콜레라 독소는 Vibrio cholerae에서 생성되는 분자량 84,000 daltons인 장내독소이다.

콜레라 독소는 소장 점막 세포에서 monosialoganglioside (GM^^1) 수용체와 결합하여 sti

mulatory guanine nucleotide-binding protein(Gs)에 ADP-ribosylation을 촉매시킴으로써

adenylate cyclase를 지속적으로 활성화시키고, 그 결과 c-AMP가 증가됨으로 장내로의

전해질 분비가 나타난다고 보고되고 있다. 그러나 임상적으로 protein kinase 활성을 억

압하는 chlorpromazine이 콜레라 독소에 의한 장 분비 증가를 억제한다는 것이 알려지면

서 콜레라 독소 작용에 있어서 protein kinase의 역할이 대두되고 있다. 실제로 chlorpro

mazine, dibucaine 또는 인지질에 작용하는 약물이 calcium-activated phospholipid-depe

ndent protein kinase(protein kinase C) 작용을 억제한다고 보고되고 있다.

따라서 이번 실험에서는 콜레라 독소를 적출한 고양이 이자에 투여한 후 이자 외분비

항진 기능에 대한 chlorpromazine, diazepam, dibucaine의 영향을 검색하여 콜레라 독소

작용에 대한 protein kinase의 관련성을 추구하고자 하였다.

실험 동물로는 몸무게 2kg 안팎의 잡종 고양이를 암수 구별없이 사용하였으며 secobarb

ital로 마취한 다음 이자를 적출하여 Krebs-Ringer bicarbonate buffer(KRB8)로 관류시켰

다. 콜레라 독소는 1 μg/ml농도로 KRBB에 혼합하여 90분간 재순환시키면서 관류하였고

롤레라 독소 투여후 이자액 유출량이 일정하게 유지되었을 때 chlorpromazine, diazepam

또는 dibucaine을 40분간 관류하여 그 효과를 관찰하였다.

실험 성적을 요약하면 다음과 같다.

1. 콜레라 독소를 1 μg/ml농도로 97분간 관류하였을 때 관류 60∼90분 후부터 이자액

유출량이 증가하기 시작하였으며, 독소 관류 중단 후 이자액 유출량은 현저히 증가하여 2

∼4시간에 고평부(plateau)에 도달하였다. 그러나 amylase기초 분비 및 CCK자극 분비는

별 변동이 없었다.

2. 콜레라 독소로 유발된 이자액 유출량은 chlorpromazine에 의해 감소되었으나. CCK자

극에 의한 amylase 분비량은 차이가 없었다.

3. 콜레라 독소로 유발된 이자액 유출량은 diazepam 10**-5 M농도에서는 증가되었으나

10**-4 M농도에서는 감소하였다. 그러나 CCK자극에 의한 amylase 분비량은 증가 경향을

보였다.

4. 콜레라 독소로 유발된 이자액 유출량은 dibucaine 10**-6 M 농도 관류시 감소하였고

, amylase분비도 감소하였다.

이상의 결과로 보아 콜레라 독소에 의한 이자 외분비 항진 기전에는 adenylate cyclase

활성화뿐만이 아니라 protein kinase부위에서의 조절이 관여된다고 생각한다.

[영문]

Cholera toxin is known to activate adenylate cyclase via covalent modification of

stimulatory guanine nucleotide binding protein. As a result, it causes persistant

increase in cellular cyclic AMP, which causes a sustained increase in intestinal

and pancreatic fluid secretion. Recently it has been reported that chlorpromazine,

which inhibits protein kinase, can inhibit cholera toxin-induced intestinal

secretion. Therefore, it is speculated that the action of cholera toxin is related

to the activation of protein kinase.

In an effort to probe the possible intermediary mechanism of cholera

toxin-induced secretion in exocrine pancreas, the effects of three protein kinase

inhibitors, i.e., chlorpromazine, diazepam and dibucaine, on the cholera

toxin-induced secretory process were evaluated in the isolated saline perfused

preparation of cat pancreas. The pancreas was isolated from mongrel cats of either

sex weighing about 2 kg, and perfused with Krebs-Ringer bicarbonate buffer (KRBB)

gassed with a mixture of 95% O^^2 and 5% CO^^2 to maintain the pH at 7.4. Cholera

toxin was added to the perfusion fluid at the concentration of 1 μg/ml and was

recirculated into the pancreas for 90 minutes. Chlorpromazine, diazepam or

dibucaine were added to the KRBB and perfused for 40 minutes after pancreatic

exocrine secretion was reached plateau by cholera toxin. CCK-8 (5ng) was given as a

bolus injection to see the effects of cholera toxin on the pancreatic enzyme

secretion. The pancreatic juices were collected every 10 minutes and analyzed for

its volume and amylase activities.

Results obtained are summarized as follows.

1. The pancreatic flow began to increase at 60∼90 min, by perfusion of cholera

toxin and reached plateau at 2∼4 hours after cessation of toxin perfusion. But the

amylase secretion was little changed.

2. Cholera toxin-induced pancreatic flow was inhibited by 10**-5 M of

chlorpromazine, but CCK-8 evoked amylase secretion was little affected.

3. Cholera toxin-induced pancreatic flow was inhibited by 10**-4 M diazepam,

while the perfusion of 10**-5 M diazepam enhanced the secretion. CCK-8-stimulated

secretion of amylase was tended to be enhanced by diazepam.

4. Cholera toxin-induced pancreatic flow was inhibited by 10**-5 M dibucaine, and

CCK-8-stimulated secretion of amylase was also inhibited.

With the above results, it is suggested that modulation of protein kinase, in

addition to increase in c-AMP, is involved in the stimulatory effects of cholera

toxin on the exocrine pancreatic secretion.
