살아있는 상피세포에서 케라틴 및 비멘틴 중간세사의 운동성을 알아보기 위하여 세포 내에서 green fluorescent protein(GFP)-keratin 및 GFP-vimentin을 발현시킬 수 있는 cDNA를 만들어서 rat kangaroo의 신장 상피 세포인 PtK2 세포에 transfection시키고 살아있

는 세포 내에서 중간세사의 움직임을 시간의 경과에 따라서 관찰하였다. PtK2 세포 내에서 합성된 GFP-keratin 및 GFP-vimentin 단백질은 세포 내에 원래부터 존재하는 각각의 중간세사의 망상구조에 편입되어 형광을 나타내었다. 미세소관을 해중합(depolymerizatio

n)시키는 nocodazole 및 미세사를 해중합시키는 cytochalasin B를 PtK2 세포에 처리하면 케라틴 중간세사는 큰 변화 없이 망상구조가 유지되지만 비멘틴 중간세사의 망상구조는 붕괴됨으로써 비멘틴 중간세사의 망상구조를 유지하는 데에는 미세소관과 미세사의 완전

한 망상구조가 필요함을 확인하였다. 망상구조를 이루고 있는 케라틴 및 비멘틴 중간세사는 시간의 경과에 따라서 형태변화와 위치 이동을 보였다. 특히 케라틴 중간세사에서 특이한 파동성 운동이 관찰되었는데 nocodazole이나 cytochalasin B를 세포에 처리한 후에도 계속 관찰되었다. 중간세사 위에 형성된 광탈색 부위의 운동 속도는 비멘틴 중간세사에서 분당 0.15μm이었고 케라틴 중간세사에서는 분당 0.06μm이었다. 케라틴 중간세사의 운동은 nocodazole에 의해서만 억제되었으나 비멘틴 중간세사에서는 nocodazole과 cytochalasin B 모두에 의해서 운동이 억제되었다. 광탈색후 형광회복(fluorescence recovery after photobleaching)은 비멘틴 중간세사에서 원래 형광의 50%가 회복되는 시간(t 1/2)이 5.5분으로서 케라틴 중간세사에서의 106분 보다 20배 정도 빨랐으며, 케라틴 중간세사의 광탈색후 형광회복은 nocodazole이나 cytochalasin B의 영향을 받지 않는 반면, 비멘틴 중간세사의 광탈색후 형광회복은 두 가지 약물 모두에 의해서 억제되었다. 양쪽 끝에 free ends를 갖는 squiggle은 케라틴과 비멘틴 모두에서 관찰되었다. 비멘틴 squiggle의 운동 속도는 분당 3.5μm로서 케라틴 squiggle의 분당 0.24μm 보다 10배 정도 빨랐다.

비멘틴 squiggle은 관찰된 것들 중에서 36.2%가 움직였고, 이 중에서 58%가 세포막 쪽으로 이동하였으며, 케라틴 squiggle은 15%만이 움직였으며 이들 대부분이 핵 쪽으로 이동하였다. 케라틴과 비멘틴 squiggle의 움직이는 비율과 속도는 nocodazole에 의해서 억제되었다. 확장되는(spreading) PtK2 세포에서 케라틴 squiggle들은 세포의 말단 부위에서 서로 연결되거나 꼬이면서 합쳐져 점차적으로 망상구조를 형성하였으며 이러한 현상은 nocodazole이나 cytochalasin B를 처리해도 계속 관찰되었다.

이상의 결과로 미루어 볼 때 케라틴과 비멘틴 중간세사는 매우 역동적인 구조로서 비멘틴 중간세사의 운동성은 미세소관과 미세사에 매우 의존적인 반면 케라틴 중간세사의 운동성은 미세소관과 연관이 있을 것으로 생각되지만 그 관계를 단순하게 규명하기 어렵고, 미세사나 다른 세포 내의 요인들과 함께 복잡한 관계가 있을 것으로 생각된다.

In order to study the dynamic properties of keratin intermediate filaments(IF) and vimentin If in live epithelial cells, PtK2, cDNA constructs encoding green fluorescent Protein(GFP)-keratin 8, 14 and 18, and GFP-vimentin were made.

Following transfection, fluorescence microscopy revealed that GFP-keratin and GFP-vimentin was incorporated exclusively into endogenous keratin and vimentin If networks in PtK2 cells respectively. Tonofilaments exhibited bending and kinking

movements in time-lapse observations. Kinks propagated along tonofilaments and in some cases they completely disappeared. GFP-vimentin fibrils within these same cells also changed their shapes. Bleach areas on both vimentin fibrils and keratin tonofilaments moved during fluorescence recovery at an average rate of 0.15μm/min and 0.06μm/min respectively. Treatment of the cells with cytochalasin B or nocodazole reduced the rate of bleach area movement on vimentin fibrils, while keratin fibrils showed reduced rate of bleach area movement only with nocodazole.

Fluorescence recovery after photobleaching(FRAP) analyses showed that the fluorescence recovery rates were similar to GFP-keratin 8, 14 and 18 with the recovery half-time(f)f) of about 106min. In contrast, bleach area across GFP-vimentin fibrils recovered their fluorescence with t 1/2 of 5.5min. The recovery rate of GFP-keratin was not changed with cytochalasin B or nocodazole, while GFP-vimentin showed slower recovery with either of the drugs. Squiggles, short filamentous structures with two free ends, were found in the cell periphery About 15.5% of keratin squiggles translocated at an average rate of 0.24μm/min, all of which moved toward nuclei. About 36.2% of vimentin squiggles were motile with average speed of 3.5μm/min, 58% of which moved to the cell periphery. The ratio and rate of moving vimentin or keratin squiggles were reduced after nocodazole treatment. Keratin squiggles appeared in the peripheral regions of spreading cells and became interconnected with one another to form the typical configurations of the tonofibrils comprising keratin IF networks.

These results suggest that keratin and vimentin IF networks are highly dynamic and exhibit motile properties in live cells. Vimentin IF network has a close relationship to microtubule and microfilament network, while keratin IF network is thought to have a complected relationship to microtubule and possibly to

microfilament network.
