골수조직에서 탈회과정이 면역염색에 미치는 영향에 관한 연구

Abstract
[한글]

본 연구는 골수의 조직 및 세포형태를 잘 보존하면서 최상의 면역 염색성을 나타내는 고정 및 탈회조건을 구하기 위하여 시행하였으며, 부검시 늑골을 채취한 후 고정액과 탈회액 및 탈회기간을 달리하여 lysozyme과 leukocyte common antigen(LCA)에 대한면역조직화학적 염색을 시행하고 그 결과를 비교하였다. 고정액으로는 10% 중성 포르말린과 B5용액을 선택하고, 탈회액으로는 0.5M EDTA, Plank-Rychlo용액, Jenkin용액, 5%질산, von Ebner용액을 사용하였으며, 면역염색은 peroxidase label led streptavidin biotin(LSAB)법

을 이용하였다.

결과를 요약하면 다음과 같다.

1. 탈회액의 종류에 관계없이 10%포르말린에 고정한 경우에 보다 좋은 면역 염색성을 보였다.

2. 전반적인 조직구조를 가장 잘 보존한 탈회액은 Plank-Rychlo용액이었고 그 다음은 0.5M EDTA였다.

3. 탈회액에 따른 적정한 시간동안 탈회한 경우 면역 염색성이 가장 좋은 것은 0.5M EDTA였고 그 다음 Plank-Rychlo용액이었으며, von Ebner용액이 가장 나쁜 결과를 나타내었다.

4. LCA 항원성은 10% 포르말린용액으로 고정하고 0.5M EDTA로 탈회한 경우에만 보존되었다.

5. 0.5D EDTA를 사용한 경우에는 3일이 최적 탈회기간이었고, Plank-Rychlo용액의 경우에는 1일이었다.

6. Jenkin용액과 5% 질산용액을 사용한 경우 각각 12시간 및 4시간 탈회할 때에는 조직구조가 비교적 잘 보존되었으나 탈회가 고르지 않고 면역 염색성도 고르지 않았다.

이상의 결과를 종합하면 0.5M EDTA가 비록 다른 탈회액보다 긴 탈회시간을 요하지만 면역 염색성이 가장 우수한 것을 알았으며, Plank-Rychlo용액도 권장할 만한 탈회액이라고 생각하였다.

[영문]

The procedure of evaluating the suitable fixation and decalcification preserving the best immunoreactivity and histological structure of bone marrow was examined. The comparison of results of iumunohistochemistory for lysozyme and leukocyte common antigen(LCA) were studied using various fixatives, decalcifying agents and the time period of decalcification with extration from normal rib bone during

autopsy. In this study, 10% neutral formalin and B5 solution were selected for fixatives, 0.5M EDTA, Plank-Rychlo solution, Jenkin's fluid, 5% nitric acid and von Ebner's fluid were used for decalcifying agent and peroxidase label led streptavidin-biotin(LSAB) method for immunostain.

The results are as follows:

1. The best suitable immunoreactivity were found in tissues placed in 10% formal in fixative regarding decalcifying agent.

2. Tissues decalcified in Plark-Rychlo solution showed the best preservation of histological structure. 0.5M EDTA was the next decalcifying agent.

3. Immunoreactivity of tissues decalcifing most obviously in 0.5M EDTA solution and the best alternative solution for 0.5M EDTA was Plank-Rychlo. But the loss of immunoreactivity was evident in tissues placed in von Ebner's fluid.

4. LCA antigenicity was preserved only in tissues fixed in 10% formalin solution and decalcifided in 0.5M EDTA.

5. Decalcification was good in tissues placed in 0.5M EDTA for 3 days. Plank-Rychlo solution for 1 day.

6. Preservation of histological structure of tissues was usually good in Jenkin's fluid for 12 hours, otherwise tissues were partially decalcified and immunostained. As for 5% nitric acid, 4 hours was needed to occur the same result of the former.

According to the result, tissues decalcified in 0.571 EDTA solution yield the best immunoreactivty though it requires relatively more time period of decalcification than others. Plank-Rychlo solution appears to be another recommandable decalcifier.
