dc.description.abstract[한글] 위장홀몬은 위장관계 뿐 아니라 중추신경계에도 분포하며, 이들중 상당수가 중추신경계에 작용하여 위산분비를 조절하고, 이 작용에는 미주신경이 중요한 역할을 한다고 한다. Vasoactive intestinal polypeptide (VIP)는 28개의 아미노산으로 구성된 peptide로서, 위장관계 및 중추신경계에 다량 분포되어 있으며, 최근에는 신경전달물질로서의 역할이 제시되었다. VIP를 말초적으로 투여할 때 위산분비를 억제한다고 하나 종에 따라 차이가 있으며, 특히 이에 대한 VIP의 중추작용에 관해서는 보고가 적고, 그 실험결과 역시 일치하지 않는다. 이 실험에서는 VIP의 뇌실내 투여에 의한 위산분비 기능 변동과 그 기전을 추구하였으며, 아울러 결박 스트레스에 의한 위궤양 발생에 대한 VIP의 영향을 관찰하였다. 실험동물로는 200g 안팎의 흰쥐를 암수 구별없이 사용하였고 측뇌실내 삽관후 5∼7일간 사육한 다음 위관류 실험을 시행하였다. 실험 결과는 다음과 같다. 1. 뇌실내 VIP 0.1, 0.3㎍ 투여로 위산 및 pepsin분비는 용량의존적으로 항진되었으나, 3, 10㎍투여로는 오히려 위산분비의 증가가 미약하였다. 2. 뇌실내 VIP 투여로 인한 위산 및 pepsin분비 항진 효과는 양측 미주신경절단으로 완전히 소실되었다. 3. Atropine 및 pirenzepine 전처치로 뇌실내 VIP 투여로 인한 위산 및 pepsin 분비 항진 효과는 억압되었다. 4. 뇌실내 VIP 투여로 인한 위산 및 pepsin 분비 항진 효과는 hemicholinium이나 desipramine전처치로 소실되었고, metyrosine 전처치로는 위액분비항진효과가 지연되어 나타났다. 5. Cimetidine 전처치로 위산 및 pepsin분비가 전반적으로 억제되며, 뇌실내 VIP투여로 인한 위산 및 pepsin분비 항진 효과는 cimetidine으로 봉쇄되었다. 6. 스트레스 위궤양 발생은 뇌실내 VIP 투여로 증가되었다. 이상의 성적으로 보아 VIP는 중추신경계에 작용하여 위산 및 pepsin분비를 증가시키고 스트레스 위궤양을 악화시킴을 알 수 있었으며, 이 효과는 미주신경을 경유한 콜린성 섬유의 활성에 기인함을 알 수 있었다. [영문] Vasoactive intestinal polypeptide (VIP) is a peptide composed of 28 amino acids that was first isolated from the porcine duodenum. Although VIP was primarily considered to be a gut hormone, it has been shown to occur in central and peripheral neuron. At present, it is accepted that VIP acts as a neurotransmitter or a neuromodulator both in central and peripheral nervous system. Recently immunohistochemical studies have shown that the vagus nerve contains not only acetylcholine but also other neurotransmitters including VIP. These findings suggest that the central control of acid secretion is considerably complex and raised the possibility that multiple neurotransmitters may affect the secretory function of the gastric parietal cell. In this context, VIP might be involved in the central regulation of acid secretion, which is yet to be investigated. In present study, the effects of intracerebroventricular (icv) administration of VIP on the gastric secretion as well as its mechanism and stress ulceration were investigated in Sprague-Dawley rats of either sex weighing about 200g. The results are summarized as follows. 1. ICV injection of VIP increased secretion of gastric acid and pepsin in a dose-dependent manner upto 1㎍. However, higher doses (3∼10㎍) fail to stimulate further the gastric secretion. 2. The secretion of gastric acid and pepsin induced by icv VIP was abolished by bilateral vagotomy as well as by atropine or pirenzepine. 3. The effect of icv VIP on gastric secretion was prevented by hemicholinium or desipramine. Metyrosine delayed the stimulatory response of gastric function induced by icv VIP. 4. Cimetidine inhibited the basal and icv VIP-induced gastric secretion 5. Stress-induced ulceration in pylorus-ligated restraint rats was aggravated by icv injection of VIP. From the above results, it is demonstrated that intracerebroventricular VIP stimulates gastric acid and pepsin secretion, and aggravates stress-induced ulceration, and is suggested that these effects are mediated by cholinergic mechanism via central vagal activation.-
