dc.description.abstractEndothelial cell is an interphase between blood and tissue that acts as a media for active interactions between plasma and intracellular environment for homeostasis. Proteins in blood can have an effect on the proteins of endothelial cells. However, endothelial cell proteins influenced by blood components according to the aging process are not known yet and it is difficult to identify biomarkers in blood component because that is low abundant proteins. This study is to identify human microvascular endothelial cell(HDMEC) proteins changed by human sera from aged man. HDMECs were subcultured with sera from healthy children(< 6 yrs), adults in their 4th decades, and elderly in their 7th to 8th decades. We applied ProteomeLabⓡ PF2D system that uses chromatofocusing according to pI and non-porous reversed-phase column chromatography according to hydrophobicity. we could obtain 2D proteome map by PrteoVue software. Using nano LC-MS/ MS, we identified differential protein. As the result, we could identify adipocyte-fatty acid binding protein(A-FABP), which showed highly increased expression on HDMEC treated with adult to elderly sera. The further study about the delicate functional mechanism of A-FABP on endothelial cell aging will be needed.-
