Abstract
[한글]원하는 유전자를 효율적으로 표적세포에 전달하기 위한 운반체로써 현재 가장 빈번하게 사용되고 있는 것 중의 하나가 레트로바이러스 운반체이다. 하지만 이는 형질도입의 효율이 낮고, 사람혈청에 의해 쉽게 비활성화되는 단점 등이 부각되면서 그 사용이 점차 제한되어지고 있다. 본 연구는 이러한 레트로바이러스의 단점을 이해하고 보완하여 유전자 형질도입율을 개선함을 목표로 하였다. 이를 위하여 본 연구는 먼저 레트로바이러스 감염에 의한 유전자 형질도입의 일차적 관문에 결정적인 영향을 주는 바이러스 막단백질의 구성을 변경시키거나, 다양한 종류의 화학적 첨가제를 사용하여 사람 암세포주들에 대한 유전자 형질도입 효율을 증대시키고자 하였다. 형질 변형된 레트로바이러스의 유전자 전달효율을 알아보기 위하여 우선 murine leukemia virus (MLV)를 기본골격으로 하는 pBabepuro 레트로바이러스 벡터에 LacZ 유전자가 도입된 pBabepuro/LacZ 레트로바이러스 벡터를 제작하였다. 다음으로 서로 다른 성질의 막을 가진 레트로바이러스를 생산해낼 수 있는 PA317, PG13, FLYRD18, Bing 및 293T/G/GP 포장세포주를 이용하여 pBabepuro/LacZ 바이러스를 생산하였다. 또한 양이온 리포솜을 포함한 여러 가지 화학적 첨가제의 도입 및 초원심분리로 바이러스를 농축시킨 후 각종 바이러스들의 유전자 전달의 효율성 증대 유무를 비교하였다. 그리고 레트로바이러스의 단점중의 하나인 비분열세포에 대한 낮은 유전자 형질도입율을 보완하기 위하여 MLV 대신 feline immunodeficiency virus (FIV) 벡터를 개발하여 그 특성을 규명하였다. FIV 벡터는 포장세포주의 도움없이 3 plasmids (포장용 플라즈미드, transfer 벡터, VSV-G를 발현하는 플라즈미드) cotransfection에 의하여 vesicular stomatitis virus (VSV)의 G 단백질을 막단백질로 가지는 FIV 벡터를 제작하였다. 실험결과 레트로바이러스로부터 유래한 막단백질을 가지는 레트로바이러스는 polybrene 등의 첨가물이 있어야만 세포내로 삽입될 수 있음을 확인하였다. 이러한 화학적 첨가물중 특히 리포솜이 우수한 효과를 보여주었으며, 특히 PG13/LacZ 바이러스의 경우 Lipofectamine이 존재하면 5 moi (multiplicity of infection)에서 SK-Hep1이나 U251-N 세포 등에서 첨가물이 없는 경우에 비하여 유전자 형질도입율이 100배 이상 증가하여 전체세포의 30%가 감염되는 효과를 보였다. 한편, VSV-G를 막단백질로 가지는 293T/G/GP/LacZ 바이러스는 어떤 다른 형태의 레트로바이러스보다 개선된 유전자 형질도입율을 보여 주었으며, 특히 뇌암세포주에서 유전자 형질도입율이 뛰어났다. VSV-G를 막단백질로 가지는 바이러스의 가장 큰 장점은 화학적 첨가물의 도움없이 효과적으로 세포를 감염시킬 수 있었다는 것이다. 이 결과를 바탕으로 in vivo에서의 유전자 형질도입율을 조사한 결과, 뇌암세포가 이식된 쥐에서 VSV-G 레트로바이러스를 단독으로 처리하는 경우 유전자 형질도입율이 10%에 도달하였다. 더불어 VSV-G 레트로바이러스에 의한 유전자 전달효율을 결정하는 주된 인자는 VSV-G 막단백질에 대한 수용체로 알려져 있는 phosphatidylserine 함량과 세포증식율에 기인하는 것을 알 수 있었다. 한편, 본 연구에서 자체적으로 제작한 FIV 벡터는 MLV를 근간으로 하는 기존의 레트로바이러스와는 달리 비분열세포와 분화된 세포에서 상대적으로 우수한 유전자 형질도입율을 보여주었다. 특히 바이러스의 vif 유전자내에 미성숙 종결코돈을 삽입시 킨 돌연변이 형태의 FIV는 세포에 대한 독성이 상당부분 제거되었음을 확인할 수 있었다. 또한 VSV-G를 막단백질로 가지는 MLV 또는 FIV는 초원심분리를 통해 고역가로 손쉽게 농축할 수 있으며 이러한 처리는 바이러스의 유전자 전달 효율을 더욱 향상시킴을 확인하였다. 따라서 이상의 결과들로부터 VSV-G를 포함하는 MLV 근간의 레트로바이러스의 in vivo 형질도입 우수성을 검증할 수 있었고, MLV 대신 FIV를 근간으로 하는 레트로바이러스는 기존의 레트로바이러스에 비해 비분열세포 및 분화세포에 유전자를 효율적으로 전달해줌을 알 수 있었다.

[영문]A retroviral vector constructed from murine leukemia virus (MLV) can only express transgenes in the cells undergoing mitosis, indicating the suitability as a delivery vehicle for cancer gene therapy. However, the retroviral vector, one of the most popular gene transfer vehicle, has recently experienced the limited usage due to its poor transduction efficiency in a variety of human cells. To overcome this drawback, we attempted to enhance the transduction efficiency by employing a variety of retroviral packaging cell lines and additive chemicals. For the complementation of low transduction efficiency of retrovirus to nondividng cells, we also explored the potential of FIV vector by modifying the packaging and transfer constructs. To evaluate the transduction efficiency, we constructed a pBabepuro/LacZ encoding LacZ as a reporter gene driven by the MLV LTR. Various kinds of retroviruses were produced by transfecting pBabepuro/LacZ into various packaging cell lines, such as PA317, PG13, FLYRD18, Bing or 293T/G/GP. For the generation of VSV-G pseudotyped FIV, 3 plasmids (packaging plasmid, transfer vector, VSV-G gene containing plasmid) were cotransfected into 293T cells. The addition of cationic chemicals exerted the most striking effect on the transduction efficiency with PG13-derived retrovirus. Especially, the transduction efficiency in SK-Hep1 or U251-N was substantially increased by the addition of Lipofectamine during the viral infection. 293T/G/GP/LacZ virus, a VSV-G pseudotyped retrovirus, was superior in transduction efficiency in most cancer cells, particularly in brain tumor cells, compared to that of other retroviruses, such as PA317-, PG13 or Bing-derived. In addition, VSV-G retrovirus could efficiently transduce human cancer cells regardless of the absence of chemical additives. Transduction efficiency of VSV-G retrovirus was mainly influenced by cell growth rate and phosphatidylserine expression level on the plasma membrane of target cells, which indicating both the relative growth rate and phosphatidylserine level were major determinants of transduction efficiency. Based on these results, we chose the VSV-G pseudotyped retrovirus to evaluate the transduction efficiency in brain tumor model. An average of 10% gene expression was routinely obtained exclusively in tumor mass when the concentrated virus was directly administrated to pre-established brain tumors in animal models. Modified FIV vector of vif gene mutated with premature termination was proven to be much less cytopathic to target cells and relatively superior in transduction efficiency to MLV-derived retrovirus in nondividing or differentiated cells. Taken together, these results suggested that the transduction efficiency of retrovirus in human cancer cells can be certainly improved when an appropriate viral vector or chemical additive is chosen.
