dc.description.abstract[한글] Human T cell에서 표현하는 Killer Cell Immunoglobulin-like Receptor (KIR)는 CD8+ T 세포 중에 활성화된 세포나 기억 세포에 주로 표현되어 있다. 그러나 T 세포에서 KIR의 구체적인 기능과 이의 표현 정도를 결정하는 기전은 아직까지 잘 알려져 있지 않고 있다. 최근의 연구 결과에 의하면, T 세포에서 표현되고 있는 KIR 역시 Natural Killer (NK) 세포에서와 유사하게 T 세포의 살상 능력을 억제하는 역할을 하는 것으로 알려져 있다. 또한 KIR의 표현 기전에 대한 최근의 연구 결과에 의하면, TCR를 통한 세포 활성화 과정 중에 이의 세포 표면 표현이 증가하는 반면 KIR의 cross-linking에 의해 그의 세포 표면의표현이 감소한다는 보고가 있다. 이는 평상시 정상 세포에서 표현하는 MHC class Ⅰ에 KIR가 결합함으로써 T 세포의 기능을 억제하지 않을 정도로 KIR의 표현 정도가 감소해 있다가, 그 T 세포가 제대로 된 기능을 수행하는 즉, T 세포의 활성화 신호가 오면 과활성화 되는 것을 억제하기 위해서 KIR의 표현이 증가할 것이라는 논리로 T 세포에서의 KIR의 기능과 그의 조절 기전을 설명할 수 있다. KIR+ T 세포가 인체 면역 조절 기전에 중요 기능을 수행할 것이라는 가능성과 중요성이 크게 부각됨에 따라 KIR의 표현 조절 기전을 구체적으로 규명해볼 필요가 있다. 흥미롭게도 T 세포의 여러 수용체들 (CD3g, CD4/8, CTLA-4)의 down-regulation에 Protein Kinase C(PKC)가 관여한다고 알려져 있는데 이와 유사하게 KIR의 internalization에도 PKC가 관여할 가능성에 대해 생각할 수 있다. 본 연구에서는 KIR의 표현을 조절하는 중요 분자와 이러한 효과를 가지도록 하는 KIR의 중요 부위들을 제시하여 KIR가 어떠한 기전으로 조절되는지를 규명하고자 하였다. 실험 결과 KIR 표현 Jurkat T 세포는 대조군에 비해, PMA에 의한 자극으로 KIR의 표현이 현저하 게 증가하였으며, 이는 PMA에 의해 활성화되는 PKC가 KIR의 표현에 영향을 미치는 것을 시사하는 결과이다. 실제로 CD8a와 KIR cytoplasmic tail의 융합 단백질을 표현하는 Jurkat T 세포주를 이용한 실험 결과 KIR cytoplasmic tail의 두 번째 PKC 결합 부위가 KIR의 표현을 조절할 가능성이 있음을 알아내었다. 또한 In vitro kinase assay로 PKC가 KIR cytoplasmic tail을 인산화시킴을 확인하였다. 마지막으로 conventional PKC isoform과 novel PKC isoform 저해제를 처리하였을 때 KIR 표현이 다른 양상을 보였으며, PKCa와 PKCd를 선택하여 PMA 처리 후 전체 단백질 양과 활성을 확인해본 결과 PKCa의 전체 단백질 양과 활성은 점차 감소하였고 반면 PKCd의 경우 전체 단백질 양과 활성은 감소하지 않았다. 이는 PKCa와 PKCd가 서로 다른 시점 혹은 서로 다른 부위에 서로 다른 기전으로 작용하여 KIR의 세포 외 표현을 조절함을 시사하는 결과이다. 예를 들면 PKCa는 KIR의 표현을 감소시키는 방향, 즉 degradation에 관여할 가능성, PKCd는 KIR의표현을 증가시키는 방향, 즉 trafficking에 관여할 가능성을 생각할 수 있다. 본 연구 결과를 통해, Jurkat T 세포주에서 KIR의 표현 기전을 이해하는데 도움을 주었으며, 앞으로 PKC가 KIR의 표현을 조절하는 기전과 여기에 관여하는 단백들에 대한 보다 구체적인 연구가 필요하다. [영문] Killer cell immunoglobulin-like receptors (KIRs) are expressed on human NK cells and a subset of CD8+ T cells, especially either on activated or memory phenotype T cells. Earlier studies have shown that KIR molecules expressed on T cells inhibit target cell cytolysis and cytokine release. Recently, it has been reported that KIR expression on T cells is dynamically regulated by TCR-mediated up-regulation and KIR-mediated down-regulation. These results suggest that, in the absence of T-cell receptor (TCR) engagement, KIRs expressed on CD8+ T cells are slowly down-regulated by KIR ligands expressed on antigen-presenting cells, and that the resulting expression level of KIR is no longer able to inhibit T-cell function. Whereas, TCR engagement activates T cells and re-induces functional level of KIR expression after ligand-induced down-regulation of KIR. To better understand the immunological function of KIR in T cells, the regulatory mechanism of KIR expression in Jurkat T cells was investigated in this study. Interestingly, several receptors (CD3, CD4/8, CTLA-4) of T cells are known to be down-regulated by PKC. Since the cytoplasmic tail of KIR contains putative PKC phosphorylation sites, it is tempting to speculate that KIR expression might be also regulated by PKC. As expected, KIR expression was strongly increased in the surface of Jurkat T cells after treatment of a potent PKC activator, phorbol myristate (PMA). Total cellular protein level of KIR was also increased after treatment of PMA. Analysis of Jurkat stable cell lines expressing CD8a and KIR3DL1 fusion proteins allowed to reveal that the second PKC substrate site of KIR cytoplasmic tail affect the expression of KIR. In support of this result, PKC appered to phosphorylate KIR cytoplasmic tail in in vitro kinase assay. In addition, PMA-mediated KIR up-regulation was almost completely blocked when either a conventional PKC inhibitor, Gõ6976, or a novel PKC inhibitor, Rottlerin was treated in media. These results suggest that KIR expression is regulated by PKC activation pathway, which includes both conventional and novel PKCs family members. Interestingly, total protein level and activity of PKCa decreased after treatment of PMA, while total protein level and activity of PKCd sustained. These results indicate that PKCa and PKCd regulate KIR expression in Tcells in a different manner. It seems likely that PKCa might decrease KIR expression by enhancing the degradation pathway, whereas PKCd might increase KIR expression by affecting the trafficking pathway. In conclusion, it is demonstrated that KIR expression in Jurkat T cells is regulated by PKC activation pathway. Further studies are necessary to better understand detailed molecular mechanism of the regulation of KIR expression. Particularly, it would be interesting to reveal which molecule(s) is involved in the regulation of KIR expression via PKC activation.-
