Abstract
[한글] 본 연구에서는 구강내 biofilm을 형성하는 몇 가지 미생물에 대한 항균력을 평가하기 위해 Curcuma xanthorrhiza 추출물인 Xanthorrhizol과 불소 또는 자일리톨을 1:1과 1:2의 비율로 혼합하여 각 항균물질의 minimum inhibitory concentration (MIC)과 속효성에 대해 알아본 결과 다음과 같은 결과를 얻었다.1. Xanthorrhizol, stannous fluoride, sodium fuoride의 MIC 값은 각각 S. mutans에 대해 5ppm, 2500ppm, 5000ppm, A. viscosus에 대해 5ppm, 1000ppm, 1000ppm, F. nucleatum에 대해 5ppm, 1000ppm, 5000ppm으로 나타났으며 자일리톨은 실험에 이용된 농도에서는 MIC 값이 관찰되지 않았다.2. Xanthorrhizol과 stannous fluoride, sodium fluoride, 또는 자일리톨이 1:1로 섞여있는 경우, MIC 값은 각각 S. mutans에 대해 1000ppm, 5000ppm, 10ppm, A. viscosus에 대해 500ppm, 250ppm, 5ppm, F. nucleatum에 대해 2500ppm, 1000ppm, 50ppm으로 나타났다.3. Xanthorrhizol과 stannous fluoride, sodium fluoride, 또는 자일리톨이 1:2로 섞여있는 경우, MIC 값은 각각 S. mutans에 대해 1000ppm, 5000ppm, 100ppm, A. viscosus에 대해 500ppm, 500ppm, 50ppm, F. nucleatum에 대해 5000ppm, 2500ppm, 100ppm으로 나타났다.4. 단일 항균물질의 속효성을 알아본 결과, Xanthorrhizol, stannous fluoride의 경우, 1분 안에 균을 사멸시킬 수 있는 농도는 S. mutans에 대해 25ppm, 2500ppm, A. viscosus에 대해 50ppm, 2500ppm, F. nucleatum에 대해 100ppm, 5000ppm으로 나타났으나 sodium fluoride, 자일리톨은 실험에 이용된 농도에서는 1분 안에 균을 사멸시키지 못했다.5. 혼합 항균물질의 속효성을 알아본 결과, Xanthorrhizol과 stannous fluoride 또는 자일리톨이 1:1로 섞여있는 경우, 1분 안에 균을 사멸시킬 수 있는 농도는 각각 S. mutans에 대해 2500ppm, 100ppm, A. viscosus에 대해 2500ppm, 100ppm, F. nucleatum에 대해 2500ppm, 250ppm으로 나타났으며, Xanthorrhizol과 sodium fluoride가 1:1로 섞여있는 경우에는 2500ppm의 농도에서 A. viscosus 균주를 거의 모두 사멸시켰지만 S. mutans나 F. nucleatum 균주에 대해서는 1분 안에 균을 사멸시키지 못했다. Xanthorrhizol과 stannous fluoride, sodium fluoride, 또는 자일리톨이 1:2로 섞여있는 경우, 1분 안에 균을 사멸시킬 수 있는 농도는 각각 S. mutans에 대해 5000ppm, 5000ppm, 500ppm, A. viscosus에 대해 2500ppm, 1000ppm, 250ppm, F. nucleatum에 대해 5000ppm, 5000ppm, 500ppm으로 나타났다.불소나 자일리톨의 경우 Xanthorrhizol을 섞었을 때 불소나 자일리톨을 단독으로 사용할 때 보다 항균력이 증가하였다. 하지만 Xanthorrhizol은 그 자체로서 항균력이 매우 뛰어났지만 불소 또는 자일리톨과 섞을 경우 Xanthorrhizol 자체의 항균력은 떨어졌다. 따라서 치약의 경우처럼 불소를 배제할 수 없는 경우 Xanthorrhizol을 첨가하면 불소의 치아 재광화 효과와 함께 항균력 효과까지도 기대해 볼 수 있을 것이다.
[영문]The Purpose of this study was to evaluate the effect of Xanthorrhizol containing fluoride or Xylitol on bacteria related to biofilm formation. Minimum inhibitory concentrations (MIC) and the fast effectiveness antibacterial agents were examined for the evaluation. The results were as follows;.1. MICs of Xanthorrhizol, stannous fluoride, and sodium fluoride were 5ppm, 2500ppm, 5000ppm on S. mutans, were 5ppm, 1000ppm, 1000ppm on A. viscosus, and were 5ppm, 1000ppm, 5000ppm on F. nucleatum respectively. However, the MIC of Xylitol is still under observation.2. MICs of Xanthorrhizol mixed with stannous fluoride, sodium fluoride or Xylitol in the ratio of 1:1 were 1000ppm, 5000ppm, 10ppm on S. mutans, were 500ppm, 250ppm, 5ppm on A. viscosus, were 2500ppm, 1000ppm, 50ppm on F. nucleatum respectively.3. MICs of Xanthorrhizol mixed with stannous fluoride, sodium fluoride or Xylitol in the ratio of 1:2 were 1000ppm, 5000ppm, 100ppm on S. mutans, were 500ppm, 500ppm, 50ppm on A. viscosus, were 5000ppm, 2500ppm, 100ppm on F. nucleatum respectively.4. As a result of the fast effectiveness of only single antibacterial agents, Xanthorrhizol or stannous fluoride was able to completely inhibit growth of bacteria when they were exposed to these antibacterial agents for 1 minute in concentrations of 25ppm, 2500ppm on S. mutans, 50ppm, 2500ppm on A. viscosus, 100ppm, 5000ppm on F. nucleatum respectively. However, Xylitol couldn''t inhibit growth of bacteria when they were exposed to these antibacterial agents for 1 minute.5. According to the fast effectiveness of mixed antibacterial agents, Xanthorrhizol mixed with stannous fluoride or Xylitol in the ratio of 1:1 was able to completely inhibit growth of bacteria when they were exposed to these antibacterial agents for 1 minute in concentrations of 2500ppm, 100ppm on S. mutans, 2500ppm, 100ppm on A. viscosus, 2500ppm, 250ppm on F. nucleatum respectively. However, Xanthorrhizol mixed with sodium fluoride in the ratio of 1:1 inhibited only growth of A. viscosus at 2500ppm but couldn''t inhibit growth of S. mutans and F. nucleatum when they were exposed to these antibacterial agents for 1 minute. Xanthorrhizol mixed with stannous fluoride, sodium fluoride or Xylitol in the ratio of 1:2 was able to completely inhibit growth of bacteria when they were exposed to these antibacterial agents for 1 minute in concentrations of 5000ppm, 5000ppm, 500ppm on S. mutans, 2500ppm, 1000ppm, 250ppm on A. viscosus, 5000ppm, 5000ppm, 500ppm on F. nucleatum respectively.As the result of this study, Xanthorrhizol containing fluoride or Xylitol had more antibacterial effect than only fluoride or Xylitol. Therefore, it is expected that dentifrice containing Xanthorrhizol and fluoride will have more effect on tooth remineralization with antibacterial effect.
