수지상세포(dendritic cell)는 형태학적으로 수상돌기를 갖고 있으며 가장 강력한 항원전달능력이 있어 일차 면역반응 및 T 세포매개 면역반응에 가장 중요한 기능을 수행한다. 최근 이러한 수지상세포를 사용하여 효과적으로 종양특이 세포독성 T 세포를 유도함으로써 생체의 항암 면역작용을 증가시키는 방법이 연구되고 있다.

세포고사(apoptosis)는 손상 받았거나 원하지 않는 세포를 생체 내에서 제거하여 생물의 항상성을 유지하는데 중요한 역할을 한다. 이때 수지상세포가 강력한 항원전달세포로 작용하여 세포고사된 항원을 T 세포에 전달하여 면역반응을 일으키는 것으로 알려져 있다. 세포고사된 종양세포가 수지상세포를 이용한 면역치료에 좋은 항원으로 사용될 수 있는 이유는 세포고사된 종양세포가 세포괴사(necrosis)된 종양세포에 비해 교차장전을 잘 일으켜서 CD8+ T 세포를 더 잘 유도하여 결과적으로 종양억제 효과가 크기 때문이다. 하지만, 세포고사된 항원이 면역치료에 좋은 항원으로 사용되지 못한다는 상반된 연구도 많은데 이는 세포고사된 종양세포를 수지상세포가 T 세포로 항원전달할 때 필요한 성숙인자(maturation factor)의 부족이 그 이유로 생각된다.

이 실험에서는 마우스 악성 흑색종주(B-16/F10)를 mitomycin-C (50 μg/ml)로 48시간 동안 처리하고 뜬 세포(floating cell)만 채취한 후, 2500 rpm에서 5분간 원심분리하고 상청액(supernatant)을 제거한 후 남은 침전물(pellet)을 세포고사 항원으로 사용하였다. 세포괴사된 항원은 마우스 악성 흑색종주를 각 3분간 냉동과 해동을 3번 반복한 후 2500 rpm에서 5분간 원심분리하고 상청액만을 채취하여 사용하였다. 이러한 세포고사와 세포괴사의 방법으로 처리한 마우스 악성 흑색종주를 배양 6일째의 미성숙 수지상세포와 함께 배양하여 항원을 전달하였다. 그 후 수지상세포를 성숙인자(CD40 ligand와 lipopolysaccharide)로 재자극한 후 표면항원(CD80, CD86, major histocompatibility complex(MHC) class II 분자)의 발현과 interleukin(IL)-12의 생성을 관찰하였고 T 세포 증식 정도를 조사하였다.

그 결과 각 실험군에서 단순히 항원만 처리하였을 때보다는 성숙인자를 추가한 경우, 더욱 증가된 표면항원의 발현과 IL-12 생성을 관찰할 수 있었다. 그리고 T 세포 증식 검사에서는 세포고사시킨 항원을 수지상세포와 배양한 후, 성숙인자로 재자극한 군이 세포고사시킨 항원과 배양하였으나 성숙인자로 재자극하지 않은 군이나 세포괴사시킨 항원을 사용한 군보다 더욱 강력한 T 세포 증식을 보였다.

이상의 결과로 수지상세포와 세포고사된 종양세포를 항원으로 반응시킨 후, 성숙인자를 추가하였을 때 표면항원의 발현과 IL-12 생성의 증가와 함께 강력한 T 세포 증식효과가 보임을 관찰하였다. 따라서 수지상세포에 세포고사된 항원을 처리한 후 성숙인자로 재자극하는 방법이 향후 더욱 강력한 종양면역 효과를 얻기 위한 방법으로 이용될 수 있으리라 생각된다.

[영문]

Dendritic cells(DCs) are antigen-presenting cells that induce T cell responses, which play roles in autoimmune and allergic diseases, transplantation, and cancer. DCs can present exogenous antigens to T cells not only in the context of major histocompatibility complex(MHC) class II, but also of MHC class I molecules, a phenomenon termed cross-priming. DCs efficiently present antigens derived from apoptotic cells, stimulating class I-restricted CD8+ cytotoxic T lymphocytes. However, DCs that have ingested apoptotic cells may not become activated, therefore inducing T cell tolerance unless further DC maturation is promoted by additional maturation factors.

In this study, the aim was to improve the methods for antigen preparation in DC immunotherapy. We postulated that apoptosis antigen would be a more efficient antigen with additional full DC maturation factors.

To remove the supernatant which included late apoptotic and necrotic cells, floating cells, which were previously treated with mitomycin-C for 48 hours, were centrifuged and the pellet was obtained. When the pellet was stained with annexin-V/propidium iodide, more than 80% of the cells were early apoptotic cells. We also observed an increased expression of MHC class II, CD80 and CD 86 molecule after antigen pulsing with maturation factors(CD40 ligand and lipopolysaccride(LPS)). However, there were no definite differences in the expression of DC surface molecules, according to the method of preparation of tumor antigens. DCs pulsed with apoptotic cells, followed by activation with CD40 ligand and LPS, had enhanced T cell stimulatory activity compared to other experiment groups. When cultured DCs, which were pulsed with apoptotic cells, received simultaneous stimulation with CD40 ligand and LPS, interleukin(IL)-12 secretion was increased compared to the group stimulated with necrotic cells.

These results reflect that DCs pulsed with apoptotic cells, with the supplement of additional maturation factors, induced enhanced T cell proliferation and secreted high levels of IL-12. It also suggests the utility of mitomycin-C induced apoptosis, supplemented with additional maturation factors, as a mean to generate efficient immunity for tumor in vivo.
