dc.description.abstract | [한글] TRAIL에 저항성을 가지는 암세포에 사이클로헥시마이드를 TRAIL과 병용 처리 하였을 경우 감작 현상이 일어 난다. 하지만 아직 그 감작 기전은 알려 지지 않은 실정이다. 본 실험에서는 TRAIL에 저항성을 가지는 암세포에서 높은 프로테인 카이네이즈 CK2의 활성도 및 이로 인한 procaspase-2의 serine 140부위가 인산화됨을 확인 하였고, 이런 높은 프로테인 카이네이즈 CK2의 활성도는 사이클로헥시마이드의 처리로 인해 억제 될 수 있었으며, 이를 통해 procaspase-2의 인산화 억제 및 이분체 형성을 통한 활성화를 확인 하였다. Procaspase-2의 활성화가 억제되면 death receptor를 매개로한 세포 사멸이 일어날 수 없었다. 따라서 DRB와 같은 프로테인 카이네이즈 CK2 억제제를 처리해 주면 procaspase-2의 인산화가 억제되었고 이분체 형성 및 활성화가 일어남으로 인해 TRAIL에 저항성을 갖는 암세포에 TRAIL과 병용처리 할 경우 효과적인 암세포 사멸을 확인할 수 있었다. 뿐만 아니라 TRAIL에 저항성을 갖는 악성 신경 교종을 이식한 누드 마우스에서도 프로테인 카이네이즈 CK2 억제제와 TRAIL의 병용처리는 간세포 독성없이 효과적으로 암세포의 성장 억제 및 사멸을 유도하였다. 이 결과를 미루어 볼 때 프로테인 카이네이즈 CK2와 procaspase-2의 상호 작용을 특이적으로 억제 할 수 있는 작은 분자를 찾는다면 그것은 사이클로헥시마이드의 TRAIL에 대한 감작효과를 나타낼 수 있을 뿐만 아니라 TRAIL을 이용한 암치료에 효과적으로 사용될 것이다. [영문]Cycloheximide (CHX) is widely used as it potently sensitizes TRAIL-resistant cancer cells to TRAIL. However, the mechanism(s) involved in the sensitization is unclear. Here we show that TRAIL-resistant cancer cells have high protein kinase (PK) CK2 activity and that PKCK2 directly phosphorylates procaspase-2 on its serine-140 residue, thereby inhibiting the dimerization and activation of procaspase-2. CHX sensitizes TRAIL-resistant cancer cells to TRAIL by downregulating PKCK2 activity. Without the activation of procaspase-2, death-receptor mediated apoptosis cannot occur in cancer cells that originate from different tissue types. DRB, a PKCK2 inhibitor, sensitizes TRAIL-resistant glioma and induces complete tumor regression in mice without concomitant hepatocyte apoptosis. These results suggest that a small molecule(s) that can specifically prevent the interaction between PKCK2 and procaspase-2 may mimic the potent sensitizing effect of CHX and thus could broaden the therapeutic applications of TRAIL.
