Abstract

내이의 림프액은 크게 두 가지 성분으로 구성된다. 막성미로 내에는 내림프액이 들어 있고 막성미로와 골성미로 사이에는 외림프액이 들어있다. 내림프의 이온성분은 세포내액과 유사한 조성(고농도의 K+, 저농도의 Na+)을 가지고 있으며, 외림프액의 이온성분은 세포외액의 조성과 유사하다. (저농도의 K+, 고농도의 Na+) 내림프액이 세포내액과 유사한 농도를 유지하기 위해서는 계속해서 K+이 내림프액 내로 들어와야 하는데, 이러한 내림프액 내로의 K+의 이동은 변연세포의 기저외측부에서 주로 Na, K-ATPase와Na+-K+-2Cl- cotransporter(NKCC1)를 통하여 이루어진다. NKCC1은 상피세포와 비상피세포 모두에서 세포막을 통해 Na+와 K+가 두 개의 Cl-와 동시에 세포 내로 이동하여 전해질 균형을 맞추는 체계이며, 신장, 비점막 뿐만 아니라 전해질 이동이 있는 모든 기관에 분포한다. 본 연구의 목적은 이러한 NKCC1이 결핍된 백서에서 전정기관의 조직학적 변화를 알아보기 위함이다.

정상과 NKCC1이 결핍된 백서의 내이 조직 중에서 전정 기관을 채취하여 광학현미경을 이용하여 조직학적으로 비교 관찰하였다.

정상 백서에 비해 NKCC1이 결핍된 백서는 조직학적으로 전정과 반고리관은 불규칙한 모양을 보였고, 그 크기가 감소하였고, 골성 구조의 두께가 얇아져 있었다. 그러나 내이의 골격 자체는 대체로 그 형태를 유지하고 있었다. 막성미로가 위축되어 있었으며, 이로 인해 내림프강의 용적은 감소해 있었다. 반고리관의 팽대부릉의 점액성 물질은 소실되어 있었으며, 감각세포의 수도 감소되어 있었다. 이석 기관에서도 이석의 수가 정상에 비해 감소되어 있었다. 결론적으로 NKCC1이 결핍되어 있을 경우 전정기관의 조직학적으로 심한 변형이 있었으며, NKCC1은 전정기관에서 주요 부분의 형성과 전해질 균형에 중요한 역할을 할 것으로 사료된다.





[영문]The fluid of inner ear is composed two components; perilymph and endolymph. The ion concentration of endolymph is similar to intracellular fluid(low sodium and high potassium). For maintaining of this electrolyte homeostasis, potassium is continuously influxed to endolymph with ion transport system, such as Na, K-ATPase and Na+-K+-2Cl- cotransporter(NKCC1). NKCC1 is cation-coupled chloride transporter, which participates in vectorial slat transport, ion gradient homeostasis and cell volume regulation in diverse tissues. The purposes of this study were to investigate the morphology in the vestibular system of KCC1-deficient mice(knock out mice).

The NKCC1-deficient mouse line used for this study was Sla12a2. Histologic examination of inner ear especially vestibular system was performed in normal and knock out mouse.

Histologic exam revealed irregular shape and decreased size and wall thickness of vestibule and semicircular canal, collapse of membranous labyrinth and endolymphatic space, absence of gelatin material in crista ampullaris and reduced hair cells.

In summary, NKCC1 plays essential roles in the formation of critical components of vestibular apparatus and electrolyte homeostasis in vestibular organ.
