There have been many reports on the relationship between the reticuloendothelial

system and allergy but opinions differ among the authors.

Owing to many investigators, our knowledge of reticuleothelial system has been

expanded, especially as regards to its close relationship to immune bodies.

Many investigators disclosed that blockage of reticuloendothelial system with

India Ink has inhibiting effect on anaphylactic shock.

Some claimed that blockage of reticuloendothelial system with small amount of

India Ink enhances an allergic reaction but with large amount of India Ink an

allergic reaction is inhibited.

But some insisted that blockage of reticuloendothelial system influences on the

antibody formation of the system thereby inhibiting allergic reaction.

On the other hand, other reticuloendothelial system hormone which is produce by

blockage of reticuloendothelial system.

Thus there is no clear-cut conclusion as to the exact nature or mode of such

effect.

The purpose of the present experiments is to study the relationship between the

function of the reticuloendothelial system and the allergic inflammation of testis.

Materials and Methods

Normal rabbits weighing around 2kg, were used. Normal fresh horse serum 1.0c.c.

per kg, body weight was subcutaneously injected for 4 consecutive days for

sensitization and 0.1c.c. per kg, of body weight was injected into the testicle 2

weeks later.

The rabbits were divided into 8 groups as follows;

Group 1. control.

Group 2. Reticuloendothelial system block with large amount of India Ink before

sensitization.

Group 3. Reticuloendothelial system block with small amount of India Ink before

sensitization.

Group 4. Reticuloendothelial system block with large amount of India Ink before

reinjection.

Group 5. Reticuloendothelial system block with small amount of India Ink before

reinjection.

Group 6. Normal rabbit serum injection control.

Group 7. The rabbit serum injection group and reticuloendothelial system block

with large amount of India Ink.

Group 8. The rabbit serum injection and reticuloendothelial system block with

small amount of India Ink.

Results

1. The histologic change of the testicle after reinjection revealed acute

inflammation of the testicle with maximum changes occuring at 48 hours after

reinjection and the changes subsided gradually. Two weeks after reinjection the

changes cleared up except for necrotic foci with leucocytic Infiltration.

2. The 2nd group revealed similar changes with those of the control groups.

3. The allergic changes of the 3rd group were milder than those of the control

group.

4. The 4th group showed the allergic changes of the testicle to be definitely

milder than those of the control group.

5. The 5th group showed the allergic changes of the testicle to be milder than

those of the control group.

6. The 6th group showed the allergic changes of the testicle to be same as those

of the normal control group.

7. The 7th group showed the allergic change of the testicle to be definitely

milder than those of the control group.

8. The 8th group showed the allergic changes of the testicle to be mildest among

any other groups.

Conclusions

1. India Ink blockage of the reticuloendothelial system of rabbit has the

inhibitory effect on the testicular allergy induced by horse serum.

2. The inhibitory effect on the testicular allergy is not based on the inhibitory

effect of the antibody formation by reticuloendothelial system but, on the

inhibitory effect of a substance in the rabbit serum, which is produced by

reticuloendothelial system blockage with large amount of India Ink. The blockage 2

weeks before reinjection did not appear to produce enough amount of the effective

substance which has inhibitory effect on the testicular allergy.

3. The reticuloendothelial system blockage with small amount of India Ink did

produce enough amount of the effective substance which has inhibitory effect on the

testicular allergy.
