총단백질 합성량을 이용하였다. α1-AR/Gh를 통한 심근세포의 비대반응의 기전을 알아보기 위하여 신생 백서 심근세포에 RGS4와 Gh를 과발현시킨 다음, norepinephrine으로 자극하여 mitogen-activated protein kinases(MAPKs)와 그의 상위 조절인자인 protein kinase C(PKC), Ras, mitogen activated ERK activating kinase (MEK) 1,2의 활성과 원종양 유전자의 발현을 관찰하였다.

Norepinephrine 자극에 의한 extracellular signal-regulated kinases(ERKs)의 활성화는 α1-AR blocker에 의해 차단되었으나, β-AR blocker는 영향을 주지 않았다. Gh의 과발현은 norepinephrine에 의한 ERKs의 활성화를 촉진시켰으며, α1-AR blocker에 의해 이러한 효과는 차단되었다. Gh와 RSG4를 모두 과발현시킬 경우, norepinephrine에 의한 비대반응은 PKC/MEK1,2/ERKs의 활성화를 통한 c-myc의 발현의 증가로 이루어졌다.

이상의 연구 결과를 토대로 norepinephrine은 신생 백서의 심근세포에서 α1-AR를 통해 비대반응을 일으키고, Gh가 norepinephrine에 의한 PKC/MEK1,2/ERKs 의 활성화에 부분적으로 관여함을 알 수 있었다. 그리고 Gh에 의한 PKC/MEK1,2/ERKs의 활성화는 원종양유전자인 c-myc의 발현을 증가시켜 단백질의 합성을 증가시켰다.





[영문]Background-In cardiac myocytes, stimulation of α1-adrenoceptor (α1-AR) leads to hypertrophic phenomenon. The Gh protein (Transglutaminase II, TGII) is tissue type transglutaminase and transmits the α1B-AR signal with GTPase activity. Little is known about Gh-mediated intracellular signal transductions to develop the hypertrophy. The aim of this study was to elucidate how Gh mediates norepinephrine-stimulated intracellular signal transductions leading to the hypertrophic responses in RGS4-transfected neonatal rat cardiomyocytes.

Methods and Results-To elucidate the molecular mechanisms of an α1- adrenoceptor/Gh-mediated hypertrophic responses, we examined the effects of norephinephrine on the activation of MAP kinases and their up-stream regulators, protein kinase C, Ras, and MEK1,2 in RGS4-transfected neonatal rat cardiomyocytes. The protooncogenes expression and the protein synthesis were also examined to address the hypertrophic responses. Norepinephrine-induced ERKs activation was inhibited by an α1-AR blocker (prazosin), but not by a β-AR blocker (propranolol). Overexpression of the Gh protein stimulated norepinephrine-induced ERKs activation, which was inhibited by α1-AR blocker (prazosin). Co-overexpression of Gh and RSG4, the regulatory protein of Gq, leaded to the partial decrease of norepinephrine- induced hypertrophic responses and activated PKC/MEK1,2/ERKs, leading up-regulation of c-myc.

Conclusion-Norepinephrine induces hypertrophy in neonatal rat cardiomyocytes through α1-AR stimulation. Gh is partly involved in norepinephrine-induced PKC/MEK1,2/ERKs activation. Activation of Gh-mediated PKC
