백서 간장핵내 apurinic DNA endonuclease의 분리 및 특성에 관한 연구

dc.description.abstract[한글] AP DNA endonuclease는 물리적 또는 화학적 인자들에 의해 손상을 받은 DNA를 다시 정상 DNA로 수정하는 과정에서 purine이나 pyrimidine 염기가 제거된 DNA의 phosphodiester 결합을 절단하는데 관여하는 효소이다. AP DNA endonuclease는 박테리아 뿐만 아니라 사 람을 포함한 대부분 동물조직에 널리 분포되어 있으며 각기 다른 종이나 조직에서 분리된 이 효소들의 특성이 서로 다르고 그 종류도 다양하다고 보고되었다 (Verly등, 1981: Shaper등, 1982;Friedberg, 1984). 본 연구에서는 백서 간장핵에 존재하는 AP DNA endonucle ase의 분포 상태를 관찰하고 염색질에 존재하는 AP DNA endonuclease를 순수 분리하여 이들의 물리 화학적 성질의 일부를 밝히고 특히 기질에 대한 특이성을 밝혀 자외선이나 화학 발암물질에 의해 손상을 받은 DNA의 수정기전의 일부를 해명하고져 하였다. AP DNA endonuclease는 백서 간장핵 염색질 뿐만 아니라 핵질에도 분포되어 있으며 핵질에는 간장 조직 g당 18.9±8.6EU의 활성도를 가진 적어도 두가지 형태의 AP DNA endonuclease가 그리고 염색질에는 간장조직 g당 7.1±2.8 EU 활성도를 가진 3가지 형태의 AP DNA endonuclease가 존재하였다. 염색질에 존재하는 3가지 형태의 AP DNA endonuclease (APcⅠ, APcⅡ, APcⅢ) 중 APcⅡ와 APcⅢ를 1M KCl 추출, DEAE Trisacryl, Sephadex G-150 그리고 AP DNA cellulose affinity column chromatography를 시행하여 거의 순수한 형태로 분리하였다. 순수 분리된 APcⅡ, APcⅢ의 활성도는 각각 83.3과 52.0 EU/mg protein이었고 이들의 분자량은 각각 42,000과 13,000 daltons이었으며 등전점은 각각 6.3과 6.2이었다. AP DNA는 APcⅡ, APcⅢ가 모두 작용하는 공통 기질이었으며 UV DNA는 APcⅡ와 APcⅢ가 모두 작용하는 기질이었다. 그러나 3'-Me MAB DNA adduct는 APcⅢ만이 작용하는 유일한 기질이었다. 이상과 같은 결과로 미루어 AP DNA endonuclease는 백서 간장 염색질 또는 핵질에 널리 분포되어 있으며 종류도 다양하여 핵질에는 적어도 2개 염색질에는 3개의 AP DNA endonuclease가 존재하며 이들은 서로 특성이 다르고 기질에 대한 특이성이 서로 다른 효소들이었다. [영문] Apurinic(AP) DNA endonucleases are enzymes which are responsible for the catalytic hydrolysis of phosphodiester bond of apurinic or apyrimidinic DNA generated in the course of the repair of DNA damaged by physical or chemical agents. It has been reported that AP DNA endonucleases were widely distributed not only in bacteria but also in most mammalian tissues including man, and that the nature of the enzymes isolated from various sources of different species was variable (Verly et al. 1981; Shaper et al. 1982; Friedberg. 1984). In the present study an attempt has been made to clarify the repair mechanism of DNA damaged by ultraviolet light (UV) or chemical carcinogen through the test for substrate specificity of AP DNA endonuclease purified from rat liver nuclei. AP DNA endonucleases were localized not only in chromatin but also in nucleoplasm. There were at least two forms of AP DNA endonuclease in the crude nucleoplasm which had an activity of 18.9±8.6 EU/g liver, and the chromatin extract had an activity of 7.1±2.8 EU/g liver. Three forms of AP DNA endonucleases (APcⅠ, APcⅡ and APcⅢ) were extracted with 1M KCI from nuclear chromatin and were purified to near homogeneity by DEAE Trisacryl, Sephadex G-150 and AP DNA cellulose chromatography The specific activities of the purified APcⅠ, APcⅡ and APcⅢ were 62.5, 83.3and 52.0 EU/mg protein, and the molecular weights of these enzymes were 30,000, 42,000 and 13,000 daltons, respectively. Isoelectric points of APcⅠ, APcⅡ and APcⅢ were 7.2, 6.3 and 6.2, respectively, and AP DNA was for APcⅡ and APcⅢ, but 3'-Me MAB DNA adduct was the only substrate for APcⅢ. From these results, it is concluded that various forms of AP DNA endonucleases are present in rat liver nuclei and 3 forms of them are present in nuclear chromatin, which have different physicochemical natures and substrate specificities.-
