Functional expression of chemokine receptors in gastric cancer

Abstract

[한글]최근 키모카인과 키모카인 수용체의 작용이 암의 전이와 관련 있다는 일련의 보고들이 있어왔다. 본 연구에서는 여러 종류의 키모카인 수용체 중 암의 전이와 관련이 있는 것으로 보고된바 있는 CC, CXC 키모카인 수용체들이 다양한 위암세포주들에서 RT-PCR 방법을 이용하여 표현되는지를 확인한 결과 CCR4, CCR6, CCR10, CXCR4 등이 흔하게 발현됨을 확인하였고 이 들 중 기존에 고형암에서 보고된 바가 없는 CCR4를 이용하여 이는 western blot과 FACS 등을 통해 단백질 수준에서도 표현됨을 확인하였다. CCR4가 실제 위암세포와 조직에서 어떠한 형태로 분포하는지 알아보기 위해 면역조직화학염색을 통해 세포막에 존재하는 것을 확인하였고 이들 CCR4 수용체가 실제로 작용하는지를 알아보기 위해 그 리간드인 CCL17을 이용하여 migration assay, migration inhibition assay, proliferation assay, confocal microscopy 등을 시행한 결과 CCR4가 위암의 증식과 무관하게 위암세포의 운동에 관여하며 이러한 운동능력은 리간드의 억제에 반응하여 운동능력 또한 저하됨을 관찰하였다. 임상적으로 CCR4의 발현이 어떠한 의의를 갖는지 알아보기 위해 tissue miscroarray를 해 본 결과 위암조직에서의 CCR4 발현율이 환자의 연령이나 성 또는 조직학적 분화 등과는 관계없이 위암의 전이와 상관관계가 있으며 이러한 연관성은 다변량 분석에서도 동일한 결과를 얻을 수 있었다. 이러한 연구결과들에 기초하여 CCR4가 위암에서 고발현되며 임상적으로는 전이와 상관관계가 있어 CCR4를 억제함으로써 위암의 전이를 억제할 수 있는 가능성을 제시하였다.

[영문]There is increasing evidence that chemokines and chemokine receptors are casually involved in cancer metastasis. However, little is known about the possible role of chemokine receptors in gastric cancer metastasis. We here report that gastric cancer cells have CC chemokine receptors, CCR4, and that CCL17, a ligand of CCR4, that promote migration and proliferation, of gastric cancer cells that express its cognate receptor CCR4. CCR4 positive patients experienced tumor recurrence more than those with CCR4 negative. Logistic regression analysis showed that CCR4 positive adding to depth of tumor invasion and lymph node metastasis were independant risk factors for recurrence. Our findings suggest that the existence of CCR4, and its migration, may represent an important mechanism underlying metastasis of gastric cancer.
