dc.description.abstract[한글] 타액선 세포막에는 여러가지 수용체 즉, 무수카린성, 아드레날린성, 그리고 peptidergic 수용체 등이 있으며 특히 무수카린성 수용체를 자극하면 세포내 calcium (Ca**2+)이 증가된다. 이 결과 Ca**+ -activated K**+ -channel이 열려 세포내 K**+ 이 세포외부로 나오는데 (K**+ -release), 이와같이 K**+ 통로 (channel)의 전도도 (conductance)를 조절하는 세포내 Ca**2+ 은 1) 세포막을 통한 세포외부로 부터의 Ca**2+ 의 유입과, 2) inositol 1,4,5-trisphosphate (IP**3) 의존성 Ca**2+ 저장고 (IP3-sensitive Ca**2+ pool, ISCP)로부터 Ca**2+ 유리에 의하여 증가되지만, 최근에는 IP**3 비의존성 Ca**2+ 저장고 (IP3-insensitive- Ca**2+ pool, IICP)가 세포내 Ca**2+ 농도를 직접적으로 조절할 뿐만 아니라 caffeine(CAF)-sensitive Ca**2+ pool (CSCP)이 존재하는 것으로 알려져 있어, 이하 선 선세포에서의 Ca**2+ -activated K**+ -release에 관여하는 CAF의 역할을 규명하고자 본 실험을 시행하였다. 흰쥐 (Sprague Dawley) 암컷의 이하선을 분리한 후 미세 조직절편을 만들었다. 이하선 조직절편을 perifusion chamber에 넣고 Krebs-Ringer bicarbonate (KRB) 용액을 관류시키면서 1) tetraethylanmmonium chloride (TEA)에 의한 carbachol (CCh)-induced K**+ -release의 변화, 2) CAF이 CCh-induced K**+ -release에 미치는 영향, 그리고 3) Na**+ -K**+ pump ATPase의 억제재인 ouabain이 CCh과 CAF을 동시 투여한 후 CAF을 제거했을 때 K**+ -release에 미치는 영향 등을 K**+ 감지전극 (K**+ -sensitive electrode)으로 계측하고 이를 analog-digital converter (A/D converter)를 통해 personal computer (PC)에 연결하여 연속 측정하였다. 이와같이 측정된 K**+ -release는 세포네 Ca**2+ 변화로 간주하였고 이를 비교 분석하여 다음과 같은 결과를 얻었다. 1. 이하선 조직 절편의 CCh (10**-5 M)-induced K**+ -release는 Ca**2+ -activated K**+ -channel 억제재인 TEA (5 mM)에 의하여 52.35 ± 3.61% 감소되었다. 2. CAF (10 mM) 전처치 후 CCh-induced K**+ -release는 CCh 단독 투여에 의한 효과보다 69.95 ± 7.25% 감소되었다. 3. CCh과 CAF을 동시 투여한 후 CAF을 제거하였을 때 K**+ -release는 CAF이 존재하는 경우에 비하여 44.30 ± 7.19% 줄었다. 4. CCh과 CAF 동시 투여 후 CAF만을 제거하였을 때 K**+ -efflux는 CCh과 CAF이 같이 존재하는 조건과 비교하여 현저히 감소하였으나 Ca**2+ 존재와는 무관하였다. 5. CCh과 CAF을 동시 투여한 후 CAF을 제거했을 때 증가된 K**+ -release는 세포외부에 Ca**2+ 존재와 관계없이 동일할 뿐아니라 (P>0.05) Ca**2+ 통로 억제재인 Ni**2+ (0.8mM)과 Mn**2+ (10**-4 M)는 CCh과 CAF 동시 투여한 후 CAF을 제거했을 때 증가된 K**+ -rel ease에 아무런 영향을 미치지 않았다 (P>0.05). 6. Na**+ - K**+ ATPase의 강력한 억제재인 ouabain (1 mM)은 CCh과 CAF을 동시 투여한 후 CAF을 제거시 야기된 K**+ -release를 91.8 ± 1.69% 감소시켰다. 이상의 실험결과로 미루어 CAF은 이하선 선세포에 있는 IP₃ 의존성 Ca**2+ 저장고에서의 Ca**2+ 유리를 억제하므로써 K**+ -release를 감소시키는 것으로 생각되지만, CCh과 CAF을 동시에 투여한 후 CAF 제거시 유발된 K**+ -release의 증가율이 CCh 단독으로 나타난 K**+ -release의 증가율과 다르다는 사실은 ISCP과 CSCP과는 별개의 Ca**2+ 저장고가 세포내에 존재할 가능성을 암시하는 것이다. 아울러 CCh과 CAF을 동시에 투여한 후 CAF만을 제거하였을 때 증가된 K**+ -release가 ouabain에 의하여 현저히 감소되어 CCh-induced K**+ -release는 세포내 Na**+ 농도와 밀접한 관계가 있는 것으로 생각되나 세포내 Ca**+ 증가와 관련된 CAF 및 Na**+ 농도와의 상호관계는 앞으로 추구하여야 할 과제로 평가된다. [영문] In acinar cells from rat salivary glands. there are several receptors, such as muscarinic, adrenergic, and peptidergic receptor etc. Of these receptors, the stimulation of muscarinic receptor results in the increase in the intracellular calcium (Ca**2+) level via inositol trisphosphate (IP^^3). Thus, a rise of Ca**2+ activates K**+ channels in the basolateral membrane and Cl- channels in the apical membrane, which in turn, evokes the salivary secretion. Cytosolic Ca**+ modulating K**+ channel is mobilized via two