항이디오타입 항체가 하이브리도마 세포의 항DNA 항체 생산에 미치는 영향

항이디오타입 항체는 항체의 가변부위의 항원 결정기 즉 이디오토프를 인지하는 항체로서 면역반응의 조절에 참여한다. 항이디오타입 항체는 생체에서 뿐만 아니라 시험관내에서 IgM 항체 생산 세포의 항원 수용체를 교차결합(cross linking)시킴으로써 항체 생산을 억제할 수 있다. 이때 항이디오타입 항체가 항체 생산을 억제하는 기전에 대해서는 확실히 알려져 있지 않다.

이에 이 연구는 항이디오타입 항체가 하이브리도마 세포의 항체 생산과 세포증식 및 면역글로불린 유전자의 전사수준에 미치는 영향을 관찰하여 항이디오타입 항체가 세포의 증식을 조절함으로써 항체 생산을 조절하는지 또는 면역글로불린 유전자의 전사과정을 조절하여 항체 생산을 조절하는지 알아보고자 하였다.

IgG2a 항DNA 항체를 생산하는 H24l 세포에서는 항이디오타입 항체가 항체 생산과 세포증식 및 면역글로불린 유전자의 전사수준에 영향을 미치지 않았다. 그러나, IgG3 항DNA 항체를 생산하는 2B8 세포에서는 항이디오타입 항체가 항체 생산을 증가시키고 세포증식을 촉진시켰다. 이때 항이디오타입 항체가 2B8 세포에서 전체 세포수에 대한 항체 생산 세포 수의 비와 면역글로불린 유전자의 전사수준에 영향을 미치지 않았다.

이상의 결과로 항이디오타입 항체는 IgG를 생산하는 하이브리도마 세포의 면역글로불린 유전자의 전사과정에 영향을 미치지 않으나 세포 증식을 촉진시켜 항체 생산을 증가시킬 수 있음을 알 수 있었다.

항이디오타입 항체가 IgM을 생산하는 항체 생산 세포에서 항체 생산을 억제하였던 종래 보고와 비교할 때 이 연구의 결과는 항체 생산 세포에서 항원수용체의 isotype의 차이에 따라 항이디오타입 항체가 항체 생산을 증가시킬 수 있음을 시사한다.

Anti-idiotype antibody(anti-id Ab) which recognize idiotope in the variable region of immunoglobulin(Ig) have been thought to regulate Ab production by B cells. Although regulatory mechanisms of anti-id Ab are not fully understood, it has been reported that anti-id Ab can suppress IgM production by lymphocytes or hybridoma cells in vitro, whereas it affects neither proliferation of the cells nor secretion of Ab.

In the present study it was investigated whether or net polyclonal anti-id Ab regulates the production of anti-DNA Ab by hybridoma cells. In addition, the effects of anti-id Ab on the proliferation of cells and on the transcription levels of Ig genes were studied to elucidate the mechanisms for regulation of anti-DNA Ab production in hybridoma cells.

Anti-id Ab did not affect the production of anti-DNA Ab by H24l cells(IgG2a producer), the proliferation rate and mRNA levels of Ig genes. Although the treatment of 2B8 cells (IgG3 producer) with anti-id Ab did net increase transcription levels of Ig genes, it increased production of Ab and stimulated cell proliferation.

Taken together these data indicate that anti-id Ab can stimulate Ab production by stimulation of cell proliferation. Furthermore, theme results and previous reports suggest that effects of anti-id Ab on production of Ab may be determined by isotype of Ab produced by B cells.
