사람 간암세포주에 대한 Desferrioxamine의 증식억제 작용기전

dc.description.abstract[한글] 철분은 종양세포를 포함한 모든 세포의 성장에 필수적인 영양소로 DNA합성의 첫단계를 촉매하는 효소인 ribonucleotide reductase의 활성화에 관여하는 것으로 알려져 있다. 그 러나 철분이 과잉상태가 되면 종양발생의 위험성이 높아지며, 반대로 철분의 결핍상태는 종양세포의 성장을 억제할 수 있다고 알려져 있다. 철분을 킬레이트화하는 대표적인 약제인 desferrioxamine(DFO)이 백혈병 세포 및 신경 모세포종 세포등 몇몇 종양에서 항종양효과를 갖고 있음이 알려졌으며, 최근에는 우리나 라에서 흔한 악성종양의 하나인 간암세포에서도 중식억제효과가 있음이 보고되었다. DFO 의 종양세포 증식억제기전은 DNA합성을 감소시키고 세포주기 중 S기를 차단하기 때문이라 고 알려져 있으나, DNA합성을 억제하지 않는 농도에서도 증식억제효과가 있다는 보고도 있고, 철분 킬레이트화 이외의 작용도 있을 수 있다는 보고도 있으며 세포의 종류 및 상 태에 따라 세포주기에 미치는 영향이 다르다는 주장도 있는 등 아직 논란이 있다. 사람 간암세포에서도 DFO에 의한 증식억제효과가 보고되었으나 DFO가 DNA합성이나 세포주기에 미치는 영향에 관해 아직 알려져 있지 않다. 본 연구에서는 사람 간암세포주인 Hep 3B 및 Hep G2 세포를 대상으로 [(3)**H] thymidi ne섭취검사를 이용하여 DFO가 DNA합성을 억제하는지를 알아보고, 또한 DFO가 세포주기에 미치는 영향을 알아보고자 propidium iodide 및 항 bromodeoxyuridine(BrdU)항체를 사총 한유식세포분석(flow cytometry)을 시행하여 다음과 같은 결과를 얻었다. 1. DFO가 DNA합성에 미치는 영향을 알아보기 위하여 0, 2, 6, 20, 60 및 120㎍/㎖의 DF O를 Hep G2 세포배양액에 첨가하고 2, 24, 48, 72 및 96시간 배양한 후 [(3)**H] thymidi ne섭취검사를 시행한 결과 첨가한 DFO의 농도에 비례하여 [(3)**H] thymidine 섭취양이 감소하였다. 2. DFO가 세포주기에 미치는 영향을 알아보기 위하여 HeP 3B 및 Hep G2 세포에 0, 2, 6 및 20㎍/㎖의 DFO를 첨가하여 2, 24, 48 및 72시간 배양한 후 유식세포분석을 시행하였던 결과 Hep 3B와 이배수성(diploid) 및 사배수성(tetraploid)의 Hep G2 세포 모두에서 첨 가한 DFO의 농도에 비례하여 S기는 증가하였고 G0/G1기는 감소하였다. 3. DFO가 relative movement(RM) 및 S phase duration(Ts)에 미치는 영향을 알아보기 위하여 Hep 3B 세포에서 BrdU 표지법으로 양가유식세포분석을 시행한 결과 DFO를 첨가하 지 않고 배양하였던 경우는 시간경과에 따라 RM이 증가하였으며 Ts는 9.9시간이었으나, 2 0㎍/ml의 DFO를 첨가하여 24시간 배양하였던 경우에는 BrdU 표지세포가 S중기 및 S후기로 이동하지 않고 계속적으로 S초기에 축적되었으며 Ts가 54.1시간으로 5.5배 지연되었다. 4. DFO가 세포주기에 미치는 영향의 가역성을 알아보기 위하여 HeP 3B 세포에 20㎍/ml 의 DFO를 첨가하고 24시간 배양한 다음 신선배지로 교환하여 DFO의 작용을 중지시키고 0, 2, 6, 15 및 24시간째에 양가유식세포분석을 시행하였던 결과 시간이 경과함에 따라 S초 기로 부터, S중기, S후기 및 G2/M기로의 순차적인 증가가 관찰되었다. 이상의 실험결과로 사람 간암세포주에 대한 DFO의 증식억제 작용기전은 DNA합성의 억제 에 의하며, 이는 세포주기 중 S초기와 S중기의 경계부위 또는 S중기를 차단하기 때문인 것으로 생각된다. [영문] Iron is essential for the growth of all living cells. However, iatrogenic iron overload has been associated with neoplasia, and it was postulated that the depletion of iron might evert an antitumor effect. Desferrioxamine(DFO), an iron chelator, has been shown to have antiproliferative activity in a variety of malignant cell lines. Recently, there were several reports on the antitumor activity of DFO against cultured cell lines of human hepatocellular carcinoma, which is one of the most common causes of death due to cancer in Korea. The antiproliferative effect of DFO might be caused by decreased ribonucleotide reductase activity resulting in decreased DNA synthesis, and BFO has been known to be a reversible blocker in the S phase or Gl-S interface. However, there remains some debate over the exact mechanisms by which iron chelators exert their antiproliferative effects. The effect of DFO on DNA synthesis and cell cycle of human hematoma cells has yet to be clarified. Furthermore, if DFO is to be used as a modulator of tumor growth or as an adjunctive mode of treatment with cell cycle-specific chemotherapeutic agents, then an understanding of the mechanisms involved is necessary to maximize such effects. This study was conducted to investigate "whether DFO inhibits the DNA synthesis of cultured hepatoma cells" and "what effect does DFO have on the cell cycle of these cells". Using HeP 3B and Hep G2 cells as hematoma cell lines, DNA synthesis was measured by [(3)**H] thymidine incorporation, and DNA flow cytometry which included bivariate analysis was used to analyze the cell cycle. The results obtained were as follows: 1. In Hep G2 cells, [(3)**H] thymidine uptake decreased in a dose dependent manner after the addition of DFO in the culture media. 2. In both Hep 3B and Hep G2 cells, the proportion of S phase cells increased and that of GO/Gl phase cells decreased in a dose dependent manner after the addition of DFO in the culture media. 3. In Hep 3B cells, the S phase duration was 9.9 hours when cultured without DFO, but it was Prolonged markedly(54.1 hours) after the addition of DFO. In DFO treated Hep 3B cells, the increment in the proportion of the early S phase which continued without shift to the mid-S and late-S suggested that the cell cycle was blocked and delayed in the S Phase by DFO. 4. In Hep 3B cells, after removal of DFO from the media following 24 hours of incubation with 20㎍/ml of DFO, a sequential increase from early-S through mid-S and late-S to G2/M Phase was observed. In conclusion, the antiproliferative effect of DFO on cultured human hepatoma cell lines was caused by inhibiton of DNA synthesis by DFO, which was due to a block in the early-mid S interface or mid S phase of the cell cycle.-
