골아세포내의 Ca2+ 활성도 변화가 골형성에 미치는 영향

dc.description.abstract[한글] 세포내에 소량의 농도로 존재하는 Ca**2+ 은 세포내의 자극 전달 경로에 중요한 역할을 하는 이차 매개체로 알려져 있다. 한편 골아세포의 골형성 작용이 골아세포내의 Ca**2+ 활성도 증가와 관련이 있다는 것은 부갑상선 호르몬 및 비타민 D에 의해 골아세포내 Ca** 2+ 활성도가 증가하고 골절 후 골유합 치료에 사용되는 전기적 자극이 골아세포내 Ca**2+ 활성도를 증가시킨다는 사실을 미루어 알 수 있다. 이에 본 연구는 골아세포의 세포내 Ca**2+ 활성도 변화가 골절유합에 미치는 영향을 조사하고자, Ca**2+ ATPase를 억제시켜 세포내 Ca**2+ 활성도를 증가시키는 vanadate와 Ca**2+ 통로를 억제시켜 세포내 Ca**2+ 활성도를 감소시키는 verapamil을 Sprague-Dawley계 흰쥐의 경골 골절 부위에 주입시켜 약물의 효과적인 농도를 알아본 후, 이 농도의 약물로 처치한 골아세포를 골절 부위에 주입하여 그 결과를 조직학적으로 비교 분석하였다. Hematoxylin-eosin(H-E) 염색으로 골절 유합 과정을 관찰하였으며, 또한 분열기의 세포를 확인할 수 있는 bromodeoxyuridine-항 bromodeoxyuridine(BrdU) 면역조직화학적 방법으로 분열기 세포의 수를 계측(BrdU 표지 지수)함으로써 골절유합 초기의 골아세포 활성화를 관찰하여 다음과 같은 결과를 얻었다. 1. 골형성을 가장 효과적으로 촉진시킬 수 있는 vanadate의 농도는 10**-6 ∼10**-5 M이었으며, 골형성을 억제하는데 유효한 verapamil의 농도는 10**-6 ∼10**-5 M이었다. 2. H-E 염색상 골절 초기에 있어서 vanadate 처치군은 대조군 보다, 대조군은 verapamil 처치군 보다 더 풍부한 골아세포 및 유골조직을 형성하였고, 더 많은 원시간엽세포의 분열상을 보였으며, 연골세포 및 유연골 조직의 출혈과 직골로의 성숙이 빨리 이루어졌다. 3. Vanadate 처치 골아세포의 BrdU 표지 지수는 대조군보다 높았으며, verapamil 처치군의 BrdU 표지 지수는 대조군보다 낮았다. 이와같은 결과로 미루어 보아 골아세포내 Ca**2+ 활성도 변화는 골유합의 기전에 작용하는데 세포내 Ca**2+ 활성도를 증가시키는 vanadate는 골형성을 촉진시키는 효과가 있으며, 골아세포내 Ca**2+ 활성도를 감소시키는 Verapamil은 골형성을 감소시키는 효과가 있 는 것으로 사료된다. [영문] Bone formation by osteoblast may be closely related to the increase in intracellular Ca**2+ activity of osteoblast. Observations of the facilitated fracture healing by parathyroid hormone, vitamin D and electric stimulation which increase Ca**2+ activities in osteoblast provided the evidence for the role of Ca**2+ as a second messenger for intracellular pathway of signal transduction. In order to study the effects of chanties in Ca**2+ activity of osteoblast on fracture healing, the author has undertaken experiments where Ca**2+ activities of osteoblasts from the Sprague-Dawley rat were raised by vanadate, Ca**2+ ATPase inhibitor, and lowered by verapamil, Ca**2+ channel blocker. Upon confirming the effective dose range of these two drubs for fracture healing by way of direct injection of them into the fracture site of tibia of rat, cultured osteoblasts were treated by a dose in the confirmed effective range. The treated osteoblasts were then injected directly into fracture sites. The whole process of fracture healing was observed by hematoxylin-eosin(H-E) stain while mitotic activity of osteoblasts was specifically observed by bromodeoxyuridine(BrdU) stain enabling the count of labeling index. The results were as follows: 1. The most effective range of concentration which could facilitate bone formation was 10**-6 to 10**-5 M, while the most effective range of concentration capable of verapamil, suppressing one formation was 10**-6 to 10**-5 M. 2. H-E stain showed more abundant osteoblast and osteoid tissues, more active mitotic division of osteoblast, and earlier appearance of chondroblast and chondroid tissue, making the maturation of woven bone faster in the vanadate-treated than in the control group. The opposite was true in the verapamil-treated group compared with the control group. 3. BrdU labeling index showed more active osteoblastic proliferation in the vanadate-treated than in the control group. The opposite was observed in the verapamil-treated group compared with the control group. In conclusion, the fracture healing appears to be facilitated and decelerated by vanadate which apparently increases intracellular Ca**2+ activity of osteoblast and verapamil which decreases it, repectively. Therefore the change of intracellular Ca**2+ activity of osteoblast can be considered to be one of fracture healing mechanisms and expected to be applied for clinical purpose.-
