자유생활 아메바의 병원성 감별을 위한 세포화학적 연구

dc.description.abstract자유생활 아메바의 병원성을 식별하기 위하여 형태학적 소견, 실험동물에의 감염능, 면역반응, 조직배양 세포에 대한 아메바의 세포 병변효과(cytopathic effect), 생화학적 방법으로 phytoagglutinin인 concanavalin A에 의한 응집반응등이 날리 이용되고 있다. 본 연구에서는 미세구조적 세포화학(ultrastructural cytochemistry) 방법을 도입하여 자유생활 아메바의 병원성과 미세구조와의 관계를 알아보고자 하였다. CGV 배지 또는 CGVS 배지 (Willaert 및 LeRay, 1973)를 사용하여 배양된 병원성 및 비병원성 자유생활 아메바를 대조군과 실험군의 2군으로 나눈 다음 실험군은 다시 concanavalin A 처리군, horseradish poroxidase, type Ⅱ (Sigma) 처리군 및 concanavalin A-horseradish peroxidase 처리군의 3군으로 나누어 이들 아메바의 미세구조를 전자현미경을 통하여 비교 관찰하여 병원성과의 관계를 관찰하였다. 그 결과를 요약하면 다음과 같다. 1. Concanavalin A로 처리하였을 때 병원성인 Acanthamoeba species인 YM-4와 Acanthamoeba lenticulata에서 세포막의 전자밀도가 약간 증가되어 있었으나 병원성인 Naegleria fowleri에서는 변화가 없었다. 2. Horseradish peroxidase로 처리하였을 때 병원성 및 비병원성 자유생활 아메바 모두에서 mitochondria와 Iysosome의 전자밀도가 현저히 증가되는 반면 세포막등 미세구조는 별 변화가 없었다. 3. Concanavalin A와 horseradish peroxidase로 동시에 처리하였을때 병원성 아메바인 Acanthamoeba lenticulata, Acanthamoeba sp., YM-4 및 Naegleria fowleri에서 세포막의 전자밀도가 현저히 증가된 반면, 비병원성 아메바인 Acanthamoeba royreba에서는 세포막의 전자밀도에 변화가 없었다. Mitochondria와 Iysosome의 전자밀도는 모든 아메바에서 peroxidase 처리군과 동일하였으며, 기타 미세구조는 대조군이나, concanavalin A 처리군 또는 peroxidase 처리군과 비교하여 뚜렷한 변화는 없었다. 이상과 같이 자유생활 아메바를 concanavalin A와 horseradish peroxidase로 처리하여 전자현미경을 통한 미세구조를 관찰하였던 바 병원성 유무에 따라 그 미세구조 특히 세포막에 현저한 차이가 있음을 알 수 있었다. Pathogenicity of free-living amoebae may differ by strain and by environmental conditions. Hitherto, a considerable research has been done by various workers to identify pathogenic strains from non-pathogenic ones by means of morphology, serology or by physico-chemical methods such as the concanavalin A-induced agglutination method(Culbertson etc, 1966 and others). In the present study, the concanavalin A-horseradish peroxidase method was employed in ultrastructural cytochemistry to visualize the ultrastructures of free-living amoebae with reference to virulence. The free-living amoebae investigated in the study were: non-pathogenic Acanthamoeha royreba, and pathogenic Acanthamoeba lenticulata, Acanthamoeba sp. (YM-4) and Naegleria fowleri. Free-living amoebae were cultured in the non-nutrient agar plate and CGV or CGVS medium. Comparative observation of the ultrastructures of free-living amoebae which were treated with concanavalin A, horseradish peroxidase, type Ⅱ (Sigma) or concanavalin A-horseradish peroxidase respectively, was undertaken through the electron microscope (Hitachi-H 500). The results are as fellows; 1. In the pathogenic Acanthamoeba lenticulata and Acanthamoeba sp. (YM-4) which were treated with concanavalin A, the electron density of the surface of the cell membrane was slightly increased, but there was no obvious difference compared to the electron density of the cell surface of non-pathogenic Acanthamoeba royreba and pathogenic Naegleria fowleri. 2. In all free-living amoebae which were treated with horseradish peroxidase, the electron-dense reaction product was found exclusively on the mitochondria and lysosome, but there was no difference with regard to the electron density of the surface of the cell membrane or other ultrastructures. 3. Ultrastructural demonstration of concanavalin A-horseradish peroxidase activity of the surface of the cell membrane was obvious only in pathogenic Acanthamoeba lenticulata, Acanthamoeba sp. (YM-4) and Naegleria fowleri, but not in the non-pathogenic Acanthamoeba royreba. The electron density of mitochondria and lysosomes was similar with the group treated with horseradish peroxidase. No obvious difference was observed with regard to electron density of other ultrastructures between the control and the treated group with concanavalin A or horseradish peroxidase respectively. The overall results suggest that concanavalin A-horseradish peroxidase activity is clearly demonstrated on the cell membrane of the pathogenic strains of free-living amoebae.-
