dc.description.abstract[한글] 평활근 세포의 수축력의 크기를 결정하는 주요 인자의 하나인 세포 내 Ca2+ 이온의 농도는 세포막을 통한 Ca2+ 이온의 유출량과 유입량의 차이, 그리고 세포 내 Ca2+ 저장소로부터의 Ca2+ 유리량과 재흡수량의 차이에 의해 결정된다. 특히 평활근 세포막의 막전압 의존성 Ca2+ 통로는 Ca2+ 이온이 세포 내로 유입되는 중요한 통로로 작용하고 있어서, 어떤 인자에 의해 이의 활성이 변화되는 경우 평활근 세포 내 Ca2+ 이온 농도의 변동과 수축력의 크기 변화를 초래하게 됨은 잘 알려져 있다. 실험적으로 고콜레스테롤증을 유발한 혈관 평활근 세포의 경우 Ca2+ 전류의 활성이 대조군에 비해 증가되고, 실험적으로 산화 LDL을 투여하였을 때 평활근 세포 내 Ca2+ 농도가 증가한다는 보고 등은 죽상동맥 경화증이 유발된 혈관의 평활근 세포막의 Ca2+ 통로의 활성이 증가되고, 이를 통해 비정상적 혈관 반응이 유발되었을 가능성을 시사하고 있으나 이의 자세한 기전에 대해선 아직 명확치 않다. 따라서 본 실험에서는 whole patch clamp 방법을 사용하여 죽상경화증 혈관에서 그 농도가 크게 증가한다고 알려진 lysophosphatidylcholine (LPC)이 평활근 세포의 막전압 의존성 Ca2+ 전류의 활성에 미치는 효과와 그 작용 기전을 규명함으로써 죽상경화증 혈관에서 관찰되는 비정상적인 혈관 수축 현상이 나타나는 기전을 규명하고자 하였다. 실험을 통해 얻은 결과는 다음과 같다. 1. 평활근 절편의 긴장도 및 세포 내 Ca2+ 농도는 LPC에 의해 가역적으로 증가하였다. 2. 평활근 세포의 Ca2+ 전류의 활성은 LPC 투여 시 가역적으로 증가하였으며, 이같은 효과는 모든 막전압 값에서 동일하게 관찰되었다. 3. LPC 투여 시 Ca2+ 전류의 막전압 의존성 활성화 곡선이 오른쪽으로 일부 이동하였으나 막전압 의존성 비활성화 곡선에는 변화가 없었다. 4. LPC에 의한 Ca2+ 전류 증가 효과는 protein kinase A (PKA) 촉진제인 forskolin 전처치에 의해서는 영향을 받지 않았으나 protein kinase C (PKC) 촉진제인 phorbol myristic acid (PMA)를 전처치하는 경우 소실되었다. 5. PKC inhibitor인 staurosporine이나 chelerythrine을 전처치하는 경우 LPC에 의한 Ca2+ 전류 증가 효과가 소실되었다. 이상의 실험 결과로 볼 때 LPC에 의한 Ca2+ 전류의 증가 효과는 세포 내 PKC 활성 변화를 매개로 하여 이루어지며, 이에 의한 평활근 세포 내 Ca2+ 농도 증가 효과는 죽상경화증 혈관에서 관찰되는 혈관운동 기능이상의 발생에 중요한 역할을 한다고 생각된다. [영문] Impairment of relaxing response and augmentation of contractile response to vasoactive substances have been reported in atherosclerotic arteries. These alterations in vascular reactivity are considered as an underlying mechanism for the development of acute vasospasm in atherosclerotic artery. Recently, it has been reported that lysophophatidylcholine (LPC), an oxidative metabolite of low density lipoprotein causes this functional abnormality. However, the precise mechanism of LPC induced change of vascular reactivity is still uncertain. In this study, to elucidate the underlying mechanisms of abnormal vascular reactivity in atherosclerotic artery, we directly examined the effect of LPC on whole cell Ca2+ current using patch clamping technique. Application of LPC (1μM) induced a significant increase in ICa (142±9%, p<0.01), which was readily reversed by washout of LPC. Steady state voltage dependency of activation or inactivation properties of ICa was not significantly changed by LPC. The PKC inhibitor, staurosporine (10 nM) or chelerythrine (3μM) inhibited ICa by 54±6% (p<0.01), 47±6% (p<0.01), respectively. After inhibition of ICa by PKC inhibitors, application of LPC had no significant change in the amplitude of ICa. The PKC activator, phorbol myristic acid (PMA, 100 nM) stimulated ICa by 124±3% (p<0.01). After stimulation of ICa by PMA, application of LPC had no significant change in the amplitude of ICa. These findings suggest that LPC increase ICa by a pathway that involves endogenous PKC in vascular smooth muscle cells, and LPC-induced increase of ICa might be, at least in part, responsible for development of abnormal vascular reactivity in atherosclerosis.-