different routes: 1) Ca**+ entry through the Ca**2+ channel, 2) Ca**2+ mobilization released from the intracellular Ca**2+ pool. In the regards of Ca**+ mobilization, the existence of IP^^3-insensitive Ca**2+ pool (IICP) has bean reported. In addition, close intercommunication between caffeine (CAF)-sensitive Ca**2+ pool (CSCP) and IP^^3 -sensitive Ca**2+ pool (ISCP) has been proposed to account for the mechanism of intracellular Ca**2+ mobilization. The aims of the present study to examine 1)tetraethy1ammonium chloride (TEA) effect on carbachol (CCh)-induced K**+ -release, 2)the changes in CCh-induced K**+ -release in response to CAF, and 3) alterations in CCh-induced K**+ -release by the exposure to the ouabain. Rat (Sprague Dawley) parotid gland was dissected out and minced the tissue with scissor. The minced tissue was placed in the perifusion chamber wi th the K**+ -sensitive electrode (Orion Research), followed by perifusing the parotid acini with Krebs-Ringer bicarbonate (KRB) solution. To stimulate the parotid acini, CCh was perifused with KRB solution. Then K**+ -efflux from parotid acini was measured using K**+ -sensitlve electrode and interpreted in terms of intracellular ca**2+ concentration. 1. In rat parotid acini, TEA (5 mM), the blocker of Ca**2+ -activated K**+ -channel, inhibited CCh (10**-5 M)-induced ^^K+-release by 52.35 ± 3.61%. 2. CCh-induced K**+ -release from acini under the condition of prior exposure to CAF(10 mM) was reduced by 68.95 ± 7.25% of the effect caused by CCh alone. 3. Removal of CAF after exposure to CCh and CAF caused another increase in CCh-induced K**+ -release, which corresponded to 44.30 ± 7.19% of initial condition(CCh plus CAF). 4. CCh-induced K**+ -efflux (initial rate of K**+ -release) by the removal of CAF was remarkably blunted compared to that in the presence of the CCh and CAF, however, such an CCh-induced K**+ -efflux was independent on the Ca**2+ in the external medium. 5. The increase in CCh-induced K**+ -release by removal of CAF from the solution with CCh and CAF, was not only related to external Ca**2+, but also not affected by the exposure to Ni**2+ and Mn**+ which are Ca**2+ channel blockers. 6. Ouabain (1 mM), strong inhibitor of Na**+/K**+ ATPase, blocked CCh-induced K**+ -release caused by removal of CAF by the 91.80 ± 1.69%. Taken all together, CAF seems to inhibit Ca**2+ mobilization from the ISCP, following the reduction of CCh-induced K**+ -release. Otherwise, after stimulating the parotid acini with CCh and CAF, the omission of CAF resulted in the increase in CCh-induced K**+ -release, while decreased CCh-induced K**+ -efflux. However, such an increase was completely inhibited by ouabain. Consequently, these facts might suggest the possibility that the parotid acini have a another intracellular Ca**2+ pool, furthermore, Ca**2+ -activated K**+ -release may be closely associated with the intracellular Na**+ concentration.-
