사람 이자 [췌장(膵臟)] 효소단백의 HPLC분석

dc.description.abstract[영문] [한글] 이자 효소단백의 분비 양상에 대한 연구는 주로 전기영동법을 이용하여 수행되어 왔으 며, 단백질의 분자량과 등전점의 차이를 이용한 이차원 전기영동법의 개발은 이자 효소단 백의 분리와 정량에 많은 공헌을 하였다. 그러나 이방법은 과정이 복잡하고 많은 시간이 소요된다는 단점이 있다. 근래 크로마토그래피 기술이 발전함에 따라 high performance l iquid chromatography(HPLC) 방법으로 이자효소를 분리 정량하려는 노력이 진행되고 있다 . 초기에는 역상(reversed phaae) column을 이용한 방법이 시도 되었는데 이 방법으로는 단백 각 분획을 정확히 인지하기가 곤란하였다. 최근 흰쥐 이자액을 hydrophobic interac tion column을 이용하여 효소 활성을 유지한 단백을 분리, 인지한 보고가 발표되었다. 본 연구는 hydro-phobicinteraction column을 이용하여 사람 이자액을 분석하는 첫번째 시 도로서, 사람 이자액 내 여러 주요 효소 단백을 HPLC 방법으로 분리 정량하는데 있어 표 준을 설정하고, 사람의 이자외분비에 대한 연구 및 질병의 진단과 치료확인에 대한 응용 가능성을 추구하고자 하였다. 실험 재료로는 총 14명의 환자로부터 채취한 이자액을 사용하였으며, 이 중 10개 시료 는 수술 후 이자관에 삽입한 카데타로 부터, 4개 시료는 역행성 담취관 조영술을 시험하 면서 채취하였다. 이자액의 단백함량은 Lowry법을 이용하여 측정하였으며, 효소단백은 TS K phenyl 5PWhydrophobic interaction column을 이용하여 HPLC 방법으로 분리하였고, 분 리한 각 분획의 효소 활성은 화학적 방법으로 확인하였다. 실험 성적은 다음과 같다. 1. Ammonium sulfate가 함유된 완충액과 저농도의 ethanol이 함유된 완충액의 조성을 시간에 따라 변화시킨 이동상(mobile phase)을 사용하여 사람 이자액 속에서 총 18개 분 획을 분리할 수 있었다. 2. 각 분획의 효소활성을 측정한 결과 chymotrypsinogen, lipase, 및 trypainogen의 활 성은 각각 3개 분획에서, amylase, procarboxypeptidase A 및 B의 활성은 각 1개 분뵉에 서 확인하였으며 1개 분획에서는 trypginogen롸 chymotrypainogen의 2가지 효소 활성을 나타내었다. 3. 사람 이자 효소단백의 HPLC 크로마토그램은 이미 보고된 횐쥐의 크로마토그램과 많 은차이가 있었다. 4. 환자의 병력과 이자액 각 분획의 크기를 가변수 처리한 다중 회귀분석 결과 환자의 성별, 시료 채취방법, 혈청 amylase치와 관련된 분획들이 발견되었다. 5. 이자석회화를 동반한 만성 취장염 환자의 이자액 1예에서 약한 chymotrypainogen 분 획 이외의 다른 단백 분뵉은 검출되지 않았다. 이상의 성적으로 보아 hydrophobic interaction colum을 이용하여 HPLC 방법으로 분리 한 사람 이자의 효소 단백 분비 양상은 흰쥐의 크로마토그램과 다르며, 효소 단백의 HPLC 분석 방법을 사람 이자액 효소 단백의 효과적인 분리, 정량 방법으로서 앞으로 이자 질환 진단에 유용하게 쓰일 수 있다고 생각한다. Analysis of human pancreatic juice proteins by high-performance liquid chromatograhy using hydrophobic interaction column Min Goo Lee Department of Medical Sciences, The Graduate School, Yonsei University (Directed by Professor Kyung Hwan Kim, M.D.) The human pancreas posseses an extraordinary capacity to synthesize and secrete a wide range of products, mainly the digestive enzymes. Knowledge of the secretory profiles of pancreatic exocrine enzyme is important for the understanding of physioiogical as well as pathological processes in the pancreas. In the past, studies on human exocrine pancreatic proteins hate relied on the use of electrophoretic procedures and an important progress was done by two dimensional(2-D) gel electrophoresis. Although 2-D gel electrophoresis produces a well resolved separation of pancreatic juice proteins, it has certain drawbacks. The preparation, running, and staining of 2-D gels require significant skill and need about 30 hours to be completed. To overcome these problems the reversed phase high-performance liquid chromatography(HPLG) technique was developed. However, the characterization of individual separated protein was difficult because the acetonitrile used as a solvent denatures the proteins. Recently, HPLC methods using hydrophobic interaction column(HIC) was developed to separate pancreatic juice protein. The solvent used to HIC did not denature the proteins during the separation of rat pancreatic juice proteins. The present study was the first approach to resolve and characterize enzymes in human pancreatic juice by HPLC using HIC, and the clinical usefulness of this technique to diagnosis and therapy of human pancreatic disease was also discussed. Pancreatic juice obtained from 14 human subjects was studied,10 samples were from pancreatic duct catheter after surgery and 4 samples ware collected by endoscopic retrograde cannulation of the main pancreatic duct. The pancreatic juice protein was separated by HPLC using TSK phenyl 5PW HIC and the enzyme activities of each peak were identified. The results are as follows : 1. Decreasing four-stage ammonium sulfate concentration in the gradient of motile phase, it was possible to obtain an ideal chromatogram of human pancreatic juice protein, and total 15 peaks were observed in the chromatogram. 2. From the chromatographic fractions,3 chymotrysinogens,3lipases,3 trpsinogens,1 amylase,1 procarboxypeptidase A, and 1 procarboxypeptidase B peaks were identified. 3. The secretory enzyme profile of human pancreatic juice was quite different from that of rat pancreatic juice. 4. The multiple regression analysis between characteristics of the patient and each peak area of the chromatogram revealed certain peaks related to sex, sampling method, or serum amylase level. 5. The pancreatic juice chromatogram of patient suffered from chronic pancreatitis with pancreatic calcification showed only a small chymotrypsinogen peak. These results suggest that the HPLC chromatogram of human pancreatic enzyme protein is different from rift, and the HPLG analysis using hydrophobic interaction column allows rapid, reproducible separation of the major human pancreatic enzyme proteins, and it mas be useful in the diagnosis of human pancreatic diseases.-
