구강암종에서 은행잎 추출물 (EGb 761) 구성 성분에 의한 항암작용

dc.description.abstract[한글] EGb 761은 항산화기능, 항산화효소 증강기능, cyclooxygenase-2 및 inducible nitric oxide synthase 억제, 신생혈관 억제능 등을 보이며 대장암과 전립선암 등에서 항암작용이 있다고 한다. 최근 연구에 의하면 구강암종 세포주에서도 세포고사 작용을 보인다고 한다. 그러나 EGb 761의 구성 성분 중 어느 성분이 구강암종 세포주의 세포고사를 야기하는지에 대해서는 알려진 바가 없다. 이에 첫째, 구강암종 세포주에서 EGb 761의 어떤 구성 성분이 세포증식을 억제하는지 알아보고 둘째, 구강암종 세포의 성장억제가 세포고사에 의한 것인가를 확인하며 셋째, 구성 성분에 의한 세포고사 유도가 EGb 761과 마찬가지로 caspase-3 의존성인가를 보고자 하였다. EGb 761의 성분으로 ginkgolide A, ginkgolide B, bilobalide, kaempferol, quercetin 등을 투여한 결과 kaempferol과 quercetin은 40 μM부터 세포증식을 유의하게 억제하였고 세포증식 억제는 유세포 측정분석 상 세포고사에 의함을 알 수 있었다. SCC-1483, SCC-25와 SCC-QLL1 등의 모든 구강암종 세포주에서 세포고사를 유도하였으며, PARP 분할도 유도하여 kaempferol과 quercetin 성분에 의해 구강암종 세포주에서 세포고사가 유도됨을 확인하였다. 또한 각각을 20 μM씩 혼합하여 처치하는 경우에 가장 강력하게 세포고사를 유도하였으며 EGb 761과 비교 시 10배 이하의 용량에서 세포고사 유도가 가능하였다. 그러나 세포에 따라 세포고사의 정도는 상이하였다. 또한, kaempferol과 quercetin에 의한 세포고사 유도는 caspase-3 의존성임을 알 수 있었다. 결론적으로 향후 EGb 761의 임상 적용에서 kaempferol과 quercertin 등의 구성 성분 투여가 효과적이며 향후 세포고사 유도 기전에 대한 추가적인 연구가 필요할 것으로 생각한다. [영문] EGb 761, extracted from Ginkgo biloba leaves, has been proven to suppress colorectal and prostate cancer. Previous studies have suggested that various mechanisms are involved in the chemopreventive effects of EGb761 such as antioxidant effects, inhibition of COX-2, and inhibitory effects on angiogenesis. Furthermore, the antiproliferative effects of EGb761 were recently demonstrated in oral cavity cancer cell lines. Since EGb761 is a composition of various components, it is essential to identify which component is responsible for its anticancer effects in oral cavity cancers. In the present study, we investigated which component of EGb 761 has antiproliferative effects on oral cavity cancer cell lines and whether each component induces cellular apoptosis. Additionally, we investigated which signal pathways are involved in the apoptotic effect of each EGb 761 component. Oral cavity cancer cells (SCC-1483) were treated with different components of EGb 761 including ginkgolides A and B, bilobalide, kaempferol, and quercetin at various concentrations. The results of cell proliferation assay showed that kaempferol and quercetin significantly inhibited cellular proliferation at a concentration of 40 μM. FACS analysis showed that the antiproliferative effects of each component were due to increased apoptosis. Kaempferol and quercetin induced apoptosis in all oral cancer cell lines (SCC-1483, SCC-25 and SCC-QLL1) and showed cleavage of PARP. Cotreatment of 20 μM of kaempferol and 20 μM of quercetin was the most potent in inducing apoptosis than any other treatment: the concentrations of the 2 components were less than one-tenth of that known to be effective for EGb 761. Although the antiproliferative effects of kaempferol and quercetin cotreatment varied among different oral cavity cell lines, apoptosis was shown to be caspase-3 dependent. In conclusion, our results suggest that the EGb 761 components kaempferol and quercetin effectively induce caspase-3-dependent apoptosis in oral cavity cancer cells, and further studies for componential applications are needed.-
