알파나프틸아민은 염료의 커플링제나 셀룰염으로 도판시 사용되며, 염료중간체, 농화학물질이나 살충제의 합성 중간체, 고무제품 등의 산화방지제, 석유제품, 제초제 생산 등에 사용되고 있다. 그리고 흡입, 섭취, 피부흡수 등의 경로를 통해 인체흡수가 일어나고, 인체발암성은 아직 명확하게 규명되지 않았지만 미국산업안전보건청에서는 발암성 의심물질로서 규제하고 있다.
염료공장에서 아조계 염료제조 중간체물질로 사용되는 공업용 알파나프틸아민의 순도를 가스크로마토그래피-질량분석계로 분석하고, 일반적으로 사용되는 요중 알파나프틸아민 분석방법들을 비교·평가하여 소변시료의 저장에 따른 안정성과 알파나프틸아민 취급근로자들의 소변시료를 분석, 평가하였다.
분석결과 공업용 알파나프틸아민의 순도는 각각 96.5±2.38%, 94.1±0.97%, 97.0±0.02%이었다.
고성능액체크로마토그래피에 의한 요중 알파나프틸아민은 물:아세토니트릴(65:35)에서 분리가 좋았으며, 전기화학검출기는 전위차 0.90V가 최적의 검출조건이었다. 요중 알파나프틸아민 전처리방법으로는 Sep-pak처리방법의 회수율이 상대적으로 우수하게 평가되었다.
This study was performed to analyze the purity of technical grade α-naphthylamine, to establish optimal analytical condition of α-naphthylamine in urine and to determine the urine sample of workers exposed to α-naphthylamine. The purity of technical grade α-naphthylamine were 96.5±2.38%, 94.1±0.97%, 97.0±0.02% by gas chromatography-mass selective detector. To analyze α-naphthylamine in urine, high performance liquid chromatography-electrochemical detector and gas chromatography-electron capture detector operating conditions have been optimized by preliminary experiment. In high performance liquid chromatography- electrochemical detector, the mobile phase was consisted of acetonitrile(35%) and water(65%), and the flow rate was maintained at 1.0 ml per minute. Optimal detective condition was. 9.0 V(10 nA/V) of electrochemical detector. The recovery of seq-pak treatment method was highly estimated as pretreatment of α-naphthylamine in urine. The free amine was isolated by gas chromatography-electron capture detector after basic hydrosis, sep-pak treatment, toluene elution and HFBA(heptafluoro-butyric anhydride) derivatization of urine. The recovery of α-naphthylamine in untie was 98.73±3.29% by gas chromatography-electron capture detector. The sensitivity was more higher than that of high performance liquid chromatography-electrochemical detector. Urinary α-naphthylamine was detected in only one worker among nine workers. The level of α-naphthylamine in urine was 6.42 ng/ml.
