CdCl₂및 Pb(Ac)₂화합물 중독에 의한 혈액학적 소견과 면양 적혈구에 대한 항체생산 세포수에 미치는 영향

dc.description.abstract[한글] 환경오염 물질로써 카드뮴, 납화합물은 급성 또는 만성적으로 건강 장해를 일으킨다. 이들은 각종 생체의 대사, 효소의 활성, 단백질 합성 등의 기능을 억제함으로써 독작용을 일으킨다. 이들의 생체에 대한 기능장해는 또한 면역능력에도 영향을 미칠 것이 예상되어 본 연구에 착수하였다. 흰쥐를 실험동물로 사용하여 무게kg당 CdCl^^2 , 8.8mg/Saline 10ml 또는 Pb(Ac)^^2 , 15mg/Saline 10ml를 1주와 3주째에 각기 2회 복강내 주사하였고 그후 2일 간격으로 3회 면양적혈구로 면역시킨 후 4주째에 흰쥐에서 적출한 비장세포의 항체생산 능력을 Jerne의 방법에 따라 면양적혈구 용혈반응의 정도로 측정하였다. 또한 각 실험군의 1주와 4주에 대한 혈액학적 변화를 측정하였다. 이상의 실험결과를 요약하면 다음과 같다. 1. 백혈구수의 변화 : CdCl^^2 , Pb(Ac)^^2 를 투여한 실험군과 이를 투여하여 중독시킨 후 면양적혈구로 면역시킨 실험군에서 대체로 대조군과 실험군 사이에 있어 혈중 백혈구 수에는 차이가 없었다. 2. Hemoglobin의 변화 : CdCl^^2 , Pb(Ac)^^2 를 투여하여 1주 경과후 실험한 군에서 대조군, 15.5±0.53mg/dl 에 비해 각기 12.2±0.81 mg/dl 로 감소하였고, 이 독성 물질들을 3주때 재 투여하고 면양적혈구를 4주때 투여한 군에서도 역시 10.7±0.40mg/dl 및 10.8±0.62mg/dl 로 감소하 였다. 3. Hematocrit의 변화 : CdCl^^2 , Pb(Ac)^^2 를 투여하여 1주 경과후 실험한 군에서 대조군, 58.9±5.03%에 비해 각기 44.2±1.87% 및 45.5±4.92%로 감소하였고 이 독성물질들을 3주때 재 투여하고 면양적혈구로 면역시켜 4주때 실험한 군에서도 역시 39.1±1.03% 및 42.4%±2.54%로 감소하였다. 4. 혈청단백중 총단백, Albumin, α-Globulin, β-Globulin, γ-Globulin, A/G (Albumin/Globulin)의 변화: CdCl^^2 , Pb(Ac)^^2 를 투여하여 1주, 2주 경과후 실험한 결과 실험군과 대조군 사이에는 유의한 차이가 없었다. 5. 용혈플라크수의 변화 : CdCl^^2 , Pb(Ac)^^2 를 투여하고 3주때 면양적혈구로 면역시켜 4주때 실험한 군에서대조군의 용혈플라크수, 144±23 plaque수 / 0.5 ml of rat spleen cell로 적었다. 이상의 실험결과는 흰쥐에 있어, CdCl^^2 및 Pb(Ac)^^2 에 의한 중독으로 hemoglobin의 감소와 면역능력의 감퇴현상을 관찰할 수 있었다. [영문] Among the environmental pollutants, cadmium and lead compounds may impair human health. And these compounds may inhibit the biological metabolic function of indirectly. This study was undertaken to investigate the effects on the immune response by intoxication of cadmium chloride and lead acetate. Cadmium chloride (8.8mg/kg, saline 10ml) and lead acetate (15mg/kg, saline 10ml) were administered intraperitoneal injectic to rats. After 4 weeks, the immune respinse of rat spleen cells was measured by the Jerne's technique. The results were obtained as follows; 1. there was no change of leukocyte counts by the intoxication of cadmium cholride and lead acetate. 2. Cadmium chloride and lead acetate reduced hemoglobin contents for most intoxicated and immunized groups. 3. Hematocrits were decreased by the intoxication of cadmium chloride and lead acetate significantly. 4. It was determined that total protein, A/G (Albumin/Globulin), α-,β-andγ-globulins in rat serum were not changed. 5. Intoxication by cadmium chloride and lead acetate reduced the number of hemolytic plaque to the sheep RBC in rat spleen cells. Therefore, the immune response of rat spleen cells was suppressed by the intoxication of cadmium chloride and lead acetate.-
