dc.description.abstract[영문] [한글] 구강내에서 분리하여 동정된 Candida albicans 36주의 ketoconazole에 대한 균 최저 발 육억제 농도를 조사함과 동시에 ketoconazole 농도를 증가시켜 균 최저 발육억제 농도를 단계적으로 상승 시킨 내성 균주의 미세구조를 전자현미경으로 관찰 하였던 바 다음과 같 은 결론을 얻었다. 1. 실험균주 36주중 20주(55.6%)는 0.39 μg/㎖에서, 7주(19.4%)는 0.19μg/㎖에서 균 최저 발육억제 현상을 보였으며, 50.0μg/㎖에서도 3주(8.3%)가 균 최저 발육억제현상을 나타냈다. 2. 내성균주의 전자현미경적 미세구조 변화 (1) 형태적인 변화로 말발굽형, 방추형, 장방형 및 기타 비정상적 형태로 관찰되었다. (2) 세포벽의 불규칙한 비후현상과 파괴현상이 관찰되었다. (3) 세포벽내에 dense vesicular structure가 축적됨이 관찰되었다. (4) 세포막의 세포벽과의 이완현상이 관찰되었다. (5) 세포막의 파괴 및 용해현상이 관찰되었다. (6) 세포질내에 많은 지방적이 증가됨이 관찰되었다. (7) 불완전한 세포분열이 관찰되었다. THE ULTRASTRUCTURE OF CANDIDA ALBICANS AFTER IN VITRO EXPOSURE TO THE KETOCONAZOLE Soo Young Rho, D.D.S. Deparment of Dental Science, Graduate School, Yonsei Univ. (Directed by Prof. Eui Wung Lee, D.D.5., M.S.D., Ph.D.) Kotoconazole is a new vystemic antifungal compound which is active by the oral route against superficial find systemic infections with Candida albicans. In this study, the author investigated the minimal inhibitory concentration of ketoconazole on C. aibicans isolated from oral cavity and observed electron microscopically the ultrastructual changes of C. albicans which were acquired resistance against ketoconazole. The results were summarized as follows: 1. The minimal inhibitory concentrations of ketoconazole on C. albicans were 0.39μg/ml in 20 strains (55.6%) and 0.19μg/ml in 7 strains (19.4%), but even 50.0μg/ml in 3 strains (8.3%) among 36 strains of Candida albicans. 2. The ultrastructual changes of acquired resistant C. albicans were observed as follows: (1) The shapes of cells showed horse-shoe shape, cone shape, rectangular shape and other abnormal shapes. (2) The cell wall became irregularly thickened and disruptured. (3) The dense veiscular structure accumulated in the cell wall. (4) The cell membrane was relaxed from the cell wall. (5) The cell membrane was disruptured and lyzed. (6) Many lipid droplets were increased in the cytoplasm. (7) Incomplete cell division was observed.-
