Antifibrotic TSG-6 Expression Is Synergistically Increased in Both Cells during Coculture of Mesenchymal Stem Cells and Macrophages via the JAK/STAT Signaling Pathway

dc.description.abstract | 전염증성 사이토카인 (pro-inflammatory cytokine)인 종양괴사인자 알파 (tumor necrosis factor-α, TNF-α)와 인터루킨-1베타 (interleukin-1β, IL-1β)는 TNF-α 자극 유전자 6 (TNF-α-stimulated gene 6, TSG-6)의 발현을 촉진한다. 그러나 중간엽 줄기세포 (mesenchymal stem cells)에서 TSG-6의 발현을 위한 최적의 조건에 대한 연구는 부족한 상태이다. 본 연구에서는 지방조직 유래 중간엽 줄기세포 (adipose tissue-derived stem cells)와 공동 배양된 대식세포 (macrophages)의 분극화 상태에 따라 TSG-6의 발현이 달라지는지 확인하고, 지방조직 유래 중간엽 줄기세포에서의 TSG-6 발현의 최적 조건을 분석하였다. M0, M1 및 M2 대식세포와 공동 배양된 지방조직 유래 중간엽 줄기세포에서 TSG-6의 발현이 증가하였고, 그 중에서 M1 대식세포와 함께 배양했을 때 가장 많은 증가를 보였다. 지방조직 유래 중간엽 줄기세포에 TNF-α, IL-1β, 인터페론 감마 (interferon-γ, IFN-γ), 그리고 지질다당류 (lipopolysaccharide, LPS)를 조합하여 처리했을 때 TSG-6의 발현이 매우 증가하였다. 또한 IFN-γ와 LPS 처리에 의한 STAT (signal transducer and activator of transcription) 1/3의 인산화가 확인되었으나, TNF-α나 IL-1β에서는 확인되지 않았다. STAT1/3의 활성화는 TNF-α/IL-1β에 의존적으로 TSG-6의 발현을 증가시켰고, JAK inhibitor는 지방조직 유래 중간엽 줄기세포와 대식세포 모두에서 TSG-6 발현을 억제하였다. TSG-6는 LX-2 간성상세포에서 전환성장인자 베타(Transforming growth factor-β, TGF-β)에 의한 Smad3 인산화를 억제하여 α-smooth muscle actin의 발현을 감소시켰다. 또한 지방조직 유래 중간엽 줄기세포 및 M1 대식세포와 공동 배양했을 때, TGF-β에 의해 유도된 LX-2 세포의 섬유화 활성이 매우 감소하였다. 이러한 결과는 STAT1/3 활성화를 통해 TSG-6 발현을 증가시킴으로써 포괄적인 염증의 미세 환경이 지방조직 유래 중간엽 줄기세포의 치료 효과를 결정하는데 중요한 역할을 할 수 있음을 시사한다. The pro-inflammatory cytokines tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-1β upregulate TNF-α-stimulated gene 6 (TSG-6); however, current knowledge about the optimal conditions for TSG-6 expression in mesenchymal stem cells (MSCs) is limited. Here, we investigated whether TSG-6 expression varies depending on the polarization state of macrophages co-cultured with adipose tissue-derived stem cells (ASCs) and analyzed the optimal conditions for TSG-6 expression in ASCs. TSG-6 expression increased in ASCs co-cultured with M0, M1, and M2 macrophages indirectly; among them, M1 macrophages resulted in the highest increase in TSG-6 expression in ASCs. TSG-6 expression in ASCs dramatically increased by combination (but not single) treatment of TNF-α, IL-1β, interferon-gamma (IFN-γ), and lipopolysaccharide (LPS). In addition, phosphorylation of signal transducer and activator of transcription (STAT) 1/3 was observed in response to IFN-γ and LPS treatment but not TNF-α and/or IL-1β. STAT1/3 activation synergistically increased TNF-α/IL-1β-dependent TSG-6 expression, and JAK inhibitors suppressed TSG-6 expression both in ASCs and macrophages. In LX-2 hepatic stellate cells, TSG-6 inhibited TGF-β-induced Smad3 phosphorylation, resulting in decreased α-smooth muscle actin (SMA) expression. Moreover, fibrotic activities of LX-2 cells induced by TGF-β were dramatically decreased after indirect coculture with ASCs and M1 macrophages. These results suggest that a comprehensive inflammatory microenvironment may play an important role in determining the therapeutic properties of ASCs by increasing TSG-6 expression through STAT1/3 activation. | - |
