Ascorbic acid 투여 경로에 따른 방수내 ascorbic acid 농도 변동

dc.description.abstract[한글] 혈청농도보다 15-69배 높은 농도로 존재하는 방수내 ascorbic acid는 항산화제로 작용하여 유해한 free radical로부터 안조직을 보호하고 자외선을 흡수하며 섬유주내의 단백질 형성에도 관여한다고 한다. 그러므로 방수내 ascorbic acid의 농도가 떨어지면 백내장, 녹내장등의 안질환의 한 원인이 될 수도 있기 때문에 ascorbic acid의 농도를 높인다면 이러한 안질환의 발생을 막을수 있다고 여겨지고 있다. 본 연구에서는 ascorbic acid투여 방법에 따른 방수와 혈청 ascorbic acid 농도 변동을 분석하여 어떤 투여 방법이 적절한지를 알아보고자 하였다. 가토를 대상으로 ascorbic acid를 정맥주사 하였을 때 투여용량의 증가에 따라 혈청 농도가 비례하여 증가하였으며 이에따라 방수내 농도도 증가하여 150mg 주사시 혈청농도는 약4배인 13.9μg/ml, 방수내 농도는 2배인 72.9μg/ml로 증가하였으나 그후 주사용량을 계속 증가시킴에 따라 혈청내 농도는 계속 증가하였으나 방수내 농도는 포화상태를 보여 별 증가가 없었다. 결막하주사로 투여시에는 주사용량에 비례하여 혈청 농도및 방수내 농도가 포화상태없이 계속적으로 증가하여 200mg 주사시 혈청 39.4μg/ml, 방수내 101.2μg /ml 이었다. 이상의 결과로 방수내 ascorbic acid농도를 높이는 방법으로는 정맥주사보다 결막하주사가 더 효과적인 방법이라는 것을 알 수 있었으며 본 연구 결과가 향후 방수내 ascorbic acid농도 변동에 따른 안질환의 발생 연구에, 그리고 안조직에서 ascorbic acid의 보호 역할에 대한 연구에 도움이 되리라 생각한다. [영문] Ascorbic acid in aqueous humor, of which the concentration is 15-69 times higher than in serum, protects ocular tissues from free radicals as an antioxidant and absorbs ultraviolet rays and modulates protein synthesis in the trabecular meshwork. Low concentrations of ascorbic acid in aqueous humor may be one of the causes of ocular diseases such as cataract and glaucoma. Thus it is regarded that prevention of ocular disease can be achieved by increasing the concentration of ascorbic acid in aqueous humor. In this study, an attempt was made to determine the most effective method of ascorbic acid ministration. Furthermore the change of concentration of ascorbic acid according to each method was analyzed. After intravenous injection of ascorbic acid in the rabbit, the concentrations of ascorbic acid in serum and aqueous humor increased in proportion to the injected dose in which injection of 157mg elevated the concentration in serum to 13.7μg/ml, and aqueous humor to 72.9μg/ml. After increasing the dosage to 400mg, the concentration increased to 79.3μg/ml in the serum and 88.0μg/ml in the aqueous humor. On the other hand, the ascorbic acid in aqueous humor and in serum increased in proportion to the dosage of subconjunctivally injection without saturation. When given subconjunctivally 200mg, the serum ascorbic acid concentration was 39.4μg/ml and that of aqueous humor was 101.2μg/ml. These results revealed that the method of subconjunctival injection is more effective than intravenous injection in increasing the concentration of ascorbic acid in aqueous humor and these findings should be helpful as a basis for performing further stu야es about the protective role of ascorbic acid and relation of changes of concentration of ascorbic acid in aqueous humor in causes of ocular diseases-
