헌팅턴 병은 헌팅틴 단백의 아미노말단에 있는 글루타민 트랙이 비정상적으로 증폭되어 발병하며 신경세포의 핵 내에 주로 아미노말단 헌팅틴 단백질 조각으로 이루어진 봉입체를 형성하는데 이 과정이 발병에 가장 중요한 요소라고 생각되어 왔다. 따라서 헌팅틴 단백이 어떠한 과정으로 분해가 되어 작은 아미노말단 조각을 형성하는지를 이해하는 것은 향후 치료제 개발에 도움을 줄것으로 생각된다. 본 연구에서는 정상과 돌연변이 헌팅틴 단백의 아미노말단에서 100 내지 200 번째 아미노산 부위에서 헌팅틴 단백은 절단되는 것을 관찰하였으며, 이 헌팅틴 단백의 아미노말단 조각은 글루타민 트랙의 수가 증가할수록 불용성임을 알수 있었다. 프로테아좀의 기능이 억제되거나 카뎁신-디가 결핍된 경우, 이 아미노말단 헌팅틴 단백조각의 발현은 증가되는것으로 미루어, 상기의 단백분해효소가 정상적으로 헌팅틴단백의 단백분해에 작용하는 것을 알수 있었다. 크기가 가장 작은, 돌연변이를 가진 헌팅틴 단백조각은 세정제에 잘 용해되지 않았고 카뎁신-디에 의하여 분해되지 않았다. 헌팅턴 환자의 뇌에서 헌팅턴 단백의 작은 아미노말단 조각이 상호 응집되는 것을 이러한 현상으로 일부분 설명할수 있었다. 시험관 내에서 헌팅틴 단백의 작은 아미노말단 조각의 형성은 펩스타틴-에이 처치시 억제 되었고 산성 버퍼 내에서 촉진되었다. 한편 현재까지 알려진 아스파틸 단백분해효소들은 헌팅턴 단백의 절단에 관여하지 않았다. 헌팅턴 생쥐와 헌팅턴병 환자의 뇌조직 추출물에서는 비슷한 크기의 헌팅틴 단백 아미노말단 조각을 포함하고 있었다. 결론적으로 헌팅턴병 환자의 뇌에서 응집되기 쉬운 헌팅틴 단백조각이 아스파틸 단백분해 효소의 성격을 가지는, 아직 알려지지 않은 단백분해 효소에 의하여 형성되는 것으로 사료된다.



[영문]N-terminal huntingtin fragments with a polyglutamine expansion form intraneuronal inclusions in the brain of Huntington’s disease patients (HD) and contribute to neuronal dysfunction and cell death in HD mice. Knowledge of the proteolytic pathways involved in forming aggregates could lead to strategies for HD therapy. A series of five wild-type and mutant huntingtin fragments released by proteolysis about 100-200 residues from the N-terminus was identified and these became insoluble with polyglutamine expansion. Proteasome inhibition or a deficiency in cathepsin D raised the levels of the huntingtin fragments, indicating that these proteases are normally involved in their degradation. The smallest mutant huntingtin fragment was the most detergent insoluble and resisted degradation by cathepsin D. These features may promote aggregation in HD brain. Unexpectedly, fragments of wild-type, as well as mutant huntingtin, formed inclusions in vitro when the proteasome was impaired. Formation of the fragments was blocked by the aspartyl protease inhibitor pepstatin A. The aspartyl proteases, cathepsin D, BACE, and gamma secretase were not involved in cleavage: the fragments still formed in cells deficient in cathepsin D and inhibitors of BACE or gamma secretase, or expression of wild-type or dominant negative presenilin had no effect on levels of the fragments. Extracts from homozygous HD mouse brain and HD patient brain had fragments of similar size. I conclude that aggregate-prone huntingtin fragments in the HD brain arise from cleavage by a novel protease with aspartyl protease activity.
