p16INK4a 유전자는 종양 억제 유전자로 9번 염색체 단완 (short arm) 부위에 위치한다. 정상적으로 p16INK4a 유전자는 cyclin D-dependent kinase인 CDK4와 CDK6를 억제하는 p16 단백질을 만들고 p16 단백질은 CDK4가 cyclin D1과 결합하는 것을 방해하여 retinoblastoma tumor suppressor protein (Rb protein)이 인산화 되는 것을 방지함으로써 세포 분열과정 중에 G1 상태에서 더 이상의 진행을 막는 역할을 한다. 이러한 p16INK4a 종양 억제 유전자의 불활성화는 악성 신경교종에서 가장 빈번하게 발견되는 유전자 이상이다. 따라서 p16INK4a 의 소실은 신경 교종의 발생 및 분화와 밀접한 연관이 있을 것이다.

본 연구는 p16INK4a 유전자가 소실되어 있는 U343 교종세포주에 full length p16 cDNA를 도입함으로써 세포주의 성장에 어떤 영향을 미치는지 분석하였다. p16INK4a 유전자 발현 이외의 모든 조건을 동일하게 하기 위해 tetracycline-repressor gene expression system을 사용하여 p16INK4a이 안정적으로 감염된 clone을 만들 수 있었다.

Hemocytometer를 이용한 세포 증식 분석결과 p16INK4a 발현을 유도한 U343-p16INK4a 세포주의 증식이 현저하게 억제된 반면, 유전자 발현을 유도하지 않는 U343-p16INK4a 세포주의 증식은 U343 모세포주의 증식과 같은 양상을 보였다 (p<0.001). 세포 주기 분석결과 p16INK4a 발현을 유도한 경우 85%를 상회하는 U343-p16INK4a 세포들이 G1 phase에 정지되어 있었으나 유전자 발현을 유도하지 않은 경우는 U343-p16INK4a 세포주와 U343 모세포주 간에 차이가 없었다 (p<0.01). 면역형광염색을 이용한 세포형태의 관찰에서는 p16INK4a 발현을 유도했을 경우 입방형의 세포돌기가 없는 모세포주의 형태가 점차 양극형의 모양과 glial fibrillary acidic protein이 잘 발달된 세포돌기를 지니는 형태로 분화되는 것을 확인 할 수 있었다.

본 연구의 결과를 토대로 p16INK4a 유전자를 가지고 있지 않는 U343 교종세포주에 인위적으로 p16INK4a 활성을 복원하여 주면 세포주기를 G1 phase에 정지시킴으로써 성장을 억제하고 교종 세포의 형태적인 분화를 정상적인 방향으로 유도함으로써 침윤성을 저하시킬 수 있을 것이다. 결과적으로 악성교종의 치료에 있어서 유전자치료 전략을 수립하는데 p16INK4a이 유용한 표적 유전자로 사용될 수 있을 것이다.





[영문]The tumor suppressor gene p16INK4a is a G1-specific cell cycle inhibitor which negatively regulates CDK4 and CDK6 by binding in competition with D-type cyclins. p16INK4a maps to human chromosome 9p21, a region frequently mutated or deleted in human cancer cell lines. One of the most frequent abnormalities in the progression of gliomas is the inactivation of tumor suppressor gene p16INK4a, suggesting that loss of p16INK4a is associated with acquisition of malignant characteristics.

We have analysed the significance of the loss of this gene in gliomas by introducing the cDNA for p16INK4a into U343 human glioma cell line which lacks p16INK4a, but expresses glial fibrillary acidic protein. With use of tetracyclin-repressor gene expression system, we obtained stably transfected clone that expressed p16INK4a upon induction.

Cell proliferation assay using hemocytometer revealed that the proliferation of U343-p16INK4a glioma cells was significantly suppressed after the induction of p16INK4a, while that of U343-p16INK4a without induction was the same to that of parent U343 glioma cells (p<0.001). Flow cytometric analysis showed that p16INK4a expression caused over 85% of the U343-p16INK4a glioma cells to arrest in G1 phase (p<0.01) and there was no difference in G1 fraction between U343 parent cell line and U343-p16INK4a cell line when p16INK4a was not induced. By using confocal microscope, we have observed that the expression of p16INK4a led tightly packed cuboidal cells without process to change into the bipolar cells with the cytoplasmic processes with well-organized glial fibrillary acidic protein.

Our results demonstrated that the restoring p16INK4a activity into p16INK4a-null U343 glioma cells significantly suppressed tumor cell proliferation by arresting cell cycle in G1 phase and potentially inhibit tumor cell invasion, thus suggesting that p16INK4a could be a useful target of gene therapy strategy for the treatment of human gliomas.
