흰쥐 말초혈액 림프구는 세포막에 vasoactive intestinal polypeptide(VIP)수용체 및 아드레날린성 β수용체를 가지고 있다. 각 수용체에 대한 신호변환 기전은 모두 흥분성 guanine nucleotide 결합 단백(G단백)의 매개에 의한 adenylyl cyclase 활성화로 세포내 cyclic adenosine monophosphate(cAMP) 농도가 증가하여 나타나는 것으로 알려져 있다. Propranolol은 아드레날린성 β수용체에 대한 경쟁적 봉쇄제이나 림프구에서 VIP에 의한 cAMP 농도 증가 및 기질 단백 인산화를 억제함이 보고되었다. 본 연구에서는 이러한 림프구에서의 VIP의 작용에 대한 propranolol의 억제 기전으로서 propranolol이 신호변환 경로상 VIP 수용체, G단백 또는 adenylyl cyclase에 직접 억제작용을 나타낼 가능성, 또는 propranolol이 가지고 있는 세포막에 대한 비특이적 작용에 의한 가능성에 대하여 알아보고자 하였다. 흰쥐 말초혈액 림프구를 이용하여 propranolol 비처치시와 전처치시의 VIP 수용체 결합 실험 및 isoproterenol, VIP, forskolin 투여시 propranolol 또는 timolol 전처치 유무에 따른 cAMP 농도 증가 변동 및 VIP의 cAMP 농도 증가 효과에 대한 lidocaine의 영향 등을 관찰하였으며, 다음과 같은 결과를 얻었다.

1. VIP 수용체 결합 실험에서 Propranolol은 VIP 수용체의 친화도나 밀도에 영향을 주지 않았다.

2. VIP에 의한 림프구 세포내 cAMP 증가는 propranolol에 의해 유의하게 억제되었으나, timolol에 의하여는 억제되지 않았다.

3. Isoproterenol은 림프구 세포내 cAMP를 증가시켰으며, 이러한 증가는 propranolol 및 timolol에 의하여 모두 유의하게 억제되었다.

4. Forskolin에 의한 림프구 세포내 cAMP 증가를 propranolol은 억제하지 못하였다.

5. Lidocaine은 VIP에 의한 림프구 세포내 cAMP증가를 봉쇄하지 못하였다.

이상의 실험결과로 보아 흰쥐 말초혈액 림프구에서 propranolol의 VIP에 대한 억제 효과는 VIP에 의한 신호변환 과정 중 VIP 수용체 또는 adenylyl cyclase에 대한 직접적인 작용보다는 그 중간 과정에 영향을 준 결과로 생각되며, 이는 propranolol의 세포막에 대한 안정화 작용과는 별개의 효과로 생각된다.

Vasoactive intestinal polypeptide(VIP) and β-adrenergic agonists have immunomodultory effects on the peripheral blood lymphocytes of rat through their own receptors. Beth of them utilize the same signal transduction pathway. That is, the stimulatory guanine nucleotide binding protein(G protein) mediates the receptor-adenylyl cyclase coupling, producing intracellular increase of cyclic adenosine monophosphate(cAMP ). In the previous experiment, propranolol, a β-adrenergic receptor blocker, inhibited the VIP-induced protein phosphorylation in lymphocytes. However, propranolol could not block the effect induced by forskolin.

Therefore, this study was designed to elucidate the mechanism of the inhibitory action of propranolol on the effects of VIP. Using peripheral blood lymphocytes of rats, the effect of propranolol on the receptor binding characteristics of VIP was

observed. And the effects of propranolol were compared to the effects of timolol on the cAMP increase induced by isoproterenol, VIP or forskolin.

The results obtained are as follows.

1. Receptor binding study showed no significant differences in the affinity or density of VIP receptor between the control and propranolol-pretreated groups.

2. VIP-induced increase of cAMP was inhibited by propranolol, but not by timolol.

3. Both propranolol and timolol suppressed the isoproterenol-induced cAMP increase.

4. Propranolol did not inhibit the increase of cAMP stimulated by forskolin.

5. Lidecaine did not block the VIP-induced cAMP increase.

These results show that the inhibitory mechanism of propranolol is not related to β-adrenergic receptor or its membrane stabilizing effect, and it is suggested that propranolol can block the effects of VIP by inhibiting the intermediate step

between the VIP receptor and adenylyl cyclase.
