dc.description.abstract[한글] 항바이러스 제재로 사용되고 있는 isoprinosine의 면역 항진제로서의 역할을 알아보기 위한 일환으로 isoprinosine 투여가 마우스 복강거식세포의 활성화에 미치는 영향을 관찰한바 다음과 같은 결과를 얻었다. (1) Isoprinosine을 마우스에 매일 마우스 마리당 100mg씩 5일 또는 10일간 경구 투여하더라도 복강세포 총수 및 복강거식세포 비율의 변화는 관찰할 수 없었다. 도한 시험관 내에서 isoprinosine 10ug/ml, 또는 100ug/ml 투여시에도, 일단 착생된 복강거식세포의 착생능에도 변화가 없었다. (2) 상기 용량으로 경구 또는 시험관 내로 isoprinosine을 투여한 마우스 복강 거식세포는, Staphylococcus epidermidis를 탐식하는 능력이 다소 증가하였다. (3) Isoprinosine은 종양세포 NS-1에 대하여 직접적인 세포독성은 없었으며, isoprinosine을 생체 내 또는 시험관 내에서 투여한 마우스 복강 거식세포는, 정상대조군에 비하여 종양세포 NS-1에 대한 세포 독성능력에 차이가 없었다. (4) 생체 내 또는 시험관 내 isoprinosine 투여는 거식세포를 선택적으로 손상시키는 carrageenan 처리에 의한 마우스 복강거식세포의 세포독성 능력의 저하를 정상으로 유지시켰다. 즉, 시험관 내에서 carrageenan 0.125mg/ml을 단독 처리한 복강거식세포는 정상대조군의 복강거식세포 보다 세포독성 능력이 저하되는데 반하여, isoprinosine을 투여한 군의 복강거식세포는 carrageenan을 처리하더라도, 복강 거식세포의 NS-1종양세포에 대한 세포독성 능력은 정상으로 유지되었다. Effect of Isoprinosine on Mouse Peritoneal Macrophages In Hong Choi Department of Medical Science, The Graduate School, Yonsei University (Directed by Professor Joo Deuk KIM, M.D.) Immunomodulatory effect of "Isoprinosine", an antiviral agents, on peritoneal macrophages were evaluated by observing their phagocytic activity and tumoricial activities in vitro. Isoprinosine was originally developed as an antiviral agent and the mechanism of antiviral activities is known to be an indirect one by potentiating host defence system rather than direct effect upon invading viruses. Their positive roles in cell mediated immune responces in infected animals were reported by numerous investigators. Thus, the possible roles of the agent and bacteria were experimentally tested in vitro. The followings are summary of the findings. 1. Total number of peritoneal exudate cell(PEC) and macrophages of the study group animal were not changed by isoprinosine (100mg/day/mouse P.O. for 5 days). Isoprinosine appeared not to affect the adhesiveness of macrophages and viability of the adherent peritoneal macrophages treated with 10ug/ml or 100ug/ml. 2. Phagocytic ability of macrophage upon Staphylococcus epider-midis was slightly enhanced when the cells were treated with isoprinosine as above dose. 3. Direct effect of the drug itself upon tumor cell line, NS-1, and macrophage mediated cytotoxicity to NS-1 was not observed when the cells were treated with isoprinosine alone in vivo or in vitro. 4. However, isoprinosine were found to be an efficient agent in restoring selectively suppressed macrophages function by carrageenan. The isoprinosine pretreated macrophages exerted justifiable degree of tumoricidal activity regardless carrageenan post-treatment, which is quite compatible with that of normal.
